2020, 45(4): 437-441, 446. doi: 10.13898/j.cnki.issn.1000-2200.2020.04.005
Abstract:
Objective To investigate the expression of necroptosis pathway related protienin diabetic myocardium, and its possible mechanism. Methods The diabetic model of rats was prepared by intraperitoneal injection of 40mg/kg of streptozotocin on the basis of highcalories diet feeding, and divided into the normal group(CON group, n=6), highcalories group(HC group, n=6) and diabetes group(DM group, n=6).After 10 weeks, the left ventricular end diastolic diameter(LVEDD), left ventricular end systolic diameter(LVESD) and left ventricular ejection fraction(LVEF) were detected using ultrasound. The heart weight(HW), body weight(BW), and heart-body ratio(HW/BW) in three groups were measured after the rats were sacrificed. In three groups, the serum levels of tumor necrosis factor-α(TNF-α), interleukin-6(IL-6) and maleic dialdehyde(MDA) were detected using ELISA method, the apical tissue were stained using Masson, and the changes of RIPK1, RIPK3 and MLKL in myocardial tissue were detected using RT-PCR and Western blotting. Results Compared with the CON group, the expression levels of HW, TNF-α, IL-6 and MDA increased, and the protein expression levels of RIPK1, RIPK3, and MLKL increased(P < 0.05 to P < 0.01), but the changes of the mRNA expression levels of MLKL were not obvious in HC group(P>0.05).Compared with the HC group, the HW/BW increased, the LVEF decreased, the levels of TNF-α, IL-6 and MDA expression increased, the mRNA and protein levels of RIPK1, RIPK3, and MLKL increased(P < 0.01), and the blue staining of Masson aggravated in DM group. Conclusions High glucose status can induce the myocardial injury, increase the myocardial fibrosis and oxidation level, which may be related to the upregulation of the necroptosis pathway.

Abstract:
Objective To investigate the effects of nedaplatin combined with ginsenoside Rg1 on the proliferation of ovarian cancer cells OVCAR-3, and its possible mechanism. Methods The CCK-8 assay was used to detect the effects of ginsenoside Rg1, nedaplatin and their combination on the proliferation of OVCAR-3 cells.Western-blot was used to detect the expression levels of key proteins of Hedgehog pathway(PTCH1, SMO and Gli1).The flow cytometry was used to detect the changes of cell cycle of OVCAR-3 cells after drug intervention. Results Compared with the control group, the proliferation of OVCAR-3 cells in ginsenoside Rg1 group, nedaplatin group and combined drug group were inhibited, and the difference of which among three groups were statistically significant(P < 0.05).At the same time, ginsenoside Rg1 and nedaplatin showed synergistic effects.Compared with the control group, the expression levels of PTCH1, SMO and Gli1 proteins in nedaplatin group, ginsenoside Rg1 group and combined drug group decreased significantly, the difference of which among three groups were statistically significant(P < 0.05), and the synergistic effect between ginsenoside Rg1 and nedaplatin was found.Compared with the control group, only the combined drug group, the cell cycle was arrested at G2 phase(P < 0.05). Conclusions Ginsenoside Rg1 combined with nedaplatin may inhibit the proliferation of OVCAR-3 cells by modulating Hedgehog pathway related proteins to arrest the cell cycle at G2 phase, and ginsenoside Rg1 shows the synergistic effect on nedaplatin.

2020, 45(4): 447-451. doi: 10.13898/j.cnki.issn.1000-2200.2020.04.007
Abstract:
Objective To investigate the effects of hydrogen sulfide (H2S) on spatial learning and memory in diabetic rats, and explore the underlying mechanism. Methods Male rats were randomly divided into normal (CON) group, diabetes mellitus (STZ) group, diabetes+sodium hydrosulfide (NaHS) (SH) group, and normal+NaHS (CH) group, with 8 rats in each group.Type 1 diabetes was induced by a single intraperitoneal injection of streptozotocin.Four weeks after successfully making the model, rats in SH group and CH group were intraperitoneally injected with 56 μmol/kg NaHS solution once a day. After treatment for 4 weeks, the fasting blood glucose (FBG) and body weight (BW) were measured.Spatial learning and memory abilities including escape latency, time spent in the target quadrant, and escape latency to the target quadrant were assessed by Morris water maze. The morphology of hippocampus tissue was observed by HE staining. The levels of total antioxidant capacity (T-AOC), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) in hippocampus tissue were determined by the kits. The mRNA expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) in hippocampus tissue were detected using real time-quantitative polymerase chain reaction. Results Compared with the CON group, there was no significant difference in the various indexes in the CH group (P>0.05).In the STZ group, the escape latency and escape latency to the target quadrant were extended, and the time spent in the target quadrant was shortened (P < 0.01).The histological structure of hippocampus tissue was obviously damaged. The levels of FBG and MDA were increased (P < 0.01), whereas BW, the levels of T-AOC, SOD, GSH-Px activity, and Nrf2, HO-1 mRNA expression were decreased, the difference of which was statistically significant (P < 0.01).Compared with the STZ group, except for FBG, the above indicators were markedly improved in the SH group (P < 0.05 to P < 0.01). Conclusions H2S attenuates cognitive dysfunction in diabetic rats, the mechanism of which might be associated with the inhibition of oxidative stress and up-regulation of the Nrf2/HO-1 pathway in hippocampus tissue.

