Construction of rat Kir2.3 eukaryotic expression vector and its expression in PC12 cells

Abstract:
Objective: To construct the eukaryotic expression vector of rat potassium channels subunit gene Kir2.3,and to detect its expression in rat adrenal pheochromocytoma cells(PC12).Methods: The total RNA was extracted from the striatum of adult rat brain.The full length cDNA encoding Kir2.3 gene was obtained using the RT-PCR method and subcloned to eukaryotic expression vector pcDNA3-flag by restriction enzymes.The sequence of Kir2.3 gene was confirmed by blasting to Genbank.The recombinant plasmids were transfected into PC12 cells by lipofectamin method.The expression of Kir2.3 gene in PC12 cells was assayed by Western blot and fluorescence microscopy.Results: The sequence of Kir2.3 gene was correct by blasting to Genbank.Western Blot showed that Kir2.3 gene was overexpressed for at least 72 h,which was detected at beginning of 24 h after pcDNA3-flag/Kir2.3 plasmids were transfected into PC12 cells.Conclusions: The eukaryotic expression plasmid pcDNA3-flag/Kir2.3 was successfully constructed and expressed in the dopaminergic like cells-PC12 cells.
Influence of Th1 type related cytokines on phagocytosis of human peripheral neutrophils

Abstract:
Objective: To observe the influence of Th1 cytokines IL-2,IFNγ,and Th1 induced cytokine IL-12 on the phagocytosis of human peripheral blood neutrophils using flowcytometry.Methods: Escherichia coli(E.coli) and Staphylococcus aureus(S.aureus) were labeled with different concentrations of fluorescein isothiocyanate(FITC) in carbonate buffer(CB,pH 9.5) or phosphate buffer saline (PBS,pH 7.2).The FITC labeling rates of the two bacteria were measured by flowcytometry.The whole fresh peripheral blood of the healthy adults were incubated with FITC labeled bacteria for 5 min to 20 min at 37℃,followed by washing and lysing of red blood cells.The percentages of phagocytosis of bacteria by neutrophils were measured by flowcytometry.The whole blood was pretreated with IL-2(20 u/ml),IFN-γ(50 u/ml) or IL-12(30 u/ml) for 2 hours before incubation with FITC labeled bacteria for 5 min at 37℃.The increment of phagocytosis of bacteria by neutrophils was measured and calculated in comparison with control group.Results: The labeling rate of E.coli was higher in alkaline CB,and that of S.aureus was higher in PBS.The percentages of phagocytic neutrophils for E.coli and S.aureus at 5 min were 45% and 38%,respectively,and reached plateau at 15 min to 20 min,with the percentages of 86%-88% and 83%-89% for E.coli and S.aureus,respectively.Pretreated with IL-2(20 u/ml),IFN-γ(50 u/ml) or IL-12(30 u/ml),the phagocytosis rates for E.coli by neutrophils increased by 19%,19%,and 26%,respectively;and that for S.aureus increased by 25%,23%,and 35%,respectively.Conclusions: Th1 type cytokines IL-2,IFN-γ,and Th1 induced cytokine IL-12 can increase human neutrophil phagocytosis,which demonstrates that Th1 type related cytokines enhance the role of neutrophils in innate immunity.
Influence of As2O3 on the reactive oxygen species in melanoma B16 cells

Abstract:
Objective: To study the influence of As2O3 on the proliferation and the reactive oxygen species(ROS) levels in melanoma B16 cells.Methods: B16 cells were treated with 30 μmol/L As2O3 alone or together with 500 μmol/L N-acetylcysteine(NAC).The effect of As2O3 on B16 cells was determined by MTT assay,and the intracellular ROS level was measured by flow cytometry(FCM).Results: As2O3 could obviously inhibit the proliferation of B16 cells and NAC might partially antagonise the apoptosis effect induced by As2O3.ROS level in B16 cells started to rise after being incubated with 30 μmol/L As2O3 for 1 h(P<0.01),and reached the peak point of at 4 h(P<0.01),and the ROS expression levels decreased slightly at 24 h.The difference was all significant compared with the control group(P<0.01).The increasing of ROS might be partially blocked by NAC(P<0.05 to P<0.01).Conclusions: B16 cells apoptosis induced by As2O3 is related to the increase of ROS level.As2O3 induces B16 cells apoptosis by accumulation of intracellular ROS level,which may be one of the antitumor processes of As2O3.
Study on apoptosis of THP-1 cells induced by Mycobacterium tuberculosis 19 kDa lipoprotein

Abstract:
Objective: To abserve the influence of Mycobacterium tuberculosis 19 kDa(Mtb P19) on the apoptosis of THP-1 cells.Methods: Mtb P19 was prepared from cultured Mtb H37Ra.Phorbol myristate acetate-differentiated THP-1 cells were incubated with P19 at the concentrations from 5 μg/ml to 20 μg/ml at 37℃,5% CO2 for up to 4 hours.The change of cells was observed by Wright-Giemsa stain,and the apoptosis rate was detected by flowcytometry.Results: Mtb P19 could induce THP-1 cells apoptosis at final concentration of 5 μg/ml,and the apopotosis rate increased with the concentration of P19(P<0.01).Round shrinkage,pyknosis or nuclear fragmentation were observed in some cells by Wright-Giemsa stain.Conclusions: Mtb P19 induces apoptosis in THP-1 cells in a dose-dependent manner.
Influence of homocysteine on the proliferation and expression of matrix metalloproteinase 2 in A7r5 cells

