Abstract:
ObjectiveTo explore the differential expression profile of circular RNA (circRNA) in brain tissue of Alzheimer's disease (AD) model mouse, to construct circRNA-miRNA-mRNA regulatory network, and to clarify the regulatory mechanism of circRNA in the occurrence of AD.MethodsThe differential expression of circRNA in the brain of APP/PS1 transgenic AD model mice was analyzed by gene microarray.Gene ontology (GO) and kyoto encyclopedia of genes and genomes (KEGG) pathway analysis were performed on the differential circRNAs.Real-time quantitative polymerase chain reaction was used to verify the randomly selected 5 differentially expressed circRNAs, and circRNA-miRNA-mRNA was constructed to perform AD target gene function prediction analysis.ResultsA total of 52 differentially expressed circRNAs were screened, of which 28 were up-regulated and 24 were down-regulated.The enrichment analysis results of GO and KEGG showed that the differentially expressed parental genes of circRNAs were mainly involved in nervous system development, protein binding, RNA transport, signaling pathways regulating stem cell pluripotency, and Hippo signaling pathways.Biological information was used to analyze the successfully constructed competing endogenous RNA (ceRNA) network of circRNA-miRNA-mRNA, indicating that the enriched functions of these target genes might play a role through small molecule binding, plasma membrane, cAMP signaling pathway, and Rap1 signaling pathway.ConclusionsThere are various differentially expressed circRNAs in the brain of AD model mice, and these differentially expressed genes may be involved in the molecular regulation of AD occurrence through the circRNA-miRNA-mRNA regulatory network.

Abstract:
ObjectiveTo explore the mechanism of miR-199a-5p in the occurrence of liver cirrhosis.MethodsThe miR-199a-5p mimics and miR-199a-5p inhibitor sequences were designed and synthesized to study the overexpression and inhibition of miR-199a-5p.The effects of overexpression of miR-199a-5p on the proliferation activity of Lx-2 cells were detected by CCK8, 5EdU, and Western blotting methods.Using Lx-2 cell cDNA as template, the wild type primer sequence of TGF-β2 3'UTR and mutant primer sequence of miR-199a-5p potential binding site were amplified by PCR to obtain wild type TGF-β2 3'UTR sequence and mutant TGF-β2 3'UTR sequence, respectively. The regulation process of TGF-β expression was revealed by double fluorescence reporter gene experiment.ResultsIn vitro experiments, the results showed that the vitality of Lx-2 cells transfected with miR-199a-5p mimics significantly increased compared with the control group(P < 0.05 and P < 0.01), and the α-SMA protein levels was significantly higher than that of control group (P < 0.05).After 8 h, 12 h and 24 h of transfection with miR-199a-5p inhibitor, the cell vitality significantly decreased (P < 0.05 and P < 0.01), and the level of α-SMA significantly decreased compared with the control group (P < 0.05).The results of dual luciferase report showed that after transfecting the miR-199a-5p mimics, the expression of luciferase plasmid containing wild-type TGF-β2 3'UTR sequence (WT) significantly decreased compared with the control group (P < 0.01).The expression of luciferase plasmid containing miR-199a-5p binding site mutation sequence (MUT) had no significant changes (P>0.05).ConclusionsUpregulation of miR-199a-5p can promote the proliferation of Lx-2 cell line, and the overexpression of miR-199a-5p can promote the expression of matrix protein α-SMA, and activate the progression of Lx-2 toward fibrosis. The miR-199a-5p plays a role in promoting liver fibrosis and cirrhosis in the process of liver cirrhosis.TGF-β2 is one of the main regulatory factors of liver fibrosis, and miR-199a-5p can promote liver fibrosis and cirrhosis by regulating the expression of downstream TGF-β2.