2020, 45(4): 452-455, 460. doi: 10.13898/j.cnki.issn.1000-2200.2020.04.008
Abstract:
Objective To investigate the effects of inhibiting the expression of ubiquitin specific peptidase 22(USP22) on cisplatin resistance in glioma cells. Methods The cisplatin-resistant glioma cell line(U251/CP2) was induced by gradual elevation of cisplatin concentration. The expression of USP22 in glioma cell line U251/CP2 was silenced using the transfection of USP22 small interfering RNA.The expression level of USP22 mRNA, cell apoptosis, viability and drug resistance index and protein level of USP22 were detected using RT-PCR, flow cytometry, CCK-8 and Western blotting, respectively. Results The cisplatin-resistant glioma cell line of U251/CP2 and USP22 gene silencing cell lines of U251-shUPS22 and U251/CP2-shUSP22 were successfully constrcucted. The inhibition of USP22 expression could significantly increase the apoptosis rate of U251/CP2-shUSP22 cells(P < 0.01), and cisplatin could up-regulate the expression of USP22 protein in U251 and U251/CP2 cells(P < 0.01).The IC50 values to cisplatin of U251-shUPS22, U251/CP2, U251/CP2-shUSP22 and U251 cells were (0.56±0.07)μg/mL, (73.73±2.74)μg/mL, (51.25±1.09)μg/mL and (1.23±0.13)μg/mL, respectively. Conclusions The inhibition of USP22 expression can enhance the chemosensitivity to cisplatin in glioma cells, and reverse the resistance of cisplatin in resistant cell line.It is suggested that USP22 may be one of the potential molecular mechanisms of cisplatin resistance in glioma.

ObjectiveTo observe the effects of nursing intervention cuff pressure combined with nebulized inhalation on improving postoperative throat discomfort in patients with secondary hyperparathyroidism.MethodsNinety patients with secondary hyperparathyroidism were randomly divided into the A, B and C groups(30 cases each group). The group A received general anesthesia, and was admitted to the post-anaesthesia monitoring room (PACU) for routine PACU care immediately after awake extubation. The anesthesia and intubation in group B and group C were the same as the group A, the cuff pressure was dynamically adjusted using external jacket pressure gauge, and maintained at 25~30 cmH2O, group B was admitted to the PACU for routine PAUC care after awake extubation, and the group C was treated with aerosol inhalation of 2 mg budesonide and 5 mL 0.9% sodium chloride solution after awake extubation. The incidence rates and severity of sore throat, dry throat, nausea vomiting and hoarseness in three groups at entering into PACU after extubation(T1), coming out of PACU(T2), postoperative 6 h(T3) and postoperative 24 h(T4) were recorded. The satisfaction of patients was also inquired, and the effects of two nursing methods on the intervention of throat after the general anesthesia were laterally compared.ResultsAt T1, the sore throat score in C group was lower than that in group A and group B (P < 0.01), and the differences of pharyngeal dryness score among three groups were not statistically significant(P < 0.01).At T2, T3 and T4, the scores of sore throat and pharyngeal dryness in group C, group B and group A gradually increased in turn(P < 0.01).The satisfaction in group C was better than that in group B and group A(P < 0.05), and the difference of which between group A and group B was not statistically significant(P>0.05).ConclusionThe nursing intervention cuff pressure combined with atomization inhalation can reduce the postoperative sore throat and pharyngeal dryness, accelerate postoperative recovery of and improve the satisfaction of patients.

Abstract:
ObjectiveTo explore the clinical effect of nursing intervention based on mechanics principle in patients undergoing hip arthroplasty.MethodsA total of 112 patients undergoing hip arthroplasty were divided into two groups according to the nursing methods. The control group(n=56) was treated with routine nursing, and the observation group(n=56) was treated with routine nursing combined mechanics principle-based nursing. The incidence of stress injury, clinical nursing satisfaction and Harris score of hip joint(including pain, joint function, deformity and mobility) were compared between the two groups.ResultsThe incidence of pressure injury in observation group (3.57%) was lower than that in control group (14.39%)(P < 0.05).The total satisfaction of clinical nursing in observation group was 92.86%, which was significantly higher than that in control group (78.57%)(P < 0.01).Harris total score and the scores of pain, joint function, deformity and mobility in observation group were significantly higher than those in control group (P < 0.01).ConclusionThe effect of nursing intervention based on mechanics principle in patients undergoing hip arthroplasty is obvious. It can reduce the occurrence of pressure injury, improve the satisfaction of nursing, and improve the function of hip joint, which is beneficial to the prognosis and worth promoting.
