血清UCH-L1蛋白在新生大鼠缺氧缺血性脑病评估中的价值
Application value of the serum ubiquitin carboxyl-terminal esterase L1 protein level in the diagnosis of hypoxic-ischemic encephalopathy in neonatal rats

摘要: 目的:探讨血清泛素羧基末端水解酶-1(UCH-L1)蛋白在新生大鼠缺氧缺血性脑病中的价值。方法:将168只新生大鼠随机分为对照组、手术组及缺氧缺血性脑损伤(HIBD)组,每组56只;各组组内又随机分为7个亚组:6 h组、12 h组、24 h组、48 h组、72 h组、96 h组及7 d组,每亚组8只;HIBD组采用改良Rice法制备HIBD模型。各组大鼠于不同时间点处死取血,采用ELISA法检测血清中UCH-L1表达水平的变化。结果:不同时间点3组大鼠间UCH-L1蛋白表达水平差异均有统计学意义(P<0.01),HIBD组UCH-L1蛋白表达水平显著高于对照组和手术组(P<0.01),而对照组与手术组间UCH-L1蛋白表达水平差异无统计学意义(P>0.05)。HIBD组大鼠各时间点UCH-L1蛋白表达水平差异有统计学意义(P<0.01);HIBD组血清UCH-L1蛋白在6 h时检测水平明显升高,48 h到达峰值,后逐渐下降,且脑组织损伤越严重,血清UCH-L1蛋白表达水平越高。结论:UCH-L1蛋白血清表达水平与脑组织损伤的严重程度表现一致,可作为检测指标应用于评估脑组织损伤的严重程度。

Abstract: Objective:To explore the application value of serum level of ubiquitin carboxyl-terminal esterase L1(UCH-L1) in the diagnosis of hypoxic-ischemic encephalopathy in newborn rats.Methods:One hundred and sixty eighty newborn SD rats were randomly divided into the control group,sham-operation group and hypoxic-ischemic brain damage(HIBD) model group(56 rats each group).Each group was randomly subdivided into the 6 h,12 h,24 h,48 h,72 h,96 h and 7 d groups(8 rats each subgroup).The HIBD model was established using the improved Rice method.All rats were sacrificed to obtain blood at different time points.The serum levels of UCH-L1 were tested by ELISA.Results:At different time-points,the differences of the levels of UCH-L1 between three groups were statistically significant(P<0.01),the levels of UCH-L1 in HIBD model group were significantly higher than that in control group and sham-operation group(P<0.01),and the difference between the control group and sham-operation group was not statistically significant(P>0.05).The differences of the levels of UCH-L1 in HIBD model group between different time-points were statistically significant(P<0.01).In HIBD model group,the serum levels of UCH-L1 began to significantly increase at 6 h,reached the peak value at 48 h,then decreased gradually.The more severe the brain damaged,the higher the level of serum UCH-L1 was.Conclusions:The levels of UCH-L1 is positively correlated with the severity of the brain damage.The expression of UCH-L1 can be used an indicator in evaluating the brain damage. -
