2022, 47(2): 141-145. doi: 10.13898/j.cnki.issn.1000-2200.2022.02.001
Abstract:
ObjectiveTo investigate the effects of miR-18b-5p on the invasion and migration of breast cancer cells by down-regulating the expression of neural precursor cell expressed developmentally down-regulated 9(NEDD9), and its mechanism.MethodsThe expression levels of miR-18b-5p in human normal breast cancer epithelial cells MCF-10A and two breast cancer cell lines of SKBR-3 and SKBR-3/PR were detected using qRT-PCR.The surface markers of SKBR-3/PR cell-derived exosomes were detected using Western blotting. The CCK8 and Transwell assay were used to investigate the effects of the transfection of SKBR-3/PR cells with miR-18b-5p inhibitor and extraction of exosomes on the proliferation, invasion and migration of SKBR-3 cells. The targeted binding sites of miR-18b-5p to NEDD9 were predicted using Bioinformatics prediction software Target Scan.Dual luciferase assay was used to analyze the targeted regulation relationship between miR-18b-5p and NEDD9.The effects of up-regulation of NEDD9 on the invasion and migration of SKBR-3 cells were detected using Transwell assay.ResultsThe expression of miR-18b-5p in breast cancer cell line was higher than that in normal cancer cell line(P < 0.01).The expression levels of CD63 and TSG101 specific proteins SKBR-3/PR in cell-derived exosomes were high(P < 0.01).The proliferation, invasion and migration of SKBR-3 cells were significantly inhibited by the extractive exosomes of SKBR-3/PR cells transfected with miR-18b-5p inhibitor(P < 0.01).The prediction results of bioinformatics software Target Scan showed that the NEDD9 3'UTR had potential binding sites with miR-18b-5p, and the miR-18b-5p could negatively regulate NEDD9 gene expression(P < 0.01).The overexpression of NEDD9 could inhibit the invasion of SKBR-3 cells(P < 0.01).ConclusionsThe miR-18b-5p can promote the invasion and migration of breast cancer cells by targeting NEDD9.

2022, 47(2): 146-151. doi: 10.13898/j.cnki.issn.1000-2200.2022.02.002
Abstract:
ObjectiveTo explore the effect of exosomal-miRNAs from tumor necrosis factor-like weak inducer of apoptosis(TWEAK)-stimulated macrophages(TMs) on the metastasis of bladder cancer cells.MethodsThe characteristics of exosomes derived from TWEAK-stimulated TMs, THP-1 induced differentiation were observed.Through the miRNAs microarray analysis, the expressions of miRNAs in exosomes derived from TMs treated with or without TWEAK were detected. The internalization of bladder cancer ATCC5637 cells and miRNAs after the internalization were detected by the laser scanning confocal microscope. The effect of exosome from TWEAK-stimulated TMs on metastasis and signal-pathway in ATCC5637 cells were detected.ResultsTWEAK not only increased the levels of miR-17-3p in TMs and the secreted exosomes, but also resulted in an elevated levels of miR-17-3p in recipient ATCC 5637 cells, which eventually reduced the activity of the EGFR/AKT/ERK 1/2 pathway.Pre-transfection of antagomiR-17-3p in TMs substantially decreased the levels of miR-17-3p in macrophages, its secreted exosomes and the recipient ATCC 5637 cells with a concomitant enhancement of ATCC 5637metastasis, suggesting an involvement of exosomal miR-17-3p from TMs in modulating the metastasis of ATCC 5637cells.And miR-17-3p inhibited the metastasis of bladder cancer cells via the EGFR/AKT/ERK 1/2 pathway.ConclusionsTWEAK transfers exosomal miRNAs to bladder cancer cells via TMs to inhibit the EGFR/AKT/ERK 1/2 pathway, resulting in the inhibition of metastasis in bladder cancer cells.

2022, 47(2): 156-159. doi: 10.13898/j.cnki.issn.1000-2200.2022.02.004
Abstract:
ObjectiveTo investigate the isolation of skin-derived precursors(SKPs) from the skin of paraplegic patients, and induce it into nervous system cells.MethodsThe 2 cm2 skin samples were obtained from the incision edge in patients with thoracic vertebral fracture complicated with paraplegia or not, cut into 1-2 mm3 pieces, and then digested in trypsin for 4 hours. The epidermal layer was removed, and the cells were collected by centrifugation after blowing. The cell phenotype was detected by fluorescence histochemistry, and the cells were induced into the differentiation of the nervous system cells.ResultsThe SKPs could successfully be isolated from the skin of paraplegia patients and there was no statistical significance in the biological characteristics between paraplegics and non-paraplegics(P>0.05).Two kinds of cells from paraplegics and non-paraplegics could express the nestin, specific antigen marker, and be induced into neuron and Schwann cells.ConclusionsSKPs can be isolated from the skin of paraplegia patients, and induced into the nervous system cells. It is suggested that it has a good application prospect in autologous cell transplantation of spinal cord injury.

