Abstract:
ObjectiveTo compare the effects of incomplete Freund′s adjuvant(IFA) and complete Freund′s adjuvant(CFA) combined with calf Ⅱ collagen(CⅡ) in establishing the collagen-induced rheumatoid arthritis(CIA) rat model.MethodsThirty SD rats aged 8 weeks were randomly divided into the 0.9% sodium chloride solution group(control group), IFA+CⅡ group and CFA+CⅡ group(10 rats in each group). The IFA/CFA and CⅡ were blerding in equal volume, and intradermally injected into the root of rat tail. The 200 μL and 100 μL mixture were injected into rats in the model group on day 0 and day 7, respectively. The same volume of 0.9% sodium chloride solution was injected into rats in the control group using the same method. The scores of arthritis index(AI) in three groups were recorded every 7 days from the initial immunization.HE staining was performed in ankle joints of 5 rats from three groups on day 14 to observe the pathological changes.ResultsOn day 0 and day 7 after modeling, the AI values in three groups were 0.On day 14 after initial immunization, the AI values in IFA+CⅡ group and CFA+CⅡ group were(6.5±0.5) and(7.4±0.4), respectively, and the difference of which was statistically significant(P < 0.01).On day 21 after modeling, the AI values in IFA+CⅡ group and CFA+CⅡ group were(8.8±0.5) and(7.8±0.8), respectively, and the difference of which was statistically significant(P < 0.01).But the difference of AI values in IFA+CⅡ group and CFA+CⅡ group were not statistically significant on day 14 and day 21 after modeling(P>0.05).The results of HE staining of the ankle joint showed that the cartilage surface in ankle joint in the IFA+CⅡ group was rough and slightly damaged, and inflammatory cell infiltration was observed in the articular cavity. In CFA+CⅡ group, the articular cartilage was obviously damaged, presented saw tooth shape, and had obvious pannus and a large amount of inflammatory cell infiltration.ConclusionsBoth IFA+CⅡ and CFA+CⅡ can be used to prepare RA model, and the inflammatory response of IFA+CⅡ is slight, which is the preferred method for preparing RA model.

2022, 47(3): 289-292. doi: 10.13898/j.cnki.issn.1000-2200.2022.03.003
Abstract:
ObjectiveTo study the activation of ferroptosis by resveratrol in colorectal cancer SW480 cells, and to explore its possible mechanism.MethodsAfter intervening SW480 cells with different doses of resveratrol(0, 10, 20, 40, 80, 160 μmol/L) for 24, 48 and 72 h, the effect of resveratrol on cell proliferation inhibition rate was analyzed by CCK8 method. After intervening SW480 cells with different doses of resveratrol(0, 20, 40, 80 μmol/L) for 48 h, Western blotting was used to analyze the expression levels of cystine/glutamate antiporter system Xc(xCT), glutathione peroxidase 4(GPX4) and divalent metal transporter 1(DMT1), and flow cytometry was applied to detect the change of reactive oxygen specie(ROS) level which was indicated by fluorescence intensity.ResultsAfter SW480 cells were treated with resveratrol, the inhibition rate of cell proliferation was significantly increased, which showed a time- and concentr ation-dependent manner(P < 0.05);the expression level of xCT and GPX4 protein were significantly decreased, and the expression level of DMT1 was greatly increased(P < 0.05);resveratrol also induced the accumulation of ROS in SW480 cells(P < 0.05).ConclusionsResveratrol increases the accumulation of ROS by inhibiting the expression of GPX4 and xCT protein to induce ferroptosis in SW480 cells, which may involve the activation of DMT1 expression.

