Detection of intracellular phosphorylated protein tyrosine kinases by flow cytometry

Abstract:
Objective: To set up the method of detection of intracellular phosphorylated protein tyrosine kinases(PTKs) by flow cytometry.Methods: After stimulated with PMA and PHA,Jurkat cells were fixed and permeabilized by two different methods,and the phospho-tyrosine monoclonal antibody,P-Tyr-100,and a FITC labeled second antibody were used to detect intracellular phosphorylated PTKs by flow cytometry.Correspondingly the Western-blotting method was also used to compare with flow cytometry.Two-colour immunofluorescence labelling technique was further used to detect phosphorylated PTKs in Mtb-Ag activated γδT cells of peripheral blood mononuclear cells(PBMC).Results: Phosphorylated PTKs in Jurkat cells increased obviously after stimulated with PMA and PHA.The results of flow cytometry were consistent with those of Western-blotting.By using two-colour immunofluorescence labelling technique,the phophorylated PTKs was detected preferentially in γδT cells among PBMCs stimulated with Mtb-Ag.Conclusions: The intracellular phosphorylated PTKs could be detected in single cell by flow cytometry.
Study on apoptosis and expression of its related genes of umbilical vein endothelial cells induced by S.aureus and its L-forms

Abstract:
Objective: To investigate the possible mechanism of apoptosis of human umbilical vein endothelial cells(HUVECs) induced by Staphylococcus aureus(S.aureus) and its L-forms.Methods: S.aureus and its L-forms were used to infect HUVECs by incubation for different intervals of 2 hours,4 hours,6 hours,8 hours and 10 hours.Apoptotic rates in the treated cells were measured by flow cytometry,and expression of bcl-2 and Fas gene were detected by using immunohistochemical staining.Results: Both S.aureus and its L-forms induced apoptosis in HUVECs.The apoptotic rates of treated HUVECs at different time were significantly higher than that of the control (P<0.05-P<0.01).Morever the apoptotic rates induced by L-forms were obviously lower than that by S.aureus(P<0.01).At the 6 hours after infection,Bcl-2 expression in HUVECs teated with S.aureus and its L-forms was significantly lower,but that of Fas significantly higher than that in the control group.Conclusions: Both S.aureus and its L-forms can induce apoptosis of HUVECs which may be resulted from bcl-2 down-regulation and Fas up-regulation.
The effect of homocysteine on the production of nitric oxide and reactivated oxygen species in ECV-304 cells

Abstract:
Objective: To observe the effect of homocysteine on the production of nitric oxide(NO) and reactivated oxygen species (ROS) in ECV-304 cells.Methods: ECV-304 cells were incubated with different of concentrations of Hcy for 24 hours.A nitric-acid reductase method was used to detect NO in different groups with and without three stimuli of nitric oxide synthase(NOS),and ROS in cells was detected by flow cytometry(FCM).Results: After cells were exposed to 0.5 mmol/L,1 mmol/L Hcy for 24 hours,NO in cultured media was significantly less than that in control group (P<0.01).Employing three stimuli for NOS,Hcy still resulted in a gradual decline in NO,and 1.0 mmol/L Hcy produced a statistical suppression of NO (P<0.01) compared with low concentration groups and control group.When incubated with 0.5 mmol/L,1.0 mmol/L Hcy fluorescence in cells detected by FCM was higher than that in control and low concentration groups (P<0.01).Conclusions: Hcy can dose-dependently restrain the excretion of NO and promote the production of ROS.
Apoptosis of lung cancer cell line induced by all-trans-retinoic acid

Abstract:
Objective: To investigate the effect and mechanism of all-trans-retinoic acid(ATRA) in inducing apoptosis of human small cell lung cancer(SCLC) cell line SH-77.Methods: SCLC cell line SH-77 was treated with different concentrations of ATRA.The multiparameter such as cell growth inhibition rate,proliferation index(PI),apoptosis index(AI) and Bcl-2 protein expression was detected by Typan blue stainning,MTT and flow cytometry,respectively.Results: Different concentrations of ATRA could inhibit cell line SH-77 cell growth.G1/G0 stage and AI were increased,while cell count at S stage and PI were reduced.Bcl-2 protein expression was decreased (P<0.01).Conclusions: ATRA can inhibit SCLC cell lines SH-77 growth,induce cell apoptosis and reduce Bcl-2 protein expression.
Dynamic expression of nuclear factor-κB in pancreas of rats with acute pancreatitis