2022, 47(2): 215-218. doi: 10.13898/j.cnki.issn.1000-2200.2022.02.020
Abstract:
ObjectiveTo expore the effects of the overall burden of cerebral small vessel disease(CSVD) on the prognosis of recombinant tissue plasminogen activator(rt-PA) thrombolysis in ischemic stroke.MethodsA total of 100 patients with ischemic stroke were selected as the research object, and treated with rt-PA thrombolysis.According to the daily living activity(ADL) score of 3 months after the onset of disease, the patients with total score>60 points and ≤ 60 points were divided into the good prognosis group and poor prognosis group, respectively. The basic data, National Institutes of Health stroke scale(NIHSS) and total burden score of CSVD were compared between two groups, and the multivariate logistic regression analysis was used to analyze the effects of the overall burden of CSDV on the prognosis of rt-PA thrombolysis in ischemic stroke.ResultsThere was no statistical significance in the gender, alcohol history, smoking history, hypertension, coronary heart disease and infarct location between two groups(P>0.05).The proportion of patients with age >60 years old in good prognosis group was lower than that in poor prognosis group(P < 0.01), and the infarct area, total NIHSS score and total burden score of CSVD in good prognosis group were lower than those in poor prognosis group(P < 0.01).The results of multivariate logistic regression analysis showed that the age >60 years old, infarct area >3.0 cm2, NIHSS>15 points, overall burden of CSVD >2 points were the independent risk factors of poor prognosis of rt-PA thrombolysis in ischemic stroke(P < 0.05 to P < 0.01).ConclusionsThe greater the age, infarct size, NIHSS, and overall burden of CSVD are, the worse the prognosis of ischemic stroke rt-PA thrombolysis is, and the clinical diagnosis and treatment plan can be adjusted accordingly.

2022, 47(2): 231-236. doi: 10.13898/j.cnki.issn.1000-2200.2022.02.024
Abstract:
ObjectiveTo explore the differences of the bacterial species, drug resistance and inflammatory markers such as C-reactive protein(CRP), procalcitonin(PCT) and white blood cell count(WBC) between hematologic patients and intensive care patients.MethodsIn the contemporaneous area, the patients with hematological disease and intensive care patients with positive blood culture were divided into the hematological disease group(HDG) and non-hematological disease group(NHICG), respectivlely, and the patients with hematological disease and intensive care patients with negatvie blood culture were divided into the control group(HDCG) and negative control group(NCG), respectively. The levels of the PCT, CRP and WBC in all cases were detected. The patients with hematologic diseases were subdivided into the acute lymphoblastic leukemia group(ALLG group), acute lymphoblastic leukemia group(AMLG group) and aplastic anemia group(AAG group). The differences of the blood culture identification, drug sensitivity test results and inflammatory indexes among groups and subgroups were observed.ResultsComparison of the total positive rates between HDG and NHICG groups: the top three were coagulase-negative staphylococcus, Klebsiella pneumoniae and subspecies, and Escherichia coli, with only different sequencing. The bacteria in two groups were mainly Gram-negative bacteria. The positive rate, number of drug-resistant strains and bacteria in the top of drug-resistant rate were still roughly the same in the HDG and NHICG groups.Except NLR, the differences of other inflammatory indicators among HDG, HDCG, NHICG and NCG groups were statistically significant(P < 0.01).The differences of all inflammatory indicators among ALLG, AMLG and AAG subgroups were not statistically significant(P>0.05).ConclusionsThe detection rate and drug resistance rate of bloodstream infection bacteria in patients with hematological diseases and severe patients are relatively concentrated. The types and drug-resistant types of bloodstream infection bacteria in the three common blood diseases are roughly the same. The methicillin-resistant coagulase-negative staphylococcus has become a common Gram-positive pathogenic bacteria and multi-drug resistant bacteria in blood patients and severe patients. The combination of PCT, CRP, WBC and other inflammatory markers is of certain significance for the rapid and effective identification of bloodstream infection.

Abstract:
ObjectiveTo analyze the preoperative visit needs of patients treated with cervical carcinoma radical surgery, and its influencing factors.MethodsA self-designed questionnaire was used to investigate 218 patients treated with cervical carcinoma radical surgery in a grade A tertiary hospital in Qingdao city.ResultsThe total score of preoperative visit needs of patients treated with cervical carcinoma radical surgery was (155.34±22.31) points, the score of psychological support dimension was the highest, followed by hospitalization expense dimension, and the scores of environment information and surgical procedure dimension were the lowest. The results of multiple linear regression analysis showed that education level, age, and type of medical insurance were the factors influencing the preoperative visit needs of patients treated with cervical carcinoma radical surgery(P < 0.05 to P < 0.01).ConclusionsThe preoperative visit needs of patients treated with cervical carcinoma radical surgery are high.It is suggested that the nursing visitors should give more individualized and accurate preoperative guidance according to the specific conditions of the patients, pay attention to the application of psychological intervention measures, reduce the stress response of patients, and improve the satisfaction of patients.

Abstract:
ObjectiveTo observe the intervention effects of simplified finger exercise on the radial artery occlusion and related complications after coronary angiography(CAG).MethodsA total of 326 patients treated with transradial CAG were randomly divided into the observation group(163 cases) and control group(163 cases). The observation group and control group were given the routine care combined with simplified finger exercise and routine care, respectively. The palm-circumference diameter, degree of numbness and pain, and incidence rate of radial artery occlusion after 24 h and 30 d of operation were observed.ResultsThe degree of postoperative limb pain, numbness and swelling in observation group were lower than those in control group(P < 0.05 to P < 0.01).The incidence rate of radial artery occlusion in observation group after 24 h of operation was lower than that in control group(P < 0.05), and there was no statistical significance in the bleeding degree and incidence rate of radial artery occlusion after 30 d of operation between two groups(P > 0.05).ConclusionsThe simplified finger exercise can reduce limb numbness, pain and swelling after CAG via radial artery, and reduce the incidence rate of radial artery occlusion after 24 h of surgery.