Abstract:
ObjectiveTo investigate the role of miR-34c downregulated by Epstein-Barr virus (EBV) in promoting the proliferation, migration, and invasion of nasopharyngeal carcinoma cells and its mechanism.MethodsThe expression level of miR-34c in EBV-negative nasopharyngeal carcinoma SUNE1, CNE2, HK1 cells, and EBV-positive C666-1 cells was detected by qPCR.MiR-34c mimics were constructed to analyze the influence of miR-34c on the biological function of nasopharyngeal carcinoma cells.CCK-8, scratch healing assay, and Transwell cell invasion assay were applied to detect the proliferation, migration, and invasion ability of nasopharyngeal carcinoma cells.Western blotting was employed to determine the expression levels of migration and invasion-related proteins, and Erk1/2 signaling pathway proteins in cells.ResultsMiR-34c was downregulated in EBV-positive C666-1 cells (P < 0.05).CCK-8 results showed that the viability of C666-1 cells in the miR-34c group was significantly lower than that in the EBV group at 16, 24, and 48 hours (P < 0.01).The scratch assay and Transwell assay results indicated that compared with the miR-34c group, the migration and invasion ability of C666-1 cells in the EBV group were significantly enhanced (P < 0.01).Western blotting results showed that compared with the miR-34c group, the expression of migration and invasion-related proteins Vimentin, Snail, MMP-2, MMP-3, and Erk1 and Erk2 proteins of C666-1 cells in the EBV group were significantly increased (P < 0.01).ConclusionsMiR-34c is downregulated in EBV-positive nasopharyngeal carcinoma cells, and miR-34c downregulated by EBV can enhance the proliferation, invasion, and migration capacities of nasopharyngeal carcinoma cells and activate the Erk1/2 signaling pathway.

Abstract:
ObjectiveTo analyze the relationship between preoperative serum cancer antigen 125 (CA125) levels and extrauterine metastasis in patients with uterine serous carcinoma.MethodsThe clinical data of patients diagnosed with serous uterine carcinoma from January 2015 to December 2021 were retrospectively analyzed to study the correlation between preoperative serum CA125 level and extrauterine metastasis.ResultsPreoperative serum CA125 levels were related to fallopian tube, ovarian and omentum metastasis (P < 0.05).Patients with CA125 ≥ 41.36 U/mL were more likely to have extrauterine metastasis (AUC=0.797, sensitivity 70.0%, specificity 88.9%, P < 0.01).Multivariate analysis showed that preoperative serum CA125 ≥ 41.36 U/mL was a risk factor for predicting extrauterine metastasis (P < 0.01).Meanwhile, stromal invasion was also significantly associated with extrauterine metastasis (P < 0.05).The 3-year progression-free survival rate was higher in the CA125-positive group than in the negative group, but the difference was not statistically significant (χ2=0.12, P>0.05);the 3-year overall survival rate was higher in the CA125-positive group than in the negative group (χ2=5.60, P < 0.05).ConclusionsPreoperative serum CA125 ≥ 41.36 U/mL is a reliable index to predict extrauterine metastasis of uterine serous carcinoma.