Abstract:
Objective: To observe dynamic expression of NF-κB in pancreas tissues and cytokines in serum and to explore the correlation between NF-κB and cytokine in rats with acute pancreatitis.Methods: Fifty Sprayue-Dawley rats were randomly divided into normal control group,acute edematous pancreatitis group (AEP),3 h acute necrotizing pancreatitis(ANP 3 h)group,ANP 6 h group and ANP 12 h group.The AEP model was successfully made by injecting cerulein into the abdominal cavity of the rats; ANP was induced by intraductal administration of 5% sodium taurocholate.The serum amylase,interlukin-6 (IL-6),tumor necrosis factor-α (TNF-α),and the expression of NF-κB in pancreas tissues and pancreatic pathology were recorded.Results: The expression of TNF-α,IL-6 and NF-κB increased significantly in AEP group(P<0.01).The expression of TNF-α,IL-6 and NF-κB was more evident in ANP group than that in AEP group,which reached the peak at hour 6,then began to come down.Conclusions: NF-κB in pancreas tissues takes an important role in the pathogenesis of acute pancreatitis and the expression of NF-κB has a positive correlation with TNF-α,IL-6.
Experimental study on the influence of spleen on the release of inflammatory mediator in rats with acute pancreatitis

Abstract:
Objective: To investigate the influence of spleen on the production and release of inflammatory mediator in acute pancreatitis.Methods: Acute necrotizing pancreatitis(ANP) was induced in SD rats by injecting 3% sodium taurocholate and trypsin into the biliopancreatic duct.All rats were divided into four groups randomly:sham operation group,splenectomy group,acute pareatitis with splenectomy group,and acute pancreatitis group.The serum levels of TNF-α,IL-1β,IL-6 and IL-10 in each group were examined.The terminal ileum was excised to observe the injury of intestinal mucosal barrier by optical microscope and electronic microscope.Results: The serum levels of TNF-α,IL-1β and IL-10 in the acute pareatitis with splenectomy group were significantly lower than those in the acute pancreatitis group(P<0.01).Histological examination revealed mild edema of mucosal epithelium and normally appearing intact mucosal barrier in the acute pareatitis with splenectomy group,whereas acute pancreatitis group revealed severe injuries including apparent edema,necrosis of the intestinal villi,degeneration of epithelial cell and infiltration of neutrophils.Conclusions: Spleen may promote the production and release of inflammatory mediators in acute inflammatory response,and aggravate the inflammatory response.Splenectomy can reduce the production and release of inflammatory mediators,thus decrease the damage of intestinal mucosal barrier.
Influence of emodin on pancreatitic acinar cell apoptosis in rats with acute pancreatitis

Abstract:
Objective: To investigate the effect of emodin on apoptosis of pancreatic acinar cells in rats with acute pancreatitis.Methods: Thirty SD rats were randomly divided into control group,acute necrotizing pancreatitis(ANP) group,and emodin group.5% sodium dexycholate was injected into the intrapancreatobiliary duct to establish acute pancreatitis models and emodin was injected intraperitonealy to induce pancreatic acinar cell apoptosis.The changes of serum amylase,interlukin-6 and apoptosis index were observed.Results: The level of serum amylase and interlukin-6 was lower in the emodin group than that in the ANP(P<0.01).Apoptosis index was higher in the emodin group than that in the ANP (P<0.01).Conclusions: Emodin can induce pancreatic acinar cell apoptosis,decrease the level of amylase and interlukin-6,and relieve the hemorrhage and necrosis of pancreas.
An experimental study on the gut mucosal barrier dysfunction in rats with acute necrotizing pancreatitis

