果糖高脂饮食诱导建立代谢综合征大鼠模型
The establishment of metabolic syndrome rat model induced by fructose and high-fat diet

摘要: 目的:建立糖、脂代谢异常的代谢综合征大鼠模型,为进一步研究代谢综合征的发病机制及制订膳食治疗措施提供基础。方法:SD雄性大鼠30只,体质量(210±10)g,随机分为观察组和对照组。观察组采用15%果糖水配合高脂饮食,对照组采用普通饲料,喂养10周,期间检测体质量、体长、腰围、空腹血糖及血清三酰甘油(TG)、高密度脂蛋白(HDL-C)及胰岛素水平。结果:试验8、10周,观察组大鼠体质量、Lee's指数和空腹血糖、TG均明显高于对照组(P<0.01),HDL-C均明显低于对照组(P<0.01);试验4、8、10周,观察组大鼠腰围均大于对照组(P<0.05~P<0.01),空腹胰岛素和稳态模型胰岛素抵抗指数亦均高于对照组(P<0.05~P<0.01)。观察组大鼠中12只达到中心性肥胖标准,11只达到高TG标准,11只达到低HDL-C标准,12只大鼠出现高血糖。综合以上指标,4只大鼠分别有两项或三项指标未达到标准,成模率为73.3%(11/15)。结论:15%果糖水配合高脂饮食可在较短时间内诱导构建代谢综合征大鼠模型。Abstract: Objective:To establish the rat model of metabolic syndrome(MS) with abnormal glucose and lipid metabolism,and provide a basis for studying the pathogenesis of metabolic syndrome,and developing dietary treatment measures.Methods:Thirty SD male rats with(210±10)g were randomly divided into the observation group and control group.The observation group were fed with 15% fructose water mixed with high-fat diet,and the control group were fed with regular diet for 10 weeks,respectively.The levels of body weight,body length,waist circumference,fasting blood glucose,serum triglycerides,high-density lipoprotein cholesterol and insulin in two groups were detected.Results:The levels of body weight,Lee's index,fasting blood glucose,and serum triglycerides,high-density lipoprotein cholesterol in observation group were higher than those in control group(P<0.01),and the level of HDL-C in observation group was lower than that in control group after 10 weeks of test(P<0.01).After 4,8 and 10 weeks of test,the waist circumference in observation group was more than that in control group(P<0.01 to P< 0.05),the fasting insulin and steady-state insulin resistance index in observation group were higher than those in control group(P<0.01 to P<0.05).Twelve rats arrived at central obesity criteria,11 rats arrived at the high TG criteria,11 rats arrived at low HDL-C critia,and 12 rats with high blood glucose were found.Combined with the above indicators,two or three indexes of 4 SD rats failed to meet the criteria,and the rat model of metabolic syndrome rate was 73.3%.Conclusions:The 15% fructose water combined with high-fat diet can induce the rat model of metabolic syndrome in a short time.