Abstract:
Objective: To observe the influence of homocysteine(Hcy) on the proliferation and expression of matrix metalloproteinase 2(MMP-2) in A7r5 cells.Methods: A7r5 cells were treated with 0.25,0.5 or 1 mmol/L Hcy for 48 hours.Cells counting and MTT were used todetect the proliferation of A7r5 cells.Semiquantitative reverse transcription-polymerase chain reaction(RT-PCR) was used to examine the production of MMP-2 mRNA.Western blot was used to evaluate the production of MMP-2 protein.Results: The number of A7r5 cells and optical density value increased in a dose-dependent manner when the concentration of Hcy rose.There was statistical significance between groups of 0.25,0.5,1 mmol/L Hcy and control group(P<0.05-P<0.01).Hcy increased the production of MMP-2 mRNA dose-dependently.There was statistical significance between each experimental group and control group(P<0.05).Hcy up-regulated the production of MMP-2 protein dose-dependently.There was statistical significance between each experimental group and control group(P<0.05-P<0.01).Conclusions: Hcy could promote the proliferation of A7r5 cells in a dose-dependent manner at some extent.Hcy could generate the production of MMP-2 dose-dependently both in protein and mRNA level.This effect might be one of the mechanisms involved in the atherosclerosis induced by Hcy.
Effects of milrinone on TNF-α,IL-1 and IL-10 expression during acute lung injury in rabbits

Abstract:
Objective: To observe the effects of milrinone on the expression of TNF-α,IL-1β and IL-10 during acute lung injury(ALI) in rabbits.Methods: Eighteen rabbits were anesthetized with tracheotomy,and then ventilated mechanically.The animals were randomly allocated into three groups:group A(normal control,n=6),group B(endotoxin control,n=6) and group C(treatment group,n=6).Group B and C were induced into ALI by endotoxin.Group C were treated by intravenous milrinone at 5 μg·kg-1·min-1 after a bolus 50 μg/kg;group B and C received normal saline intravenously.The mean systemic arterial pressure(MAP) and mean pulmonary arterial pressure(mPAP) were recorded at the time of baseline(T1),after modeling 0(T2),and 120 min(T3).The levels of TNF-α,IL-1β and IL-10 in serum were detected,and arterial blood gas analysis was made at the same time.Polymorphonuclear neutrophils(PMN)(%) in bronchoalveolar lavage fluid(BALF) and lung wet-to-dry weight ratio(W/D)were determined after the animals were sacrificed.And lung specimens were obtained to observe the histological changes.Results: Compared with group B,the PaO2 in group C increased and mPAP decreased significantly(P<0.01),and the ventilation function improved.After treatment,the concentrations of TNF-α and IL-1β in serum decreased significantly,while the concentrations of IL-10 in serum were significantly enhanced(P<0.01).And PMN(%) in the BALF decreased significantly(P<0.01).Conclusions: Milrinone could improve the pulmonary gas exchange and attenuate endotoxin-induced ALI.The mechanism of the effects may be that milrinone could regulate the imbalance between inflammatory and anti-inflammatory mediators by enhancing the generation of IL-10 and refraining the release of IL-1β and TNF-α.
Establishment of model of isoprenaline-induced chronic heart failure in rats

Abstract:
Objective: To establish the model of isoprenaline (ISO)-induced chronic heart failure(CHF) in rats and to assess the value.Methods: The ISO-induced group(ISO group,n=50) received two subcutaneous injections of 170 mg ISO per kilogram of body and the healthy control group(NS group,n=10) received two subcutaneous injections of 0.25 ml normal saline.The left ventricular end-diastolic dimension(LVEDD),left ventricualr end-systolic dimension(LVESD),left ventricular ejection fraction(LVEF) and shortening fractionl(FS) were detected by echocardiogram at 3,6 and 18 weeks after the injection.Eighteen weeks later,the mass ratio of heart ventricle and left ventricle were measured.Results: As time went on,the degree of CHF became more serious and there was an increase of mortality(P<0.01).Along with the decrease of LVEF and FS,LVEDD and LVESD increased significantly(P<0.01);the mass ratio of heart ventricle and left ventricle increased significantly(P<0.01).Conclusions: High-dose of ISO-induction is a simple,convenient and reliable method to trigger the rat model of CHF.Echocardiographic assessment is instructive in this species.
Influence of vitamin E on the hyperphosph-orylation tau protein contents in hippocampus of Alzheimer disease rats