Abstract:
Objective: To observe the changes in gut mucosal barrier in rats with acute necrotizing pancreatitis(ANP).Methods: SD rats were randomly divided into sham operation group(n=40) and ANP group(n=40).The rat models of ANP were set up by retrograde injection of 5% sodium taurocholate in the biliopancreatic duct.The morphological changes of pancreas and ileum were observed.Plasma levels of intestinal fatty acid binding protein(IFABP) and endotoxin were examined at various time points,the bacterial translocation rates of mesenteric lymph nodes and portal vein were also examined at various time points.Results: The plasma levels of IFABP were elevated obviously 2 hours after ANP induction and reached peak value at 6 hours(P<0.01)and decreased at 24 hours.Plasma endotoxin levels also increased significantly at the early stage of ANP and reached peak value at 48 hours (P<0.01).The rates of bacterial translocation to mesenteric lymph nodes increased sharply 24 hours after SAP induction and reached six-eighths at 48 hours(P=0.013).The bacterial translocation rates of portal vein reached three-eighths at 48 hours(P=0.216).Conclusions: Gut mucosal barrier function can be injured in the early stage of ANP,and result in gut origin bacteria-endotoxin translocation,which may be the originator of septic and secondary infection of the pancreas.IFABP may be a specific marker for identification of gut mucosal barrier dysfunction in the early stage of ANP.
Significance of changes of serum neopterin for patients with acute pancreatitis

Abstract:
Objective: To evaluate the clinical significance of changes of serum neopterin levels in patients with acute pancreatitis.Methods: Twenty-eight patients were divided into mild group and severe group.Their blood samples were obtained at 0,4th,8th and 14th day after admission and the APACHE Ⅱ scores were recorded.Results: Severe patients showed a high neopterin levels,and the correlation between the mean neopterin levels and APACHE Ⅱ was observed at 8th day and 14th day(P<0.05 and P<0.01).In addition,the serum neopterin levels of patients with fatal outcome were much higher than those of surviving patients at 8th day and 14th day(P<0.05 and P<0.001).Conclusions: The rise of neopterin level in patients with acute pancreatitis is related to the severity of pancreatitis.Measurement of serum neopterin may help to predict the severity and prognosis of patients with acute pancreatitis.

Abstract:
Objective: To study the effect of GS on fatigued diaphragm of rats.Methods: Twelve SD rats were divided into two groups at random.Fatigued group (n=6) were induced to fatigue by using a 10 min stimulation program consisting of 30 trains/min of five impulses each at 5 Hz(2 ms impulse duration at supramaximal voltage),and equilibrated for 30 min;GS group were induced to fatigue and perfused 160 mg/L GS solution for 30 min.In the end,Pt,P0,CT,1/2RT,+dT/dtmax,-dT/dtmax and force-frequency relation were measured.Results: Compared with control group,P0,+dT/dtmax,-dT/dtmax of GS group increased;CT,1/2RT of GS group was shorter than that of control group.Conclusions: GS can improve contractility of fatigued diaphragm.
Effect of pituitrin on myocardiac contractile force in the heart of the toad in vitro

Abstract:
Objective: To explore the effect of pituitrin on heart contractile in the toad.Methods: Ringers solution was used to maintain the normal action of the heart,then the heart was perfused by Ringer's solution 1 ml containing pituitrin at concentration of 0.125 u/ml,0.25 u/ml,and 0.5 u/ml,and washed before adding the next dosage.The myocardiac contractile force and heart rate were recorded separately after adding each dosage.The other part of this experiment was that the deslanoside was added to the solution when the heart failure model was caused by pituitrin (0.25 u/ml).The parameters were take down.Results: The myocardiac contractile force was reduced by adding pituitrin compared with control(P<0.05).The reduction of myocardiac contractile force was not antagonized by adding deslanoside.Conclusions: The heart failure can be caused by pituitrin and can't be antagonized by deslanoside in toad.
Evaluation of transcatheter closure of congenital heart defects with occluder