Abstract:
ObjectiveTo investigate the clinical characteristics of patients with small cell lung cancer (SCLC) complicated with hyponatremia and its relationship with chemotherapy efficacy, so as to provide theoretical basis for improving the prognosis of patients with SCLC complicated with hyponatremia.MethodsOne hundred and six SCLC patients were selected, including 38 patients with hyponatremia and 68 patients with normal serum sodium level. All patients were treated with etoposide combined with platinum regimen for 2 consecutive cycles. The general clinical data of hyponatremia group and normal blood sodium group were analyzed, the short-term efficacy was evaluated according to the RECIST 1.1, and the adverse reactions were analyzed according to NCICTCAE 3.0.Th e hyponatremia group was subdivided into blood sodium corrected group and blood sodium uncorrected group according to whether the blood sodium was corrected or not after chemotherapy, and the relationship with clinical efficacy was analyzed. The patients were divided into disease control group and disease progression group according to the chemotherapy efficacy. The levels of gastrin-releasing peptide precursor (ProGRP) and neuron-specific enolase (NSE) before and after chemotherapy were compared between the two groups, and the correlation between the serum sodium level and ProGRP, NSE levels was analyzed.ResultsThe incidence of SCLC complicated with hyponatremia was 15.83% (38/240).There were significant differences in clinical stage, bone metastasis, pleural effusion, NSE level and ProGRP level between SCLC with hyponatremia group and normal sodium group (P < 0.05).The objective remission rate (ORR) and disease control rate (DCR) in the hyponatremia group were lower than those in the normal sodium group (P < 0.05).Compared with the normal blood sodium group, the patients in the hyponatremia group were more likely to have gastrointestinal reactions, and the difference of which was statistically significant (P < 0 05).The ORR and DCR of patients in the sodium corrected group were higher than those in blood sodium uncorrected group (P < 0.05).Before chemotherapy, there was no significant difference in NSE level between the disease progression group and the disease control group (P>0.05), and the level of ProGRP in the disease progression group was higher than that in the disease control group (P < 0.05).The difference of ProGRP and NSE levels in the disease progression group between before and after chemotherapy was not statistically significant (P>0.05), and the levels of ProGRP and NSE in the disease control group after chemotherapy were lower than those before chemotherapy (P < 0.05).After chemotherapy, there was no significant difference in NSE level between the two groups (P>0.05), and the level of ProGRP in the disease progression group was significantly higher than that in the disease control group (P < 0.01).The serum sodium level in SCLC patients was negatively correlated with NSE and ProGRP levels (r=-0.588, -0.642, P < 0.05).ConclusionsPatients with SCLC complicated with hyponatremia have a later clinical stage, are more likely to have bone metastasis and pleural effusion, have poor response to chemotherapy and similar adverse reactions, but the patients with hyponatremia are more likely to have gastrointestinal adverse reactions. The prognosis of patients with corrected blood sodium after treatment is better than that of patients with uncorrected hyponatremia.Serum sodium level is negatively correlated with ProGRP and NSE levels. Therefore, SCLC with hyponatremia is an indicator of poor prognosis, and correction of hyponatremia may improve the prognosis of patients.

Abstract:
ObjectiveTo evaluate left ventricular function in patients with essential hypertension by two-dimensional speckle tracking imaging (2D-STI).MethodsFifty essential hypertension patients with left ventricular ejection fraction (LVEF) >52% were selected as the hypertension group, and according to left ventricular mass index (LVMI), they were divided into left ventricular hypertrophy group (LVH group) and non-left ventricular hypertrophy group (NLVH group).Twenty-two healthy subjects were selected as the control group.Conventional ultrasound data were obtained by conventional two-dimensional echocardiography.Global peak longitudinal strain (GLS), global peak circumferential strain (GCS), global peak radial strain (GRS), left ventricular total rotation Angle (LVtor), longitudinal strain peak time dispersion (LPSD) and circumferential strain peak time dispersion (CPSD) were measured by 2D-STI.The above parameters were compared among groups and their correlation with LVMI was analyzed.ResultsThere were significant differences in GLS, GCS, LVtor and LPSD among groups (P < 0.05).CPSD: there were significant differences between control group and LVH group, NLVH group and LVH group (P < 0.05).GLS was negatively correlated with LVMI (P < 0.01), GCS, LVtor and LPSD were positively correlated with LVMI (P < 0.01), and CPSD was weakly positively correlated with LVMI (P < 0.01).ConclusionsLeft ventricular systolic function is impaired in essential hypertension patients with preserved ejection fraction, and the damage of left ventricular systolic function is aggravated with the thickening of left ventricular systolic function.GLS, GCS, LVtor and LPSD can be used as reliable indicators to evaluate the early changes of left ventricular systolic function in essential hypertension patients, and provide help for clinical diagnosis and efficacy evaluation.

