2021, 46(2): 141-145. doi: 10.13898/j.cnki.issn.1000-2200.2021.02.001
Abstract:
ObjectiveTo investigate the effect of tetramethylpyrazine (TMP) on the permeability of human umbilical vein endothelial cells (HUVECs) induced by lipopolysaccharide (LPS) and the expression of cytoskeleton-related proteins.MethodsHUVECs cultured in vitro were randomly divide into normal control group, LPS group (1 μg/mL LPS), TMP low, medium and high dose group (60 μg/mL TMP+1 μg/mL LPS, 120 μg/mL TMP+1 μg/mL LPS, 240 μg/mL TMP+1 μg/mL LPS), pure TMP group (240 μg/mL TMP).Endothelial cell permeability model was established in Transwell chamber; cell voltage resistance meter was used to measure the trans-endothelial electrical resistance in each group; FITC-dextran method was used to detect the permeability change in each group, and Pull-Down experiment was used to collect Rac1-GTPase active protein, Western blotting was used to detect the protein expression of Rac1-GTPase, Rac1, LIMK1 and p-LIMK1 in each group.ResultsTEER and permeability coefficient showed that three different concentrations of TMP inhibited the decrease of TEER induced by LPS in endothelial cells and decreased LPS-induced endothelial cell permeability(P < 0.05 to P < 0.01);Western blotting analysis showed that TMP could effectively inhibit the LPS-induced up-regulation of Rac1-GTPase, p-LIMK1, Rac1 and LIMK1 in HUVECs(P < 0.05 to P < 0.01).ConclusionsTMP can effectively inhibit the increase of permeability of HUVECs induced by LPS, which may be related to the down-regulation of Rac1-GTPase active protein and LIMK1 phosphorylation protein expression in Rac1/LIMK1 signaling pathway.

2021, 46(2): 210-213. doi: 10.13898/j.cnki.issn.1000-2200.2021.02.019
Abstract:
ObjectiveTo investigate the expression level of miR-377 in ovarian cancer tissues, and analyze the correlation between miR-377 expression, and clinicopathological parameters and prognosis of patients.MethodsFifty ovarian cancer tissue samples and 50 normal ovarian tissue samples were divided into the ovarian cancer group and control group, respectively. The clinical pathological parameters data were collected. The expression levels of miR-377 in two groups were detected using RT-qPCR, and the relationship between mir-377 expression and relevant clinical pathological parameters in ovarian cancer patients were analyzed. The relationship between mir-377 expression and prognosis was analyzed using survival curve, and the independent risk factors of ovarian cancer prognosis using multivariate COX regression method.ResultsThe expression level of miR-377 in ovarian cancer group was significantly lower than that in control group(P < 0.01).The expression level of miR-377 in ovarian cancer group was correlated with the TNM stage, clinical stage and metastasis of tumor(P < 0.05 to P < 0.01), and not related to the age and ovarian cancer type of clinicopathological parameters(P>0.05).The 5-year survival rate of patient with low expression of miR-377(55.88%) was significantly lower than that in patients with high expression(75.00%) (P < 0.01).The results of multivariate COX regression analysis showed that the low expression of miR-377 and TNM staging were the independent risk factors of ovarian cancer prognosis(P < 0.01).ConclusionsThe expression level of miR-377 in the ovarian cancer tissues decreases significantly. The expression levels of miR-377 is related to the TNM stage, clinical stage and pathological grade of ovarian cancer, and the low expression of miR-377 and TNM stage are the independent risk factors of the prognosis of patients with ovarian cancer.

Abstract:
ObjectiveTo explore the effects of anhydrous alcohol by subcutaneous injection on the appearance and morphology of skin, and establish a simple and repeatable animal model of scar.MethodsTwenty-seven New Zealand white rabbits, weighing 1.5 kg, were randomly divided into the A group, B group and C group. The 99.5% alcohol was injected into the dorsal dermis of rabbits in A, B and C groups at the doses of 1, 2 and 3 mL/kg, respectively. The changes of injection sites at 7, 30 and 90 d were observed according to the POSAS scoring standard and microscopic appearance for finding out the best dose.ResultsThe tissue similar to human hypertrophic scar in B group was found at 30 d of injection. The HE staining showed the epidermal layer significantly thickened, and the collagen fiber arrangement disorderedin the dermis. The results of type Ⅰ collagen fiber IHC staining showed that the type Ⅰ collagen was stained for brown, and the brown granules were found in the cytoplasm, and the number of type Ⅰ collagen fibers increased significantly under the microscope.ConclusionsThe hypertrophic scar formed by 2 mL/kg anhydrous alcoholic by subcutaneous injection is similar to the human hypertrophic scar, and can be used as one of the animal models in studying the mechanism of hypertrophic scar and evaluating the treatment.