2022, 47(3): 297-301. doi: 10.13898/j.cnki.issn.1000-2200.2022.03.005
Abstract:
ObjectiveTo observe the correlation between sleep disturbance and ecological executive function after acute ischemic stroke.MethodsA total of 103 patients with acute ischemic stroke were selected and divided into sleep disturbance group and normal sleep group according to the Pittsburgh sleep quality index scores.Polysomnography(PSG) and ecological behavior rating inventory of executive function-adult version(BRIEF-A) were used to evaluate sleep quality and executive function, and the difference of which between the two groups was compared.Multiple stepwise regression analysis was applied to explore the correlation between PSG sleep parameters and BRIEF-A scores.ResultsThe total score(GEC), scores of metacognition index(MI) and its factors(organization, planning, working memory, task initiation, task monitoring), and scores of behavior regulation index(BRI) and its factors(conversion, inhibition, self-monitoring, emotional control) of BRIEF-A in sleep disturbance group were higher than those in normal sleep group(P < 0.05 to P < 0.01).Compared with the normal sleep group, the total sleep time(TST) was shortened, the sleep efficiency(SE) decreased, the non-rapid eye movement sleep phase 3(NREM3) decreased, and the rapid eye movement sleep latency(RL), sleep latency(SL), wake-up time after falling asleep, proportion of non-rapid eye movement sleep phase 1 to total sleep time, and proportion of rapid eye movement sleep phase to total sleep time(REM%) increased in the sleep disturbance group(P < 0.05 to P < 0.01).Multiple stepwise linear regression analysis showed that GEC, MI, BRI were negatively correlated with TST and SE(P < 0.05 to P < 0.01), GEC and MI were negatively correlated with the proportion of NREM3 to total sleep time(P < 0.01 and P < 0.05), GEC and MI were significantly positively correlated with REM%(P < 0.01), and GEC was significantly positively correlated with REM%(P < 0.01).ConclusionsPatients with sleep disturbance after acute ischemic stroke have ecological executive function compared with those without sleep disturbance, which are related to sleep parameters in the sleep-wake cycle.

Abstract:
ObjectiveTo analyze the clinical characteristics and related risk factors of frailty syndrome in elderly patients with chronic heart failure(CHF).MethodsA total of 184 patients with frailty syndrome from January 2017 to March 2019 were selected as the study object. The clinical characteristics and related risk factors of frailty syndrome were analyzed by the questionnaires and medical records.ResultsAmong 184 elderly patients with CHF, the frailty syndrome in 66 cases were found, and the incidence rate of which was 35.87%(66/184). The female, age, junior high school education or below, solitary and grade Ⅲ+Ⅳ of the New York Heart Association(NYHA classification), complicated diseases(2-7 kinds) and creatinine level in frailty syndrome group were higher than those in non-frailty syndrome group, and the levels of hemoglobin, albumin and left ventricular ejection fraction in frailty syndrome group were lower than those in non-frailty syndrome group(P < 0.05 to P < 0.01). The results of logistic multivariate regression analysis showed that the age, number of complicated diseases, and levels of hemoglobin, albumin and left ventricular ejection fraction were the independent influencing factors of frailty syndrome in elderly CHF patients(P < 0.05 to P < 0.01).ConclusionsThe risk of frailty syndrome in elderly CHF patients is high, and whcih can be affected by many factors. The management intervention should be strengthened for elderly CHF patients with advanced age and multiple diseases to reduce the occurrence of frailty syndrome.

Abstract:
ObjectiveTo compare the clinical effects of three different postoperative analgesic regimens in laparoscopic total hysterectomy.MethodsNinety patients scheduled by laparoscopic total hysterectomy were randomly divided into the dezocine combined with tramadol group(DT group), dezocine combined with flurbiprofen axetil group(DF group) and dezocine combined with sufentanil group(DS group)(30 cases in each group). The DT, DF and DS groups were treated with 0.5 mg/kg dezocine+10 mg/kg tramadol, 0.5 mg/kg dezocine+3 mg/kg flurbiprofen axetil and 0.5 mg/kg dezocine+1 μg/kg sufentanil for postoperative patient controlled intravenous analgesia(PCIA), respectively. The VAS score ≤3 points was maintained. If VAS score > 3 points, the 0.1 mg/kg dizocine was added in a single dose. The Ramsay sedation score and BCS comfort score among three groups were recorded after 2 h, 4 h, 8 h, 12 h and 24 h of surgery. The dose of dezocine, dose of additional dezocine, number of PCIA, postoperative bowel sounds recovery time, first anal exhaust time, first time out of bed activity, nausea, vomiting, bloating and dizziness among three groups were recorded.ResultsCompared with the DT group and DS group, the postoperative bowel sound recovery time, first anal exhaust time and first time of getting out of bed activity were significantly shortened(P < 0.05), and the postoperative nausea, abdominal distension and dizziness significantly reduced in the DF group(P < 0.05). There was no statistical significance in Ramsay sedation score and BCS comfort score among three groups after 2 h, 4 h, 8 h, 12 h and 24 h of surgery, and the differences of the dose of dezocine, dose of additional dezocine and number of PCIA were not statistically significant(P > 0.05).ConclusionsDezoxin combined with flurbiprofen axetil is more beneficial to the recovery of gastrointestinal function on the basis of satisfying postoperative analgesia.

