精神分裂症是重性精神疾病的一种,世界人口患病率约1%,其不仅严重损害病人本人的社会功能,也给病人家庭和社会带来一定程度的不利影响[1]。研究精神分裂症的发病机制,寻求有效的治疗手段,是国内外学者一直以来的奋斗目标。目前研究[2]已表明,遗传是精神分裂症最主要的因素。miRNA是一种内源性的非编码RNA,通过作用于目标mRNA,在转录后水平调控基因的表达[3]。研究[4]发现,miRNA在精神分裂症病人体内差异表达,部分miRNA可能在精神分裂症的发病机制中扮演重要角色。但精神分裂症病人体内差异表达的miRNA在精神分裂症发病过程中的具体作用机制仍然不明,有待进一步阐述。本研究在既往研究的基础上,进一步运用生物信息学软件预测精神分裂症病人外周血差异表达miRNA-181b的靶基因、转录因子和相关信号通路,为精神分裂症病因学的探究提供参考。现作报道。

miRNA-181b的成熟体有hsa-miRNA-181b-3p和has-miRNA-181b-5p两种,经过计算,hsa-miRNA-181b-3p共有227个靶基因,has-miRNA-181b-5p共1 153个靶基因,miRNA-181b靶基因共1 344个,其中hsa-miRNA-181b-3p和has-miRNA-181b-5p共有的靶基因有36个(ADAMTS1、ADAMTS2、ADM、ANKFY1、ARF3、ARHGAP11A、ATP8B2、BCL2L2、CD164、CTSD、DDX21、DDX39B、DYNLL2、FEM1B、H3F3B、HIPK1、HMGA2、HNRNPF、HNRNPU、MAP7、MAPK1、MDN1、NUCKS1、PIP4K2A、PPA1、RHOB、SCD、TBL1XR1、UBE3C、VKORC1L1、WNK1、YY1、ZBTB10、ZC3H12B、ZNF432、ZNF711),显著表达的靶基因是ARHGAP11A、PPA1、YY1和ZC3H12B。

对hsa-miRNA-181b-3p和has-miRNA-181b-5p的靶基因进行GO生物学过程富集分析,以P < 0.05为具有统计学意义。结果显示,hsa-miRNA-181b-3p和has-miRNA-181b-5pmiRNA-181b-3p的靶基因共有的富集条目有27个,其聚类图见图 1,包括分子功能、RNA结合、细胞组成、细胞核核质、细胞质、细胞蛋白质的修饰过程、生物过程、生物合成过程、基因表达、酶结合、细胞氮氧化物代谢过程、离子结合、细胞器、蛋白质复合物等过程的调节,其中显著富集的条目见表 1。

对miRNA-181b-3p和miRNA-181b-5p的靶基因进行KEGG信号通路富集分析,结果显示,共有12个KEGG信号通路条目显著性富集。将P值以10为底进行负对数处理(-LgP)后做图,得到KEGG信号通路的-LgP柱状图(见图 2)。显著性富集的条目包括:内质网中蛋白质的加工、神经胶质瘤、p53信号通路、HIF-1信号通路、赖氨酸的降解、亨廷顿病和黏合连接等通路。

采用miRGen V.3数据库分析miRNA-181b的转录因子,结果表明,与miRNA-181b显著关联的转录因子有7个,分别是JUN、IRF1、EGR1、ZNF263、ESRRA、MEF2C和FOSL2(见表 2)。

目的对精神分裂症病人外周血中差异表达的miRNA-181b进行靶基因、转录因子和信号通路的分析。 方法采用DIANA-TarBase数据库分析miRNA-181b靶基因,应用DIANA-miPath V.3软件对miRNA-181b的靶基因进行GO功能富集分析和KEGG信号通路富集分析,运用DIANA-miRGen v3.0数据库分析miRNA-181b的转录因子。 结果miRNA-181b靶基因共1 344个,其中显著表达的共有靶基因是ARHGAP11A、PPA1、YY1和ZC3H12B;miRNA-181b的靶基因广泛参与生物学过程的调节,包括分子功能、RNA结合、细胞组成等过程;KEGG生物信息学分析发现miRNA-181b的靶基因富集了12条信号通路,包括hippo信号通路、p53信号通路、HIF-1信号通路、赖氨酸的降解等;与miRNA-181b显著关联的转录因子有7个,其中JUN、EGR1、MEF2C与精神分裂症有显著关联。 结论对精神分裂症病人外周血中差异表达的miRNA-181b进行生物信息学分析,发现了多个靶基因、信号通路和转录因子与精神分裂症的发病有密切关联。 -

Abstract:ObjectiveTo analyze the target gene, transcription factor and signal pathway of the differential expression of miRNA-181b in the peripheral blood of schizophrenic patients. MethodsThe miRNA-181b target gene was analyzed using the DIANA-TarBase database, the GO function and KEGG pathway enrichment of miRNA-181b gene were analyzed using DIANA-miPath V.3 software, and the transcription factors of miRNA-181b were analyzed using the DIANA-miRGen v3.0 database. ResultsThere were 1 344 target genes of miRNA-181b, and the target genes ARHGAP11A, PPA1, YY1 and ZC3H12B significantly expressed.The target genes of miRNA-181b were widely involved in the regulation of biological processes(including molecular function, RNA binding and cell composition).The results of KEGG bioinformatics analysis showed that the target gene of miRNA-181b enriched 12 signal pathways, which included the hippo signal pathway, p53 signal pathway, HIF-1 signal pathway and lysine degradation.Seven transcription factors were significantly associated with miRNA-181b, and JUN, EGR1 and MEF2C were significantly associated with schizophrenia. ConclusionsThe multiple target genes, signal pathways and transcription factors are closely related to the onset of schizophrenia using the bioinformatics analysis of the differential expression of miRNA-181b in the peripheral blood of schizophrenic patients. -