Abstract:
Objective: To study the influence of Vitamin E on the hyperphosphorilation tau protein in the brains of Alzheimer disease(AD) rats by means of oxidative stress.Methods: Thirty-six AD rats were randomly divided into three groups:sham operation group,model group and model treatment group(each consisting of 12 rats).The models were established by injecting amyloid-β protein into the left amygdale of the rats.The sham operation group received injection of solvent only,and the model treatment group were additionally fed Vit E 10 mg once a day by gastrogavage.The ability of learning and memory of the rats were detected by Y-maze and the expression of Pser-262-tau in hippocampus tissues was estimated by immunohistochemistry methods.Results: The ability of learning and memory of the rats in the treatment group was lower than that in sham operation group(P<0.01),but higher than that in model group(P<0.01).Vietmin E could depress the expression of Pser-262-tau in the hippocampus tissues of AD rats.Conclusions: Vitamin E depresses the expression of Pser-262-tau in the hippocampus tissues of AD rats and improves the learning and memory ability of the rats,suggesting that vietmin E can protect the tau protein from hyperphosphorylation in rats.
Expressions of BDNF and TrkB in salivary adenoid cystic carcinoma and its clinical significance

Abstract:
Objective: To investigate the relationship of the expression of brain-derived neurotrophic factor(BDNF) and tyrosine kinase B(TrkB) to salivary adenoid cystic carcinoma(SACC).Methods: The immunoactivity of BDNF and TrkB in 45 cases of SACC was determined by immunohistochemistry method(streptavidin biotin method).The relationship of the expression of BDNF and TrkB with the histological type,clinical stage,perineural invasion and distant metastasis of the tumor was analyzed.Results: The positive expression rates of BDNF and TrkB were 84.4% and 88.9% in SACC.The statistical analysis showed that the expression level of BDNF and TrkB was positively correlated with the clinical stage and distant metastasis potential(P<0.05-P<0.01),and the expression level of TrkB was positively correlated with perineural invasion(P<0.01).Conclusions: BDNF and TrkB can promote the invasiveness and distant metastasis of SACC,which may act as a predictor of distant metastasis and perineural invasion of SACC.
Efficacy of injection in submucosa of hydroxycamptothecin for prevention of postoperative recurrence of superficial bladder cancer

Abstract:
Objective: To study the efficacy of injection in mucosa of hydroxycamptothecin(HCPT) for prevention of postoperative recurrence of superficial bladder cancer.Methods: Sixty-five cases superficial bladder cancer in our hospital were divided into observation(A) group including 33 cases and control(B) group including 32 cases.The cases of A group were injected in submucosa then instillated.The B group only were treated with intravesical instillation.Results: Sixty-five cases had been followed up for 2 to 6 years.There was only 1 case of recurrence within 54 months,recurrence rate of 3.0%,in A group.There were 6 cases of recurrence,recurrence rate of 18.7%,in B group.Compared with B group the recurrence rate of A group is low(P<0.05).Conclusions: Injection in submucosa of HCPT can effectively prevent postoperative recurrence of superficial bladder cancer with less side effects.
Relationship between common carotid artery intima-media thickness and coronary disease

Abstract:
Objective: To investigate the relationship between common carotid artery intima-media thickness(IMT)and plaque with coronary disease(CHD).Methods: The carotid IMT and plaque score were measured in 90 patients with suspected CHD by color Doppler flow imagining and all the patients were divided into CHD group(60 cases,including 15 cases with stable angina,25 cases with unstable angina and 20 cases with acute myocardial infarction) and control group(non-CHD,30 cases) according to coronary angiography(CAG).Carotid IMT,the plaque index and the detection rate of carotid plaque were compared between CHD group and control group.Results: The IMT and plaque score were statistically higher in group CHD than those in control group(P<0.01).A positive correlation was found between IMT and the number of coronary artery with stenosis(P<0.05);there was no obvious correlation between IMT and the degree of coronary artery stenosis(P>0.05).Conclusions: Common carotid artery IMT is closely related to coronary heart disease.Ultrasonography of carotid artery could be used as a routine method to detect and screen patients with coronary heart disease in early stage.
Role of B-type natriuretic peptide in diagnosis of asymptomatic heart failure

Abstract:
Objective: To evaluate the diagnostic significance of B-type natriuretic peptide(BNP) in asymptomatic heart failure(AHF).Methods: Plasma BNP was determined in 30 patients with AHF and 30 healthy persons served as control.Results: The plasma BNP concentration in the AHF group was significantly higher than that in the control group(150.80±83.75 pg/ml vs 40.05±15.63 pg/ml)(P<0.01).Conclusions: Detection of B-type natriuretic peptide combined with previous medical history and echocardiography may help to the diagnosis of AHF.
Transurethral electrovaporization combined with intravesical instillation of pirarubicin in treatment of glandular cystitis:A report of 22 cases

Abstract:
Objective: To observe the efficacy of intravesical instillation of pirarubicin(THP) combined with transurethral electrovaporization in the treatment of glandular cystitis.Methods: Twenty-two cases of glandular cystitis were diagnosed by cystoscopy and pathological examination and treated with transurethral resection and bladder instillation of pirarubicin.Results: The follow up ranged from 12 to 36 months with mean of 14 months.Of all 22 cases,18 cases were fully cured with transitional cell covering the surface of bladder mucosa gradually,3 cases improved,and 1 case relapsed and recovered after treated with transurethral resection and bladder instillation of pirarubicin again.No case developed adenocarcinoma.Conclusions: The transurethral electrovaporization combined with pirarubicin intravesical instillation is effective in treatment of local recurrence of glandular cystitis.
Treatment of severe acute pancreatitis:A report of 58 cases