Abstract:
ObjectiveTo investigate the correlation between homocysteine(Hcy) level and MTHFR 677C/T gene polymorphism in patients with hypertension, and evaluate the effect of different doses of folic acid on different genotypes of hyperhomocysteinemia.MethodsThree hundred and fifty-nine patients with hypertension were screened for Hcy screening and MTHFR 677C/T gene detection. The gene type was divided into type CC, CT and TT, and 50 patients in each gene type were randomly divided into two groups(25 cases in each group), who were treated with 0.8 mg and 5 mg folic acid for 8 weeks to evaluate the therapeutic effect.ResultsAmong 359 hypertensive patients, 119 cases were combined with hyperhomocysteinemia, and the incidence rate was 55.43%. The incidence of hyperhomocysteinemia and plasma Hcy level in male patients were significantly higher than those in female patients(P < 0.01). The results of MTHFR 677C/T gene detection showed that among 359 patients, 161 cases were with type CC, 118 cases were with type CT, and 80 cases were with type TT. The incidence of hyperhomocysteinemia and plasma Hcy level in type TT hypertensive patients were higher than those in type CC and type CT patients(P < 0.01). There was no significant difference in plasma Hcy level between 0.8 mg group and 5 mg group of the same gene type before and after intervention(P > 0.05). The plasma Hcy level in 0.8 mg group and 5 mg group of different gene type was significantly lower than those before intervention(P < 0.01). The difference of Hcy level in different gene type in the same group after intervention was statistically significant(P < 0.01), the Hcy level of type CC in 0.8 mg group and 5 mg group was lower than that of type CT and TT(P < 0.05), and Hcy level of type CT in 0.8 mg group was lower than that of type TT(P < 0.05).ConclusionsThe Hcy level is correlated with MTHFR 677C/T gene polymorphism.Folic acid can effectively reduce hyperhomocysteinemia. There is no significant difference between high-dose folic acid and low-dose folic acid. The effect of folic acid intervention was different with different gene types.

2022, 47(3): 409-413. doi: 10.13898/j.cnki.issn.1000-2200.2022.03.034
Abstract:
ObjectiveTo analyze the influencing factors on sleep quality of shift nurses in operating room using three-factor scale model.MethodsA total of 513 operating room shift nurses were surveyed using the Pittsburgh sleep quality index(PSQI), and the data was analyzed using three-factor scale model(including sleep efficiency, sleep quality and daily life dysfunction).ResultsThe differences of PSQI among nurses with different labor relation, age and shift work experience were statistically significant(P < 0.05 to P < 0.01), while the differences of PSQI among nurses with only child, different marital status and children were not statistically significant(P>0.05).The results of multiple regression analysis showed that the shift experience was an independent influencing factor of sleep quality(P < 0.01).Among different shift work experience nurses, there was no statistical significance in the use of sleep medication and dimension of sleep disorder(P>0.05), and the differences of other dimensions and three factors were statistically significant(P < 0.05 to P < 0.01).The scores of PSQI factor 1, 2 and 3 in the shift nurses were higher than those in no shift experience nurses(P < 0.05), and the scores of PSQI factor 1 and 2 in the shift nurses within 6 months were also higher than those in no shift experience nurses(P < 0.05).The shift work experience was an independent influencing factor of sleep efficiency, sleep quality and dysfunction in daily life(P < 0.01).ConclusionsThe sleep quality of the operating room nurses with shift work experience within 6 months or shift work in progress is poor. The hospital should take appropriate intervention measures to improve the sleep quality of the operating room nurses.

Abstract:
ObjectiveTo investigate the relationship among nurses' psychological capital, occupational well-being and job performance.MethodsA total of 283 registered nurses were selected as the study subjects from two grade A tertiary hospitals in Wuhan city using convenience sampling method. The subjects were surveyed with the questionnaires of the nurse psychological capital scale, occupational well-being scale and job performance inventory.ResultsThe total score of nurses' job performance was(202.22±16.95) points. There were no significant differences in the job performance of nurses with different gender and educational background(P>0.05), while differences in the job performance of nurses with different age, marital status, working year, professional title, department, and monthly income were statistically significant(P < 0.05 to P < 0.01).Pearson correlation analysis showed that the dimensions of nurses' task performance and situational performance were positively correlated with psychological capital, occupational well-being and other dimensions(P < 0.05 to P < 0.01), the anti-production performance was negatively correlated with the hope dimension in psychological capital(P < 0.05), but there was no obvious correlation between the dimensions of task performance and welfare(P>0.05).Multiple linear regression analysis showed that hope, resilience, job value and welfare benefit were the influencing factors of nurses' job performance(P < 0.01), which together explained 50.3% of the total variation.ConclusionsThe job performance of hospital nurses is at a medium level, which varies with age, marital status, working year, professional title, department, and monthly income.Hope, resilience, job value and welfare are the predictors of nurses'job performance.
