Abstract:
Objective To endoplasmic reticulum stress cell model of human umbilical vein endothelial cells(HUVECs), to estoblish observe the effects of coptisine on the growth activity of HUVECs cells, and expression levels of CHOP and GRP78 gene and protein. Methods HUVECs were induced by tunicamycin(TM) at a concentration of 10 μg/mL for 18 h, and then treated with coptisine at different concentrations for 24 h.The CCK8 was used to detect the effects of different concentrations of coptisine on the activity of HUVECs and activity of HUVECs induced by TM.The mRNA expression levels of CHOP and GRP78 in each group were detected using RT-PCR.The protein expression levels of CHOP and GRP78 in each group were detected using Western blotting and cellular immunofluorescence.>Results With the concentration of coptisine increasing, the coptisine in the range of 1-20 μmol/L had no effect on the activity of HUVECs.After 24 hours, the concentration of coptisine in the range of 20 μmol/L played a protective effect on the activity of HUVECs induced by TM(P < 0.01).The mRNA expression levels of CHOP and GRP78 in HUVECS increased after 18 hours of TM induction(P < 0.01), after 24 hours of continuous induction with coptisine, it was found that coptisine could inhibit the mRNA expressions of CHOP and GRP78 in TM-induced HUVECs(P < 0.01).The protein expression levels of CHOP and GRP78 in HUVECs increased after 18 hours of TM induction(P < 0.01), and after 24 hours of continuous induction with coptisine, it was found that coptisine could inhibit the protein expression levels of CHOP and GRP78 in TM-induced HUVECs(P < 0.01).The results of cellular immunofluorescence staining were similar to those of Western blotting. Conclusions Coptisine can alleviate the excessive ER stress induced by TM in HUVECs, and improve the activity of damaged cells.Coptisine can improve the excessive ER stress response of HUVECs by down-regulating the expressions of CHOP and GRP78.

Abstract:
ObjectiveTo explore the effects of different fluctuations of membrane potential on the intrinsic characteristics and regulation of neural coding of GABA-ergic neurons in mouse cerebral cortex.MethodsThe brain slices were prepared from FVB-Tg(Gad GFP) 4570Swn/J mice(postnatal day 15-20), and incubated in artificial cerebrospinal fluid with sufficient oxygenation for 2 h.The effects of fast post-polarization(fAHP), slow post-hyperpolarization(sAHP), different repolarization of action potential and fast post-polarization(fADP) on the intrinsic characteristics of neurons and regulation of neural coding were further studied by whole-cell recording.ResultsCompared with sAHP, the absolute refractory period(ARP2) and threshold(vts-Vr3) of the action potential under the influence of fAHP were shorter and higher, respectively(P < 0.01).Compared with fAHP, the ARP2 was shorter when the action potential was not fully repolarized, and the ARP2 was the shortest when the threshold intensity was 0.25 times(P < 0.05).Compared with fAHP, the lower the fADP level was, the shorter the ARP2 was, and the ARP2 was the shortest at the resting potential level(P < 0.05).ConclusionsAfter the integrating synaptic input, the plasticity of intrinsic characteristics of cerebral cortex GABA-ergic neurons changes when the membrane potential fluctuates.Different input modes affect the intrinsic characteristics of action potential of cerebral cortex GABA-ergic neurons, thereby regulating the programming ability of GABA-ergic neurons to input signals.

Abstract:
Objective To investigate the clinical value of the changes of serum C-peptide and glycosylated hemoglobin in patients with type 2 diabetic nephropathy.
Methods A total of 145 hospitalized patients with T2DM were divided into three groups: normal urine albumin group(N-UAlb ,n=81), minor urine albumin group (M- UAlb ,n=48) and clinical urine albumin group(C- UAlb, n=16) according to the urine albumin-creatinine ratio(ACR). The levels of serum C-P were detected and compared between three groups as well as demographic characteristics and clinical parameters. The multiple logistic regression model was used to investigate the association between serum C-peptide, glycosylated hemoglobin and diabetic nephropathy in T2DM patients. Results Compared with the N-UAlb group, the univariate analysis showed higher level of HbA1C, blood urea nitrogen, Scr, age and diabetic duration and lower level of delta C-peptide and eGFR in C- UAlb group(P<0.05).The correlation test showed glycosylated hemoglobin positively correlated with fasting blood glucose(FBG), blood urea nitrogen(BUN) and the occurrence of chronic kidney disease(CKD)(P<0.05),whereas negative correlation were found between delta C-P and diabetic duration, serum creatinine, HbA1C and the occurrence of CKD(P<0.01).Multiple logistic regression indicated the decreasing level of serum C-peptide and HbA1C might be the independent risk factors of diabetic nephropathy in type 2 diabetes patients. The multiple-factor regression also showed that the level of serum C-peptide decreasing and HbA1C increasing were significantly with the increased risk of DN in T2DM patients(P<0.05).
Conclusion The level of serum C-peptide decreased and HbA1C increased significantly with the progress of diabetic nephropathy, which was of certain significance in monitoring the above indicators for diagnosis and prediction of type 2 diabetic nephropathy.

Abstract:
ObjectiveTo compare the changes of MRI morphological score and T2 value of cartilage injury in knee osteoarthritis(OA).MethodsSixty-five knee OA patients and 10 healthy subjects were selected to undergo MRI scanning of sagittal 3D-WATSc and T2 mapping. The patients were divided into mild(OA1), moderate(OA2) and severe(OA3) groups. The correlation between T2 value of cartilage and the whole-organ magnetic resonance imaging score (WORMS) in each sub-region of OA groups was evaluated, and the difference of T2 value in each sub-region among three groups was compared.ResultsExcept for the lateral region of femoral, T2 values were positively correlated with WORMS scores in OA groups(r=0.657-r=0.854, P < 0.05), and the differences of T2 values in other sub-regions were statistically significant (P < 0.01).The sensitivity of T2 value in patella, medial region of femoral and medial region of tibia to detect the pre-pathological changes of cartilage morphology of knee OA was 78.3%, 68.2% and 54.8%, respectively, and the specificity was 100%, 80% and 70%, respectively.ConclusionsBoth the MRI semi-quantitative evaluation index WORMS, and the MRI quantitative evaluation index T2 value can effectively evaluate the change of knee OA cartilage.T2 value can differentiate the severity of cartilage injury of knee OA, and discover the cartilage lesions before morphological changes.
