Abstract:
ObjectiveTo establish the cell line with low-expression miR-374a using CRISPR/CAS9 gene targeting technology.MethodsThe mature gRNA targeting miR-374a was designed to synthesize the DNA, which was inserted into the CRISPR/CAS9 vector to construct the pSpCas9(BB)-2A-Puro.A549 cells were transfected with the constructed vector, and screened using puromycin. The gene sequences of target site were analyzed using PCR and sequencing method. The qPCR was used to detect the relative expression levels of miR-374a and its target gene TGFA in transfected cells. The CCK8 was used to detect the proliferation activity of cells.ResultsThe gene targeting miR-374a vector was successfully constructed. The transfected cells were screened under pressure, and the sequencing results showed that the inserting/deletion mutation was found. The results qPCR showed that the expression level of miR-374a significantly decreased, and the expression level of TGFA significantly increased in transfecting gene editing vector compared with control cells(P < 0.01 and P < 0.05).The cell proliferation assay showed that the proliferation activity significantly changed in low-expressed miR-374a cells(P < 0.05 to P < 0.01).ConclusionsThe low-expressed miR-374a cell line was successfully established, which provides the basis in studying the molecular function.

Abstract:
ObjectiveTo investigate the expression level and effects of astrocyte elevated gene-1(AEG-1) in gastric cancer stem cells.MethodsThe expression levels of AEG-1 mRNA and protein in gastric stem cell lines(HGC-27 and MKN-45) were detected using RT-PCR and Western blotting, respectively. The AEG-1 was processed using transient transfection, and the cells proliferation and its mechanism were investigated using the MTS assay and cell-cycle analysis. The apoptosis was examined using the activities of Caspase-3/7 and annexin V, the effect of silencing AEG-1 on the stemness of gastric stem cells was evaluated using the tumorsphere formation assay, and the tumor formation experiment was conducted to evaluate the effects of AEG-1 on the tumorigenesis of gastric stem cells in nude mice.ResultsThe expression level of AEG-1 in gastric stem cells was higher than that in non-stem cells(P < 0.01).Cancer stem cells transfected with AEG1-siRNA resulted in the cancer stem cell cycle arrest in G1 phase, and could significantly inhibit the cell proliferation(P < 0.01).The results of Annexin V and Caspase-3/7 activities detection showed that the knockdown of AEG-1 could induce the apoptosis of gastric stem cells. The low level of AEG-1 could significantly inhibit the ability of gastric cancer stem cells to form into balls. The loss of AEG-1 inhibited the tumorigenesis of tumor stem cells in vivo.ConclusionsThe silencing expression of AEG-1 can suppress the cell proliferation, induce the cell cycle arrest in G1 phase and apoptosis of gastric cancer stem cells, inhibit the tumorigenesis, which plays a positive role in tumorigenesis of gastric cancer stem cells.

Abstract:
ObjectiveTo investigate the clinical value of thrombelastography(TEG) in predicting the lacunar stroke(LS) developing into progressive lacunar stroke(PLS).MethodsOne hundred and forty-two LS patients were divided into the PLS group(46 cases) and Non-PLS group(96 cases). The maximum amplitude, clotting angle, clotting time and reaction time of TEG parameters in two groups were detected using TEG5000 analyzer.ResultsThe maximum amplitude and clotting angle in PLS group were significantly larger than those in Non-PLS group(P < 0.01), and the clotting time and reaction time in PLS group were significantly shorter than those in Non-PLS group(P < 0.01).The results of Pearson linear correlation analysis showed the maximum amplitude and clotting angle were positively correlated with NHISS score of PLS patients(P < 0.05), and the clotting time and reaction time were negatively correlated with NHISS score of PLS patients(P < 0.01).The logistic regression analysis showed that the maximum amplitude and clotting angle could be used as the independent risk factors of the LS developing into PLS, and the clotting time and reaction time could be used as independent protective factors of the LS developing into PLS(P < 0.05 to P < 0.01).ConclusionsThe value of TEG in the prediction of the LS developing into PLS is obvious. The larger the maximum amplitude and clotting angle are, and the shorter the clotting time and reaction time are, the higher the risk of the LS developing into PLS is.

Abstract:
Objective: To explore the effect of evidence-based nursing on the incidence of pulmonary infection in elderly patients who have been in bed for a long time. Methods: 96 patients over 80 years old who were in bed for a long time from February 2016 to August 2017 were selected as the study subjects. They were randomly divided into observation group and control group, 48 in each group. The patients in the control group were given routine nursing measures. The observation group was given evidence-based nursing on routine nursing. According to the evidence-based analysis of the past patients with pulmonary infection, we found that the risk factors of pulmonary infection in the elderly patients who had been in bed for a long time were smoking history, old age, disturbance of consciousness, ability of spontaneous cough, invasive operation and hospitalization. Meanwhile, according to the above factors, the corresponding interventions were carried out to observe the incidence of pulmonary infection in the two groups after 3 weeks. Results: There were 5 cases of pulmonary infection in the observation group (10.41%) and 13 cases in the control group (27.08%). There was significant difference between the two groups (P < 0.05). CONCLUSION: Evidence-based nursing measures can effectively reduce the incidence of pulmonary infection and improve the therapeutic effect of the patients, aiming at the causes of pulmonary infection in the elderly.