Abstract:
ObjectiveTo investigate the diagnostic value of conventional ultrasound and contrast-enhanced ultrasound for TI-RADS 4 shrunken thyroid nodules.MethodsA tatal of 78 patients(80 nodules diagnosed as TI-RADS 4) undergoing conventional ultrasound and contrast enhanced ultrasound were selected and divided into the shrinkage nodule group(n=38, 38 nodules) and the papillary carcinoma group(n=40, 42 nodules) according to pathological results. Conventional ultrasound and contrast-enhanced ultrasound examinations were performed on all patients. The radiological characteristics of conventional ultrasound images and the enhancement modes of contrast-enhanced ultrasound were compared respectively.ResultsIn terms of conventional ultrasound image features, there was no statistical significance in boundary, echo, aspect ratio, and calcification type(P>0.05), but statistical significance was found in calcification location and "halo sign"(P < 0.01) between the two groups. The incidence of marginal calcification, halo sign, no enhancement or low enhancement at stellate point in the shrinkage nodule group was higher than that in the papillary carcinoma group(P < 0.01). Shrinkage nodules mainly showed no enhancement or punctate low enhancement, while papillary carcinoma mainly showed centripetal low enhancement.ConclusionsThe position of calcification of the nodule, the presence or absence of "halo sign" and the characteristic manifestations of contrast-enhanced ultrasound can distinguish thyroid shrinkage nodules and papillary carcinoma. Conventional ultrasound and contrast-enhanced ultrasound are significant in distinguishing thyroid shrinkage nodules and thyroid papillary carcinoma.

Abstract:
ObjectiveTo investigate the value of iodine base values of the hypoperfusion zone measured by dual-energy computerized tomography (DECT) in the assessment of risk stratification for acute pulmonary embolism (APE).MethodsThe clinical and imaging data of 57 patients with APE were retrospectively analyzed, and the iodine base values of the hypoperfusion area and other related parameters were measured on DECT images. The patients with APE were divided into a high-risk group (20 patients) and a non-high-risk group (37 patients) according to the hemodynamic status. The differences in radiological parameters such as the low perfusion zone iodine base value and clinical parameters were compared between the two groups.ResultsThere was a statistically significant between high-risk and non-high-risk APE patients with age, deep vein thrombosis (DVT), pulmonary trunk diameter/ascending aortic diameter, the diameter of right ventricle (RVD)/the diameter of left ventricle (LVD), pulmonary artery obstruction index (PAOI) and iodine base values in the low perfusion zone (P < 0.05 to P < 0.01). Logistic regression analysis showed that age, DVT, RVD/LVD and PAOI were all independent risk factors for the development of high risk in patients with APE (P < 0.05 to P < 0.01).ConclusionsIodine base values in the low perfusion zone are helpful to assess the severity of APE patients and to guide clinical treatment for the benefit of patients.

Abstract:
ObjectiveTo investigate the control status of risk factors for atherosclerotic cardiovascular disease (ASCVD) high risk residents in a community based on guideline recommendation, which may help general physician focus on guideline-directed management and therapy, and reduce the incidence and mortality of ASCVD.MethodsIn this cohort study, pharmacy and medical claims data from Gumei Community Health Service Center database were queried for residents with high risk for ASCVD. Telephone interview confirmed related information. Control status and related factors were analyzed.ResultsOne thousand one hundred and thirty-nine residents stratified into high risk group for ASCVD were selected, including 941 hypertensive patients and 708 diabetic patients. Among 941 hypertensive patients, blood pressure control rate was 91%, and analysis in subgroups indicated that sex, obesity, smoking and exercise habit were not associated with CR (P>0.05). Among 708 diabetic patients, HbA1c control rate was 60%, which was higher in non-smokers than smokers (62% vs 44%, P < 0.05). In all patients, LDL-C control rate was 62%, which was higher in male vs female, smokers vs non-smokers, and no regular exercise vs regular exercise subgroups (P < 0.05). Binary logistic regression analysis indicated that female, non-diabetic and regular exercise were associated with LDL-C poor control (OR=2.392, 1.383 and 1.545, P < 0.05 to P < 0.01).ConclusionsIn these ASCVD high risk residents, control rate of hypertension was optimal, but control rates of HbA1c and LDL-C were not satisfied. Female, non-diabetic and regular exercise correlates with higher LDL-C level, which neds more attention in these subgroups.