Abstract:
Objective: To investigate an optimal therapy for severe acute pancreatitis(SAP).Methods: The clinical data of 58 cases of severe acute pancreatitis treated in our department from June 1998 to January 2006 were reviewed retrospectively,and the therapy and results were analyzed.Results: The overall incidence of complication,fatality rate and cured rate were 44.8%,29.3%,and 70.7%,respectively.The incidence rates of complication in non-surgery group and surgical intervention group were 46.9% and 42.3%,(P>0.05),respectively.The fatality rate in non-surgery group and surgical intervention group were 31.3% and 26.9%,respectively(P>0.05).The cured rates in non-surgery group and surgical intervention group were 71.9% and 69.2%,respectively(P>0.05).Conclusions: Grasp of treatment time,indications and choice of treatment are vital to the prognosis of SAP.
Clinical analysis of 18 patients with reoperative meningiomas

Abstract:
Objective: To investigate the causes of reoperative intracranial meningioma and the methods of reoperation.Methods: The clinical data of 18 patients undergoing reoperation in intracranial meningiomas were analyzed retrospectively.Results: In 18 patients,the complete resection was performed in 14 patients while the incomplete resection was performed in 4 patients.There were no perioperative and postoperative death.All patients were diagnosed with meningiomas by pathology.Conclusions: The grade of resection is the significant factor related to reoperation of meningioma.The residual tumor reoperation rate after incompletely resection is high and necessitates the reoperation.
Clinicopathological study of invasive micropapillary carcinoma of the breast:A report of 14 cases

Abstract:
Objective: To investigate the clinicopathological characteristics and differential diagnosis of invasive micropapillary carcinoma(IMPC) of the breast.Methods: The clinicopathologic information,histopathology and immunophenotype of 14 cases of IMPC of the breast were studied.Results: No special chinical symptoms or signs were observed in the cases.The occurrence of IMPC was 4.8%.The incidence of metastases of the lymph node(78.6%) and the number of metastatic node(on average 7.6) occurred more often in IMPC than that of the invasive ductal carcinoma(41.0%,on average 3.5).Histpathological features manifested that the nest of IMPC was pseudopapillary and was characterized by the tumor cells within artifical hollow space in clustered/tubular shape;the tumor cells were cylindercial or cubic,containing fine granular;the nucleus was oval,deep dyeing,mildly pleomorphic,and mitotic activity was rarely seen.Immunophenotype showed the toumor cells expressed an unique antistaining for epithelical membrance antigen(EMA),and part of the tumor cells expressed ER,PR or c-erbB-2.Conclusions: IMPC is a rare type of breast carcinomas,and its histopathologic feature is pseudopapillary nest of IMPC.The biological behaviour of IMPC is highly maliganant.More attentions should be paid to the differentiation of this tumor from invasive papillocarcinoma,mucinous carcinoma and metastatic carcinoma.
Laparoscopy for varicocele:Report of 113 cases

Abstract:
Objective: To observe the effect of peritonexscopy for treatment of varicocele.Methods: The clinical data of 113 cases of varicocele treated by laparoscopic surgery were reviewed and analyzed retrospectively.All the cases underwent varicocelectomy under laparoscopy.Results: The operation time was 20-80 mins.The mean bowel recovery time was 14.6 hours,and the mean hospital stay was 3.5 days.After operation,16 patients(14.1%) developed scrotal emphysemas.56 cases were followed up for 3-36 months.Varicocele recurred in 2 cases(1.77%) and testicular hydrocele occurred on the affected side in 3 patients(2.66%).Of the 68 previously infertile patients,35 were followed up and 20(57.1%) had obvious improvement in sperm counts and motilities after operation.Conclusions: Varicocelectomy under laparoscopy is safe and effective for treatment of varicocele.
Deproteinized hemoderivative of calf blood injection for the treatment of Alzheimer disease

Abstract:
Objective: To observe the effects of deproteinized hemoderivative of calf blood injection(DHCBI) on Alzheimer disease.Methods: Sixty-six patients were randomly divided into control group and treatment group.The control group received the therapy of piracetam intravenous injection,and the treatment group received intravenous DHCBI.One course lasted fo two weeks,and then the therapeutic effects were observed.Results: The clinical symptoms and the plasma high-sensitivity C-reactive protein and various lipid indicators of the two groups improved(P<0.01),and there were more obvious effects in the treatment group(P<0.05-P<0.01).Conclusions: DHCBI can improve the plasma lipid and the inflammation levels of the elderly with Alzheimer disease.
Docetaxel combination therapy for treatment of non-small cell lung cancer:Report of 35 cases

Abstract:
Objective: To observe the efficacy and side effect of docetaxel combination therapy on advanced non-small-cell lung cancer(NSCLC).Methods: Thirty-five patients with NSCLC were treated by docetaxel 100 mg intravenous d1,combined with cisplatin 100 mg or oxaliplatin intravenous 200 mg d1,respectively.The therapy was repeated every 3 weeks.After 4 cycles the effects and side effects were evaluated by WHO criteria.Results: CR was achieved in 1,PR in 21,NC in 10 and PD in 3.The overall response rate(CR+PR) was 62.86%.The major toxicities were nauser/vomiting,neutropenia and alopecie.Conclusions: Docetaxel-based combination chemotherapy is safe and effective for treatment of advanced NSCLC.
Analgesic effect of lornoxicam or combined with bupivacaine,morphine or buprenorphine injected intra-articularly after knee arthroscopy