Abstract:
Objective: To evaluate the therapeutic effect of transcatheter closure of patent ductus arteriosus(PDA),atrial septal defects(ASD) and ventricular septal defects(VSD) using occluder.Methods: Twenty-four patients were treated with occluder through the percutancous procedure under fluoroscopy angiography and transthoracic echocardiography(TEE).Results: The success rate was 100%.No residual shunts were found 24 hours after the procedure and 1 week later.Transesophageal echocardiography(TTE) and X-ray examination showed no residual shunts in all the cases.Conclusions: Transcatheter closure of PDA,ASD and VSD with occluder is a simple and efficient method with a high success rate and good occlusion effect.
Management of acute arterial ischemia of the lower extremities: A report of 47 cases

Abstract:
Objective: To evaluate surgical treatment of acute arterial ischemia of the lower extremity and the approach to reducing the complications.Methods: Forty-seven cases of arterial embolism or thrombosis were analyzed retrospectively.Forty-two cases received surgery in addition to thrombolytic therapy,anti-coagulation and vasodilatation,and 5 cases were treated with drugs only.Results: The extremities of 32 cases were saved,11 cases were amputated,4 cases died and 5 cases receiving drug treatment had chronic ischemia.Conclusions: Early diagnosis and proper management are essential to the prognosis of acute arterial ischemia of the lower extremity.
Immediate reconstruction of mucous membrane defect with uncovered buccal fat pad

Abstract:
Objective: To investigate the reconstruction of mucous membrane defects.Methods: Fifty mucous membrane defects in different parts were repaired with the pedicled uncovered buccal fat pad graft.The defects of 20 cases were in the buccal area,18 cases in the soft palate and posterior-superior alveolar process,5 cases in the molar backside,2 cases in the pharynx and 5 cases in the oroantral fistula.Results: All the uncovered buccal fat pads survived.The wound took about 2-3 weeks to heal and its surface became epithelialiced after 4-6 weeks.Conclusions: As the nocovered buccal fat pad is located in the mouth,it is easy to excise and the operation can be completed by only one incision.It has the advantages of good appearance and minimal functional loss.It is a simply and useful method for immediate reconstruction of local mucous membrane defects.
Comparison of MxEP and EP regimen in treatment for advanced non-small cell lung cancer

Abstract:
Objective: To observe and compare the effect and major adverse reaction of the two chemotherapy regimens(MxEP and EP) in treatment of advanced non-small cell lung cancer (NSCLC).Methods: Thirty cases of advanced NSCLC (stage Ⅲ-Ⅳ) patients were treated with MxEP regimen(MxT 10 mg,d2;VP16 75 mg/m2,d1-5;DDP 30 mg/m2,d1-3),and the other 27 cases were treated with EP regimen (VP 16 75 mg/m2,d1-5;DDP 30 mg/m2,d1-3).Results: In the MxEP group,the overall response rate was 56.7%(CP+PR=17),the medium response time was 6 months and the medium survival time was 13 months.In the EP group,the overall response rate was 33.3%(CP+PR=11),the medium response time and medium survival time were 3 and 8 months.The major side effects were anorexia,nausea,vomiting,leucopenia and thrombocytopenia,which were tolerable for most cases.Conclusions: The MxEP regimen is a good choice for treatment of NSCLC.
Effect of high dose of intravenous immunoglobulin on Guillain-Barre syndrome

Abstract:
Objective: To observe the therapeutic effect of high dose of intravenous immunoglobulin (IVIG) in treatment of Guillain-Barre syndrome (GBS).Methods: Eleven patients with GBS were treated with IVIG in a dose of 0.4 g·kg-1·d-1for 5 consecutive days,and the other 16 patients were treated with steroid.The therapeutic effect was compared.Results: The IVIG-treated group was superior to the steroid-treated group in cynical grading scale and time of response(P<0.05 and P<0.001),but there was no significant difference in the total effective rate.Conclusions: Treatment of Guilian-Barre syndrome with high dose of IVIG is superior to that with steroid in clinical grading scale and time of response.
Phacoemulsification for infantile congenital cataract

