Abstract:
ObjectiveTo investigate the effect of folate deficiency on the proliferation, apoptosis, expression of spindle assembly checkpoint-related proteins, and its mechanism in L02 cells.MethodsL02 cells were cultured with different concentrations of folate(2 000, 200, 20, 0 nmol/L) for 2 weeks. Then the morphology of L02 cells was observed by inverted microscope, the cell cycle and apoptosis were detected by flow cytometry, cell proliferation was measured by CCK8, the expressions of Mad2, Bub1, BubR1 mRNA were detected by fluorescence quantitative PCR, and the expressions of Mad2, Bub1, BubR1 protein were analyzed by Western blotting.ResultsCompared with the normal control group, folate deficiency made L02 cells to become larger and irregular in shape, reduced the cell viability(P < 0.01), induced S-phase block(P < 0.05), promoted apoptosis(P < 0.05), and upregulated the mRNA and protein expressions of Mad2, Bub1, BubR1 in L02 cells(P < 0.01).ConclusionsIn the culture L02 cells, it induces folate deficiency at 200 nmol/L folate, which causes impaired spindle assembly checkpoint function, abnormal cell proliferation, and even apoptosis in severe deficiency.

2021, 46(5): 566-569. doi: 10.13898/j.cnki.issn.1000-2200.2021.05.002
Abstract:
ObjectiveTo investigate the role of miR-143 in renal cell carcinoma, the effect of sunitinib on the expression of miR-143 and its association with cell proliferation and invasion.MethodsThe human renal carcinoma 786-O cells were divided into control group, miR-143 group, sunitinib group and co-action group. The negative control sequence was transfected into the cells of control group, the miR-143 sequence was transfected into the cells of miR-143 group, sunitinib group cells were treated with sunitinib for 48h, the co-acting group was transfected miR-143 sequence and treated with sunitinib for 48h.Proliferation and invasion of 786-O cells were detected by MTT and transwell experiment, miR-143 RNA expression was detected by qRT-PCR, DNA methyltrans-ferase3B(DNMT3B).The expressions of p-Akt-S473 protein were detected by Western blotting.ResultsThe human renal cancer 786-O cells with stable and high expression of miR-143 were successfully constructed. The expression of miR-143 was increased in the miR-143 transfected group(P < 0.01).The IC50 value in 786-O cells with the treatment of sunitinib for 48 h was 6 μmol/L, which was selected as the drug concentration in the subsequent experiment. The expression of miR-143 in 786-O cells was increased after the treatment of sunitinib for 48 h(P < 0.01).Cell survival rate, cell invasion number, expressions of DNMT3B and p-Akt-S473 protein in miR-143 and sunitinib group were all lower than control group and higher than co-action group(P < 0.05 to P < 0.01).ConclusionsmiR-143 reduced the proliferation and invasion capacity of 786-O cells, which may be related to the decreased expression of DNMT3B and p-Akt regulated by miR-143.Sunitinib may inhibit the proliferation and invasion of 786-O cells by upregulating miR-143.

Abstract:
ObjectiveTo compare the incidence rate of long-term adverse events between radiofrequency ablation and oral drug therapy for atrial fibrillation(AF), and further analyze the predictive value of C reactive protein(CRP) in long-term adverse events of AF patients.MethodsThe patients with AF in the First Affiliated Hospital of Bengbu Medical College were selected, 89 patients were treated with catheter ablation, and 107 patients were treated with oral medication. Two groups were followed up for 18 months, and the incidence rate of chronic stroke, bleeding events and all-cause death in two groups were evaluated. The indicators in two groups were evaluated according to the pre-admission related indicators. The Cox regression analysis was used to predict the risk factors of long-term adverse events, and whether CRP for a predictor of AF long-term adverse events was evaluated.ResultsThe long-term stroke, bleeding events, all-cause death and all events in the patients treated with radiofrequency ablation were less than those in the patients treated with drug. The adverse events time in the radiofrequency ablation group was little compared with in the drug therapy group, but the difference of which was not statistically significant(P>0.05).The CRP was an independent predictor of long-term adverse events in AF patients, and the serum CRP ≥ 6.4 mL/L was associated with the incidence rate of long-term adverse events in AF patients.ConclusionsThe CRP level can be used as a predictor of long-term adverse events, and the high-risk population of patients with AF can be screened using CRP level.