Abstract:
Objective: To evaluate the analgesic effect of intra-articular administration of lornoxicam or combined with bupivacaine,morphine or buprenorphine after knee arthroscopy.Methods: One hundred twenty patients from the Hospitals in Tata and the Kindom of Settat in Morocco were assigned to six groups(n=20).Group A received lornoxicam 8 mg in 0.9% NaCl to 20 ml i.a,group B received morphine 2 mg in 0.9% NaCl to 20 ml i.a,group C lornoxicam 8 mg and bupivacaine 0.5% 20 ml i.a,group D morphine 2 mg and bupivacaine 0.5% 20 ml i.a,group E buprenorphine 100 μg and lornoxicam 8 mg in 0.9% NaCl to 20 ml i.a and group F morphine 2 mg and lornoxicam 8 mg in 0.9% NaCl to 20 ml i.a.The intensity of postoperative pain was evaluated by pain intensity(PI),pain intensity difference(PID) and pain relief(PAR) at 0.25,0.5,1,4,8,16,20,24 hour after operation when the passive knee flexion was 90°.The total impression of analgesic effect and side effects related to the drugs were recorded.Results: The PID values were significantly higher in group C,D and F than those in group A and B at 0.25,0.5,16,20,24 h after operation(P<0.01).The PAR values were significantly lower in group C,D,E and F than those in group A and B at 0.25,0.5,16,20,24 h after operation(P<0.01).Group A showed a higher PID value and a lower PAR value compared to group B at 8 and 16 h after operation(P<0.01).The total impression scores of analgesia among group C,D,E and F did not have statistical difference,but their scores were superior to those of the other two groups during the postoperative period(P<0.01).Abdominal distension and nausea were observed in 7 cases in group E and 9 cases in group F.Conclusions: Intra-articular use of lornoxicam combined with bupivacaine,buprenorphine or morphine provides better analgesic effect than the administration of lornoxicam alone after knee arthroscopic surgery.

Abstract:
Objective: To investigate the relations between the serum level of interleukin-6(IL-6) and the severity of acute cerebral infarction.Methods: The serum level of IL-6 in 63 patients with acute cerebral infarction and 24 healthy persons was determined.All the patients were scored by clinic neurological function deficit scale(NDS),and comparision was made between the groups.Results: The serum level of IL-6 of the patients was significantly higher than that of the normal control(P<0.01),and the serum level of IL-6 of the group with serious cerebral infarction was significantly higher than the groups with middle and mild cerebral infarction(P<0.01).Conclusions: The serum level of IL-6 is closely correlated with the severity of cerebral infarction.
Continuous drainage via a lumbar catheter in terminal cistern for treatment of postoperative cerebrospinal fluid leakage accompanied by intracranial infection

Abstract:
Objective: To explore the effective therapeutic strategy of cerebrospinal fluid fistula with intracranial infection in brain.Methods: Ten patients with cerebrospinal fluid(CSF) fistula and intracranial infection in brain admitted to our hospital in past 7 years were treated with lumbar puncture and continuous external drainage,and the relevant clinical material were reviewed and analyzed.Results: The leakages in 10 patients with CSF fistulae accompany by intracranial infection were reduced 1 to 2 days after continuous drainage and 9 patients were cured.The follow-up of 6 months proved no CSF fistula.One of patients suffered from brain infection.Although the infection tissue was resected,the intracranial infection and leakage of cerebrospinal fluid were not under the control.The patient was died at last.Conclusions: The continuous lumbar external drainage is a safe and effective method for curing CSF fistula with intracranial infection in brain.
Application of mechanical anastomosis in reconstruction of alimentary tract for patients with esophageal and cardiac carcinoma

Abstract:
Objective: To investigate the practical value of mechanical anastomosis in reconstruction of alimentary tract for patients with esophageal and cardiac carcinoma.Methods: Esophagogastrostomy was performed in 56 cases who had undergone excision of esophageal and cardiac carcinoma.Results: The operation was successful in all the cases and the time for anastomosis was no more than 10 minites.No one died after the operation.Anastomatic leakage occurred in 1 case,anastomatic narrowness in 3 cases,reflux esophagitis in 3 cases,infections of the lungs in 2 cases and arrhythmia in 8 cases.All the patients with the above-mentioned complications were cured by conservative treatments.Conclusions: Esophagogastrostomy with mechanical suture is safe and reliable,with the advantages of less operation trauma and less ocurrence of complications.

Abstract:
Objective: To explore the effect of free flap pedicled with ramus profundus arteriae plantaris medialis in repairing heel tissue defetcs.Methods: The flap pedicled with ramus profundus arteriae plantaris medialis with an area of 10 cm×8 cm to 4 cm×5 cm was used to repair the heel tissue defets of seven patients who were injured or burned and the clinical effects were observed;the skin grafting was completed at the same time.Results: The flaps of all the seven patients survived and the senses of feeling recovered.Conclusions: It is an ideal procedure to repair the heel tissue defets with flap pedicled with ramus profundus arteriae plantaris medialis,which has the advantages of simple operation,definite effect and fewer complications.