Abstract:
Objective: To investigate the operative effect of phacoemulsification on infantile congenital cataract.Methods: Of the 43 eyes of 25 infants with congenital cataract, phacoemulsification was performed on 20 eyes of 11 infants,and cataract extracapsular extraction was performed on 23 eyes of 14 infants.All the cases were followed up for 12-18 months.Results: There were significant differences in the occurrence of various complications (P<0.05-P<0.005).Within 12 months, the phacoemulsification group had a lower rate of posterior capsular opcification than the cataract extracapsular extraction group (P<0.05).Conclusions: Phacoemulsification has minimal complications and can relieve or delay posterior capsular opcification.Intact circular capsularhexis will be necessary for stage 2 capsular bag reconstruction.
Treatment of oviductal pregnancy by peritoneoscopy: A report of 30 cases

Abstract:
Objective: To investigate the effect of peritoneoscopy in the treatment of oviductal pregnancy.Methods: The clinical data of 30 cases of oviductal pregnancy treatedby peritoneoscopy operation in our department from Jun.2003 to May 2004 were analyzed retrospectively.Results: All 30 cases were operated successfully by means of peritoneoscopy.Thirteen patients who were willing to bear were retained the oviducts,and the other patients were performed salpingoectomy.The temperature of all the patients returned to normal 72 hours after the operations.No persistent ectopic pregnancy occurred.Conclusions: This technique is the choice of treatment for oviductal pregnancy with the advantages of little injury and fewer complications.
Cryosurgery for rhegmatogenous retinal detachment by using binocular indirect ophthalmoscope

Abstract:
Objective: To observe the effect of cryosurgery for rhegmatogenous retinal detachment(RRD)by using binocular indirect ophthalmoscope.Methods: Eighty six patients(91 eyes) were treated.Binocular indirect ophthalmoscope were used to find holes and to do cryosurgery,scleral compressing.Scleral buckling,releasing subretinal fluid,injecting the C3F8 or SF6 in the vitreous cavity has been done if necessary.Results: Eighty four eyes had retinal reattachment after one operation,the other four eyes had retinal reattachment after twice operation,still other 3 eyes must be done with vitrectomy.Conclusions: The cryosurgery for rhegmatogenous retinal detachment under binocular indirect ophthalmoscope is an effective and safe method.
Ectopic pancreas lying the jejunum: A report of 3 cases and review of the literature

Abstract:
Objective: To explore the clinicopathologic features of ectopic pancreas of the jejunum.Methods: The clinicopathologic data of 3 cases of ectopic pancreas situated in the jejunum were analyzed.Results: Three cases of ectopic pancreas were occasionally confirmed in the operations for gastric ulcer,gallstone of biliary tract or gastric carcinoma respectively.The maximal diameter were from 0.5 to 2.5 cm.Conclusions: Ectopic pancreas of the jejunum is rare and it is often asymptomatic.
Brain-heart syndrome of aged patients with acute stroke: A report of 416 cases

Abstract:
Objective: To investigate the clinical significance and mechanism of brain-heart syndrome (BHS) in the aged with acute stroke.Methods: The clinical data of 416 cases of BHS of the aged with acute stroke were analyzed retrospectively.Results: The incidence of BHS in the aged(38.6%) was higher than that in the young persons (17.4%)(P<0.01),and the occurrence of BHS in98% old patients was in the first week of stroke.The occurrence of BHS was associated with stroke type and lesion location.BHS was found more commonly in acute stroke of brain hemorrhage (76.5%) than in that of embolism(51.3%)(P<0.01).The features of BHS EKG were arrhythmia,similarity myocardial infarction and myocardial ischemia.The abnormality rate of myocardial enzyme was about 46.6%,and about 49% of BHS was associated with low potassium or sodium.The mortality rate of the aged with BHS was 30.29%,which was higher than that of the young persons with BHS(15.15%)and the aged without BHS(15.26%)(P<0.005).Conclusions: The key to the treatment of BHS of aged patients with acute stroke is early diagnosis,high ICP release,EKG monitoring,prevention of pump failure and timely adjustment of arrhythmia.
Alpha-fetoprotein-producing gastric carcinoma: A report of 2 cases and review of the literature