Abstract:
ObjectiveTo explore the effect of temperature on the incidence of ischemic stroke in residents.MethodsThe incidence and meteorological data of ischemic stroke from 2013 to 2020 in Pingyi county sub-cohort of Shandong Ecological cohort were collected. The distributed lag non-linear model (DLNM) was used to analyze the influence of temperature on the incidence of ischemic stroke in the whole population. In addition, the differences between genders and ages were also discussed.ResultsA total of 50 427 people were observed from 2013 to 2020, and the number of cases of ischemic stroke was 42 014, with an average daily incidence of 14 cases. It was found that there was a non-linear relationship between the average daily temperature and the incidence of ischemic stroke. High temperature was related to the increase of the incidence of ischemic stroke, and high temperature had immediate effect and lag effect on the incidence of ischemic stroke. The cumulative relative risk(RR) of high temperature lag in 0-10 days was 1.433 (95%CI: 1.079-1.902). Low temperature and lower temperature had a certain protective effect on the whole population in a specific cumulative period, and the protective effect became weaker and weaker with the passage of time. The RR of low temperature and lower temperature lagging in 0-10 days were 0.618 (95%CI: 0.455-0.840) and 0.712 (95%CI: 0.608-0.834), respectively. The results of stratified analysis showed that high temperature had a great influence on the incidence of ischemic stroke in men and ≥ 65 years old group. When the accumulation of high temperature lagged for 10 days, the risk of ischemic stroke in the male group was the highest, with an RR of 1.678 (95%CI: 1.193-2.361), while for the group aged ≥ 65 years old, the relative risk of ischemic stroke reached the highest on the day of high temperature, with an RR of 1.523 (95%CI: 1.203-1.928).ConclusionsThe temperature has a non-linear relationship with the incidence of ischemic stroke in residents. The high temperature will increase the risk of ischemic stroke in residents, and the cumulative lag effect is greater. Therefore, residents should pay attention to raising awareness of self-protection in hot weather.

Abstract:
ObjectiveTo study the relationship between atmospheric PM2.5 concentration and daily visits of children with wheezing.MethodsUsing the generalized additive model of time series on the basis of controlling for the medium and long-term effects of time, holiday effects, and week effects, we retrospectively analyzed the daily visits of wheezing children in the pediatric emergency department of a tertiary hospital in the eastern region of Shanghai influenced by weather (average daily temperature, relative humidity) and atmospheric pollutants (PM2.5, PM10, SO2, NO2 and O3). The excess risk (ER) of the corresponding concentration range of the above indicators was inferred.ResultsPM2.5 concentration in the eastern region of Shanghai had an impact on the daily visits of wheezing children. Atmospheric PM2.5 concentrations were positively correlated with PM10, SO2 and NO2 concentrations (r=0.780, 0.601, 0.658, P < 0.05), and negatively correlated with relative humidity and temperature (r=-0.015, -0.270, P < 0.05). There was a lagged effect of PM2.5 exposure on daily visits, with the greatest effect occurring with a 1-day lag. The results of the single-pollutant model showed a daily visit ER (95%CI) value of 4.251% (0.824%-7.796%) for each 10 μg/m3 increase in PM2.5 concentration. The two-pollutant model was established by introducing SO2, NO2 and O3 on the same day, respectively, and the daily visit ER (95%CI) values for each 10 μg/m3 increase in atmospheric PM2.5 concentration in PM2.5+SO2, PM2.5+NO2 and PM2.5+O3 (95%CI) were 5.442% (0.747%-10.355%), 5.371% (1.378%-9.522%) and 4.535% (1.481%-7.682%), respectively.ConclusionsExposure to high atmospheric PM2.5 concentration may affect and increase the daily visits of children with wheezing.