Abstract:
Objective: To investigate the causes and nesssesity of emergency hysterectomy in abnormal uterine hemorrhage of the pregnant and maternal woman and to seek the possible preventive measures for the incidence.Methods: A retrospective analysis was conducted in 30 cases of emergency hysterectomy.Results: Among the 30 cases,29 were cured and 1 died.The main cause of hysterectomy during pregnant period was placental factors;whereas it was infection factors during postpartum period.Conclusions: Hysterectomy is an effective procedure for abnormal uterine hemorrhage,but the time of operation should be considered.

Abstract:
Objective: To observe the effect of selective artery interventional therapy with octreotide on severe acute pancreatitis(SAP).Methods: Sixty-three SAP patients were divided into control group(36 cases) and treatment group(27 cases).The control group received only the routine therapy,while the the treatment group received interventional therapy in addition to the routine therapy.A catheter was placed to the pancreas feeding artery according to CT check and digital subtraction angiography(DSA) device,and octreotide 0.1 mg was supplied every 6 h for 3 to 7 days.The curative effects of the two groups were compared.Results: The blood amylase reduced more quickly in the treatment group than in control group.The gastrointestinal decompression was significantly different between the two groups(P<0.01) and the treatment time was shortened obviously in the treatment group(P<0.01).The complication incidence rate,change surgery rate and fatality rate were 14.81%,3.70% and 0.00%,in the treatment group,and 44.44%,22.22% and 16.67% in the control group,respectively.Conclusions: The curative effect of interventional therapy using octreotide is much better than the routine therapy for SAP.

Abstract:
Objective: To evaluate the clinical effect of tension-free mesh-plug hernia repair(mesh plug) on the aged patients with inguinal hernia.Methods: Eighty aged patients with inguinal hernia who were admitted in our hospital from Oct.1996 to Oct.2006 were reviewed retrospectively.Forty cases received traditional operation(control group) and 40 cases were performed mesh plug hernia repair(treated group).The surgery duration,hospital stay,time to start out-of-bed exercises,complications and recurrence rates were compared between the two groups.Results: Compared with the traditional operation,the mesh plug hernia repair had the advantages of shorter surgery duration(P<0.01),shorter hospital stay(P<0.01) and fewer complications(P<0.05),and patients were able to start out-of-bed exercises earlier(P<0.01).Conclusions: The mesh plug hernia repair is the first choice for treatment of inguinal hernia in the aged patients,and is worth populariting in hospitals of all grades.
Repair of the exposed wound of knee joint with superficial sural artery fascia island flap

Abstract:
Objective: To explore the application of lateral superficial sural artery fascia island flap to restore the exposed wound of knee joint.Methods: Six cases of exposed wound of knee joint were repaired using the lateral superficial sural artery fascia island flap.The supplying region was repaired by skin grafting.Results: All the 6 flaps were survived.The follow-up lasted for one to six months,and the appearance and function of the knee joints recovered completely.Conclusions: Repair of the the exposed wound of knee joint with lateral superficial sural artery fascia island flap does not injure the known veins,and reaches good results for primary repair of both the supplying region and the accepting region,which guarantees the function of the acceptor site.It is an ideal procedure.

Abstract:
Objective: To investigate the effect of surgical therapy for mixed hemorrhoid and to find a way to decrease post-surgery complication.Methods: One hundrend and eighty patients with severe mixed hemorrhoids were randomly assigned to receive hemorrhoidectomy using divest and scissors excision-ligation technique or Milligan-Morgan hemorrhoidectomy.Post-surgery complications and duration of treatment were recorded.Results: The average treatment time was 16.68 days in group one,and 24.63 days in group two.The total score of the first day post-surgery complication was 2.78±1.46 in group one,and 6.5±1.62 in group two(P<0.01).The total score of the fifth day post-surgery complication was 1.28±1.11 in group one,and 5.32±1.22 in group two(P<0.01).Conclusions: Hemorrhoidectomy using divest and scissors excision-ligation technique is easy to operate,with the advantages of a shorter length of hospital stay and a lower complication rate.

Abstract:
Objective: To summarize the etiological factors of neonatal hyperbilirubinemia.Methods: The etiological factors of 160 cases of neonatal hyperbilirubinemia were analyzed retrospectively.Results: The pathogeny of neonatal hyperbilimblnemia was as follows:infection 60(37.5%),breast milk jaundice 42(26.25%),perinatal factors 32(20.0%),unknow causes(10%) and other causes(5.0%).Conclusions: Prevention and treatment of neonatal infection,proper breast feeding,intensive perinatal care,and prevention of fetus hypoxia are essential to decrease the occurrence of neonatal hyperbilirubinemia.