Abstract:
Objective: To explore diagnosis,therapy,and prognosis of alpha-fetoprotein-producing gastric carcinoma (AFPGC).Methods: The data of 87 cases,who had been treated in our hospital and whose level of serum AFP was higher than normal,were reviewed retrospectively.The samples of the gastric cancer patients were examined by means of HE and immunohistochemistry staining.Results: Among the patients whose serum AFP levels were higher than normal,2 were diagnosed as hepatoid adenocarcinoma of the stomach.Their immunophenotypes were positive,for AFP,CEA,α1-AT,and α1-ACT.The routine chemotherapy couldn't control the progress of the disease.Both of them were reported liver metastases.Conclusions: AFPGC have aggressive behavior and its clinical or biological features are quite different from the common AFP-negative gastric cancers.AFPGC is at high risk of liver metastasis,and its prognosis is poor.Curative gastrectomy is the primary choice of treatment.Regimens of adjuvant chemotherapy including mitomycin and irinotecan are recommended,but the development of a novel multi modal therapy against AFPGC is needed.

Objective: To investigate the relationship between the infection rate of head louse and personal-social factors.Methods: The eggs,nymph or adults of pediculus humanus(P. h.) capitis on the head of the children were examined and their personal hygiene habits,parents' education level and occupation,living condition and the children number of the family were investigated.Results: The total infection rate was 19.16%.The male infection rate was 12.81% and female 29.33%(P<0.01).The middle school students' infection rate was 10.00%,and the pupils' 27.67%.The infection rate was higher in students who did not have the habits of often washing their head and having a bath or shared washing tools with their family members.The parents' education level and occupation,living condition and the children number of the family were the factors for the students' infection of P.h.capitis.Conclusions: The high infection rate of P.h.capitis of the rural students is closely associated with the bad personal hygiene habit of the kids,the parents' education level and occupation,their living condition and the children number of the family.
Comparison of barrier function to bacteria between wood pulp spunlace and white cotton cloths from market

Abstract:
Objective: To observe the barrier of wood pulp spunlace against bacteria.Methods: One to four layers of wood pulp spunlace or white cotton cloth from market were put on the surface of culture medium plate.Suspension of E.coli and S.aureus at different dilution were dropped on the dried or wet cloths.After culture,the numbers of bacteria colonies were counted.Results: One layer of dried or wet wood pulp spunlace were able to,but white cotton cloth from market did not,prevent permeation from bacteria.Only four layers of dried white cotton cloth from market were able to prevent the permeating from the bacteria.Conclusions: Wood pulp spunlace had a better barrier function against bacteria than that of white cotton cloth from market.

Abstract:
Objective: To discuss the value of liver blood pool SPECT in diagnosis of hepatic hemangioma.Methods: Twenty-two patients with clinically suspected hepatic hemangioma 1.5-9.1 cm in diameter were examined by hepatic conventional and blood pool SPECT and the results were compared with the histology and radiolgy outcome.Results: Of the 22 cases clinically suspected hepatic hemangioma,18 were diagnosed as hepatic hemangioma by SPECT,which were verified by pathological test (n=12),and clinical,radiological and serum tumor makers follow-up lasting for 2-3 years(n=6).In the other 4 cases,3 were proved hepatogenic cancers and 1 liver cyst by pathology.Conclusions: The liver blood pool SPECT is sensitive and specific in diagnosis of hepatic hemangiomas,and can be used to differentiate benign lesion from malignant lesion.

Abstract:
Objective: To evaluate coronal CT scan in diagnosis of nasal fracture.Methods: The CT images of 120 cases of nasal fracture were compared with their plain-film.Results: All the cases were confirmed fracture by CT,and only 35 cases of fracture were displayed in the plain-film.Conclusions: Coronal CT scan is the most effective means for diagnosis of nasal fracture.

Abstract:
Objective: To investigate the influence of chemotherapy on the immune system.Methods: The serum IL-2,IL-18 and TNF-α in 32 patients with malignant solid tumor were measured by indirect avidin-biotin complex technique and enzyme-linked immunosorbent assay.Results: The experiment showed that the levels of IL-2,IL-18 and TNF-α did not have significant difference before and after chemotherapy one period(P>0.05).Conclusions: The cell-mediated immunity system has strong self-regulated ability when affected by cytotoxic substance in order to maintain its stability and perform its immunity function.