Abstract:
ObjectiveTo evaluate the clinical value of the superficial inferior abdominal artery perforator flap in oral cancer patients by comparing the long-term complications of the donor site between the superficial inferior abdominal artery perforator flap and forearm flap.MethodsThe clinical data of 15 oral cancer patients(observation group) treated with free superficial inferior abdominal artery perforator flaps(superficial inferior epigastric artery perforator flaps or superficial circumflex iliac artery perforator flaps) for extensive resection of lesions and 15 oral and maxillofacial soft tissue defect patients(control group) treated with forearm flaps to repair at the same time were retrospectively analyzed. The long-term complications of the donor site(including paresthesia, hyperplasia of scar and pigmentation) were compared between two groups, and the long-term functional recovery of the donor site was evaluated in two groups.ResultsIn the observation group, the donor wound was directly pulled and sutured after stealth separation, and only linear scar was left after surgery, which had little effect on aesthetics and function. In the control group, the skin grafting was performed on the donor wounds, and the postoperative hyperplasia and hyperpigmentation of scars were obvious. The comparison results of the subjective feelings of patients in the donor site after skin flap surgery showed that there was no statistical significance in the decrease of muscle strength between two groups(P>0.05), and the paresthesia and subjective evaluation of the donor site appearance in observation group was significantly better than those in control group(P < 0.01).In the objective examination of the sensation, function and appearance of the donor site of patients in two groups, the muscle strength decreasing and paresthesia in observation group were better than those in control group(P < 0.05 and P < 0.01), and the color, thickness, vascular distribution, flexibility and total score of scar in observation group were also significantly better than those in control group(P < 0.01).ConclusionsThe superficial inferior abdominal artery perforator flap is a good choice for the repair of oral and maxillofacial soft tissue defects, which has less complications at the donor site and less influence on the appearance and function of the donor site.

2021, 46(5): 591-594. doi: 10.13898/j.cnki.issn.1000-2200.2021.05.008
Abstract:
ObjectiveTo explore the predictive value of the mean platelet volume-to-lymphocyte ratio(MPVLR) in the severity of acute cerebral infarction.MethodsA total of 109 patients with acute cerebral infarction were retrospectively analyzed, and divided into the mild group(n=64) and moderate-severe group(n=45) according to the NIHSS score. The MPVLR was calculated according to the blood routine indexes of two groups. The influencing factors of acute cerebral infarction were analyzed using dichotomous logistic regression, and the predictive value of MPVLR level on admission in the severity of acute cerebral infarction was evaluated using ROC curve.ResultsThe MPVLR and monocyte counts in mild group were lower than those in moderate group(P < 0.05).The results of binary logistic regression analysis showed that the increasing of MPVLR level on admission was an independent predictor of acute cerebral infarction. The results of ROC curve showed that the area under the curve of MPVLR predicting the severity of acute cerebral infarction was 0.745(95%CI: 0.653~0.824), and the best cutoff value, sensitivity and specificity were 5.36, 80.0% and 62.5%, respectively.ConclusionsThe MPVLR has certain predictive value in judging the severity of acute cerebral infarction.

2021, 46(5): 649-652. doi: 10.13898/j.cnki.issn.1000-2200.2021.05.023
Abstract:
ObjectiveTo investigate the correlation between the calcification volume of carotid bifurcation on non-enhanced CT and the stenosis degree of internal carotid artery on digital subtraction angiography(DSA).MethodsThe imaging data of 29 patients with carotid artery scanning were retrospectively analyzed, including non-enhanced CT and DSA scanning.Calcified plaque of carotid bifurcation was segmented in CT image and volume was measured by 3D reconstruction software. The stenosis degree of internal carotid artery was evaluated by DSA, and the stenosis degree >40% was defined as "obvious stenosis".Pearson correlation analysis was used to analyze the correlation between calcification volume and stenosis degree. The ROC curve was used to evaluate the value of calcification volume and predict the obvious stenosis.ResultsThe calcification volume of carotid bifurcation was(0.17±0.10) cm3 in all 29 patients.Among them, 25 patients(86.2%) had stenosis of internal carotid artery with different degrees, in which 17 patients had obvious stenosis.Correlation analysis showed that the calcification volume on non-enhanced CT was moderately correlated with the carotid stenosis degree in DSA(r=0.455, P < 0.05).The ROC curve showed that the cut-off value of the calcification volume used to predict obvious stenosis was 0.07 cm3(P < 0.01) with the sensitivity of 100.0% and specificity of 66.7%.ConclusionsThe calcification volume of carotid bifurcation in non-enhanced CT is related to the stenosis degree of internal carotid artery. The volume over 0.07 cm3 can be used as a screening index to predict obvious stenosis.