Abstract:
Objective: To assess the value and safety of laparoscope assisted vaginal hysterectomy for normal-positioned uterus.Methods: The clinical data of 52 patients with normal positioned benign uterine diseases treated by laparoscope assisted vaginal hysterectomy(LAVH) were retrospectively reviewed and compared with those of 50 patients receiving total abdominal hysterectomy(TAH).Results: LAVH was accomplished with earlier anus exhaustion,shorter postoperative stay at hospital and less postoperative pain than the TAH(P<0.005-P<0.001).The incidence of vaginal stump polyps was lower than that by TAH(P<0.05),and there was no significant difference in operation duration and bleeding(P>0.05).Conclusions: LAVH is safe and feasible,possessing the advantages of both laparoscopic operation and colpohysterectomy.

Abstract:
Objective: To observe whether there exists difference in the test results of far-sighted ametropia sufferers' eyesight by computer optometry instruments and by streak retinoscope respetively after their pupils were dilated,and to know if the retinoscopy optometry can be replaced by computer optometry.Methods: Twenty-five far-sighted ametropia sufferers who had accepted optometry in 1996 in our hospital were selected at random.After their pulils were dilated,the computer optometry instrument and streak retinoscope were adopted respectively to test the ametropia degree;then the statistical data were analyzed.Results: Examination results by the two methods were significantly different(P<0.01).Conclusions: The test results by computer optometry is not exact.It denotes that the retinoscopy optometry can still not be replaced by computer optometry;as the latter is simple,easy and fast in operation,it can be used as a reference for the former,and applied in popular examination as well.

Abstract:
Objective: To investigate the effect and safety of orthokeratology in treatment of young people with myopia.Methods: The myopia of 94 cases(186 eyes) were corrected by orthokeratology,and the efficacy and the complication were observed.Results: During the follow-up of 6 months,all the 94 cases improved significantly and no severe complications occurred.Conclusions: Ortheratology is safe and effective for treatment of myopia in the teenages with no serious complications.

Abstract:
Objective: To explore the clinical characteristics of children's Mycoplasma pneumonia(MP) in different age groups and to improve the recognition of the disease.Methods: Ninety-four cases of MP were divided into three groups:infants,preschool children and school children,and their clinical data were analyzed retrospectively.Results: Fourty-three cases(45.75%) of the cases were school children.The infants group was significantly different from the preschool children group and the school children group in symptoms,physical signs,sternum and specific anti-MP-IgM antibody titer(P<0.05).However,the preschool children group and the school children group were similar in those aspects(P>0.05).The extra-pulmonary complications occurred in 18 cases,with no significant difference in the incidence rates among the three groups(P>0.05).All the cases responded well to erythromycin and azithromycin.Conclusions: Children's MP occurs most frequently in school children.The clinical characteristics of different groups are relevant to the development level of immunity and immunoreaction.Macrolides antibiotics is effective for treatment of MP.

Objective: To investigate the bioequivalence of amlodipine besylate tablets in healthy volunteers.Methods: A single oral dose of 10 mg test or reference formulations were given to 20 male healthy volunteers in a randomized crossover design.The serum concentration of amlodipine was determined by LC-MS/MS method.Pharmacokinetic parameters and relative bioavailability were calculated with DAS 2.0 program to evaluate the bioequivalence of the two formulations.Results: Pharmacokinetic parameters of amlodipine besylate test formulation were obtained as follows:Tmax was(5.60±3.09) h,Cmax was(8.19±2.2) μg/L,AUC0-144 was(366±100) μg·L-1·h-1,AUC0-∞ was(397±115) μg·L-1·h-1.For reference formulation:Tmax was(6.30±2.85) h,Cmax was(8.22±1.99)μg/L,AUC0-144 was(385.8±80) μg·L-1·h-1,AUC0-∞ was(421±83) μg·L-1·h-1.Calculated with AUC0-144,the bioavailability of two formulation was(96.1±26.6)%.Conclusions: The amlodipine besylate tablets test formulation and reference formulation were bioequivalent.

Objective: To investigate the bioequivalence of valsartan dispersible tablets and capsules in healthy volunteers.Methods: Twenty volunteers were randomly divided into two groups(test and reference),with double cross-over design.The concentration of valsartan in serum was determined by high performance liquid chromatography(HPLC) and pharmacokinetic parameters were calculated with DAS2.0 practical pharmacokinetics program.Results: The pharmacokinetic parameters of valsartan dispersible tablets and capsules were as follows:t1/2 were(3.715±1.895)h and(3.585±1.901)h,Cmax were(1.797±0.539)μg/ml and(1.848±0.647)μg/ml,Tmax were(2.350±0.489)h and(2.425±0.438)h,AUC0-36 h were(10.786±3.794) μg·h-1·ml-1 and(10.52±3.96) μg·h-1·ml-1.The relative bioavalibility of valsartan dispersible tablets was(111.6±40.7)%.Conclusions: The statistical analysis showed that the test and reference preparation were bioequivalent.

Abstract:
Objective: To investigate the HBsAg situation in the newly enrolled college students and to provide a scientific basis for the prevention of hepatitis B in the university.Methods: The venous blood of the freshmen were collected on an empty stomach in the morning and the enzyme-linked immunosorbent assay(ELISA) was used to detect the HBsAg.And the students with positive HBsAg and increased ALT were detected the HBV five indicators by ELISA.Results: The rate of HBsAg infection was 4.56%,in which,male accounted for 5.70% and female 3.69%.Conclusions: The rate of HBsAg infection in the freshmen in the university is much lower than that of the social crowd.