Abstract:
ObjectiveTo explore the influence and mechanism of social support on nurses' job performance.MethodsA total of 900 in-service clinical nurses in four hospitals in a city were investigated by job performance scale, perceived social support scale, psychological capital questionnaire and organizational culture scale.ResultsThere was a significant positive correlation between social support, psychological capital and organizational culture and nurses' job performance(P < 0.01). The total effect of social support on nurses' job performance was significant(P < 0.01), the indirect effect of social support on nurses' job performance through psychological capital was significant(95% confidence interval did not include 0), and the direct effect of social support on nurses' job performance was significant(P < 0.01). Social support and organizational culture could significantly and positively predict psychological capital(P < 0.01), and the interaction effect between social support and organizational culture was significant(P < 0.01). The simple slope diagram showed that the positive effect of social support on psychological capital increased with the improvement of organizational culture level. The turning point of the standard score of organizational culture moderating effect was -1.638.ConclusionsSocial support can directly and positively predict nurses' job performance. Psychological capital partially mediates the impact of social support on nurses' job performance. Organizational culture positively moderates the mediating process of social support through psychological capital. The influence of social support on nurses' job performance is a moderated mediating effect.

Abstract:
ObjectiveTo understand the status quo of professional identity of nursing students in higher vocational college, analyze the relationship between empathy, career decision-making self-efficacy and vocational identity, and provide countermeasure and basis for improving professional identity of nursing students in higher vocational college.MethodsA total of 1 269 nursing students in higher vocational college were investigated with general information questionnaire, scale of empathy for nursing students, scale of career decision-making self-efficacy for university students and professional identity questionnaire for nurse students. The influencing factors of professional identity, and the relationship between empathy, career decision-making self-efficacy and professional identity of nursing students in higher vocational college were analyzed.ResultsUnivariate analysis showed that there were significant differences in the professional identity scores of nursing students in higher vocational college with different gender, grade, family atmosphere, relationship with classmates, relationship with teachers, learning status, school record and nursing being the first major or not (P < 0.01). Pearson correlation analysis showed that the total scores and scores of each dimension of empathy of nursing students in higher vocational college were significantly positively correlated with the total scores of professional identity (P < 0.01), and the total scores and scores of each dimension of career decision-making self-efficacy of nursing students in higher vocational college were significantly positively correlated with the total scores of professional identity (P < 0.01). Multiple linear regression analysis showed that the influencing factors of professional identity of nursing students in higher vocational college from high to low were as follows: career decision-making self-efficacy, learning status, nursing being the first major or not, empathy and relationship with teachers (P < 0.05 to P < 0.01).ConclusionsThe stronger the empathy of nursing students in higher vocational college, the higher level of career decision-making self-efficacy and professional identity are. The professional identity of nursing students in higher vocational college needs to be strengthened. School administrators and nursing educators should be aware of the importance of professional identity, and adopt targeted intervention measures to improve professional identity of nursing students in higher vocational college and stabilize nursing team.

Abstract:
ObjectiveTo explore the mediating effect of positive coping style on the relationship between psychological capital and time management of nursing students in the late-stage of practice.MethodsA total of 214 nursing students in the late-stage of practice in a tertiary hospital were selected by convenient sampling method. The survey was conducted with the general information questionnaire, positive psycap questionnaire (PPQ), simplified coping style questionnaire (SCSQ) and time management scale.ResultsNursing students' total score of time management was (176.73±32.45) points. There were significant positive correlations among psychological capital, positive coping style and time management (P < 0.01). The positive coping style played a part of mediating effect between psychological capital and time management, and the mediating effect accounted for 12.44% of the total effect.ConclusionsPsychological capital can indirectly improve time management by promoting positive coping style. The time management capacity of nursing students can be improved by training their psychological capital and positive coping style in the late-stage of practice, so as to improve the practice effect.