Objective: To study the influence of direct bilirubin(DBIL) and indirect bilirubin(IBIL) on the determination result of serum high-density lipoprotein cholesterol(HDL-C) polyanion by polymer/detergent(PPD) method.Methods: The serum mixed with different levels of DBIL(5.0,17.5,40.5,70.0,102.0 μmol/L)and IBIL(5.0,17.0,40.0,71.5,104.0 μmol/L)was prepared and divided into high[(2.54±0.22)mmol/L],middle[(1.10±0.15)mmol/L] and low[(0.50±0.08)mmol/L] groups according to the level of HDL-C.The level of DBIL and IBIL was determined by HITACHI 7170A automatic analyzer;the level of HDL-C of each serum specimen was determined by using both PPD method and phospho-wolframic acid magnesium precipitation(PTA-Mg2+) method.Results: The interference of DBIL was complicated,the serum specimen within normal DBIL level scope manifested fake advance in the determination result by the PPD method;when the level of DBIL exceeded 20.0 μmol/L,the determination result showed remarkably reduction;when the level of DBIL was 120.0 μmol/L,the HDL-C deviation was-46.29%,-36.89% and-22.73%,respectively in the high,middle and low groups of specimen.The interference of IBIL was not serious,when the level was 104.0 μmol/L,the deviation in each group was within5.0%.Conclusions: When the PPD method is used to determine HDL-C in clinical laboratory,the specimen should be diluted or treated with other experimental techniques to eliminate the interference if the level of DBIL is higher than 40.0 μmol/L.

Abstract:
Objective: To analyze the CT manifestations of transtentorial herniation due to epidural hematoma,and to sudy the relationship between the volume of the hematoma and the degree of the herniation.Methods: Fifty-two cases of epidural hematoma due to cerebral hernia were examined by CT,with the scanning line perpendicular to the line from posterior elinoid process to the lowest point of the clivus(C-C line).Results: Transtentorial hernia was diagnosed by CT in 26 cases,presenting as an isodensity mass attached to the tenporal lobe and located medial to the tentorium in the ambient cistem.The hernias varied in size,being <3 mm(n=3),3-7 mm(n=6) or >7 mm(n=11).The volume of these differnt sized hernias was 31.3-34.5 ml,32.4-42.3 ml and 40.5-69.2 ml.Conclusions: By CT study with the scanning line perpendicular to C-C line,tentorial herniation due to epidural hemstoma can be accurately diagnosed.The volume of the hematoma is consistent with the degree of herniation.

Abstract:
Objective: To explore the significance of ultrasound multiple parameters in distinguishing benign and malignant thyroid nodules,and to screen the sensitive indexes for the differential diagnosis.Methods: Three hundreds and one thyroid nodules were enrolled in this study.The ultrasonograms were retrospectively studied with multiple stepwise regression analysis.Results: With stepwise regression analysis of multivariable binary logistic regression analysis,4 ultrasound evaluation indexes were enrolled:microcalcification,internal echo of nodule,integrity of capsule and nodule boundary.Conclusions: The Logistic model can help differentiate benign from malignant thyroid nodules.

Abstract:
Objective: To investigate the incidence rate of self-image disorder in breast cancer patients who under mastectomy.Methods: One hundrend and two patients with breast cancer were surveyed with self-designed questionnaires.The relations between the occurrrence of self-image disorder and the patients' age,education level and urban and rural areas were analyzed,and the nursing and interference were applied accordingly.Results: The incidence of self-image disorder increased in accordiance with the patients' education level but decreased with the rise of age.The incidence occurred more often in urban patients than in rural ones(P<0.01 to P<0.005).Conclusions: Self-image disorder often occurs in young patients,patients with higher education level and urban patients.More care and interference should be given to them,which is of great value for the prognosis of patients with breast cancer.

Abstract:
Objective: To explore the role of interfering the body temperature of premature infants receiving blue phototherapy in incubator.Methods: Sisty prematures in our hospital were randomly divided into intervention group and control group.The body temperature(T) of the intervention group was taken before blue phototherapy.When the T was between 36.5℃ and 37.5℃,the incubator temperature was decreased 1℃;when the T was <36.5℃,the incubator temperature was decreased 0.5℃;The control group were given blue phototherapy directly.The febrile incidence between the two groups was observed during the treatment.Results: The febrile incidences of the two groups were 20% and 66.7%,respectively,with statistic difference(P<0.005).Conclusions: Regulating the incubator temperature in accordiance to the prematures' temperature taken before blue phototherapy can effectively decrease the incidence of febrile.

Objective: To summerize the causes of leakage of urine in cerebrovascular patients with an indwelling catheter and the points of nursing care.Methods: The balloon catheter was employed.The indwelling catheter was placed according to the aseptic operation of catheterization procedures.The reasons for leakage were concluded,and appropriate care measures were taken.Results: Of the 182 cases using indwelling balloon catheterization,leakage occurred in 44 cases.Of them,32 were brought under control and 12 improved.Conclusions: There are many causes for leakage in the process of using an indwelling balloon catheterization.Nurses should strictly observe the rules and take comprehensive measures to prevent the incidence of leakage of urine.
