Study on CLEC4M in promoting the proliferation and migration of brain astroblastoma

Abstract:
ObjectiveTo investigate the effect of CLEC4M on brain astroblastoma proliferation and migration, as well as the mechanism behind it.MethodsBrain astroblastoma cell lines overexpressing and interfering with CLEC4M were constructed by lentivirus transfection. Real-time quantitative PCR was used to determine the level of CLEC4M expression. The effects of CLEC4M on the proliferation and migration of brain astroblastoma were studied using the CCK-8 test and the cell scratch assay, and the effect of CLEC4M on the cell cycle of brain astroblastoma was studied using flow cytometry. Transcriptome sequencing was used to identify genes in cancer signaling pathways regulated by CLEC4M.ResultsBrain astroblastoma cell lines overexpressing and interfering with CLEC4M were successfully constructed, and the overexpression and interference effect were significant(P < 0.01). CLEC4M increased astroblastoma proliferation and migration, and it mostly affected the G0/G1 and S phases of the cell cycle to regulate cell cycle progression(P < 0.05 to P < 0.01). In addition, overexpression of CLEC4M in brain astroblastoma also modified the expression levels of 5 857 genes, with 2 897 genes highly up-regulated and 2 960 genes significantly down-regulated, according to transcriptome sequencing studies. The differentially expressed genes were largely enriched in ribosomes, cell cycle, and cancer signaling pathways, according to KEGG analysis. CLEC4M interacted with CCNB1, CCND3, APAF1, STEAP3, RRM2B and FAS genes in cancer signaling pathways(P < 0.05 to P < 0.01).ConclusionsCLEC4M can increase the proliferation and migration of brain astroblastoma.
Cytokines emxpression in the supernatant of bone marrow mesenchymal stem cells under inflammatory microenvironment and its effects on osteogenic differentiation

Abstract:
ObjectiveTo investigate the expression level of cytokines in the supernatant of bone marrow mesenchymal stem cells(BMMSCs) under inflammatory microenvironment, and its effects on the osteogenic differentiation of BMMSCs.MethodsWestern blotting was used to detect the expression levels of osteogenic proteins in BMMSCs cultured under normal and inflammatory environment. The expression levels of different cytokines in the supernatant of BMMSCs under normal and inflammatory environment were detected by ELISA, and the content change of each cytokine was compared between normal and inflammatory environment. The expression levels of osteogenic differentiation-related proteins in BMMSCs in each group were detected under the adding cytokines to normal environment.ResultsThe osteogenic ability of BMMSCs in inflammatory environment was not significantly decreased compared with normal environment. The results of ELISA showed that the expression levels of IL-6, IL-8, IL-1β, TGF-β1 and TGF-β2 in inflammatory environment were higher than those in normal environment(P < 0.01). After the different cytokines were added to the normal environment, the expression levels of osteogenesis-related proteins in BMMSCs induced by TGF-β significantly increased.ConclusionsThe TGF-β1 secreted by BMMSCs in an inflammatory environment is mainly involved in the process of osteogenic differentiation.
Study on the effects of WASF3 antisense oligonucleotides on the proliferation and migration of non-small cell lung cancer and its mechanism

Abstract:
ObjectiveTo investigate the effects of antisense oligonucleotides of Wiskott-Aldrich syndrome protein family member 3(WASF3-AS) on the proliferation and migration of non-small cell lung cancer(NSCLC) cells and its mechanism.MethodsThe expression levels of WASF3 in human NSCLC cell lung cancer cell line A549 and human normal lung cell MRC-5 were detected using RT-qPCR. The A549 cells were divide into the blank control group, WASF3-NC group(negative control group) and WASF3-AS group(antisense oligonucleotide WASF3 group). The effects of WASF3-AS on the WASF3 expression in A549 cells were detected using RT-qPCR. The CCK-8 method was used to detect the cell proliferation ability, the cloning experiment was used to detect the cell cloning ability, and the Transwell method was used to detect the cell migration ability. The mRNA and protein levels of the phosphatase and tensin homolog(PTEN), phosphatidylinositol-3-kinase(PI3K) and protein kinase B(p-Akt) in A549 cells of three groups were detected using RT-qPCR and Western blotting, respectively.ResultsCompared with the human normal lung cells MRC-5, the mRNA expression levels of WASF3 in three group increased(P < 0.05 to P < 0.01). There was no statistical significance in the mRNA expression level of WASF3 between the blank control group and WASF3-NC group(P>0.05), but it was higher than that of WASF3-AS group(P < 0.05 and P < 0.01). Compared with the WASF3-NC group, the proliferation levels of cells in WASF3-AS group significantly decreased after 48, 72 and 96 h of transfection(P < 0.05). After 14 days of cell transfection, the clone formation rate and number of cell migration in WASF3-AS group significantly decreased compared with the blank control group and WASF3-NC group(P < 0.05). Compared with the WASF3-NC group, the expression levels of PTEN mRNA and protein in WASF3-AS group significantly increased, while the expression levels of PI3K and p-Akt mRNA and protein significantly decreased(P < 0.05).ConclusionsThe expression level of WASF3 in human NSCLC cells is significantly higher than that in normal lung cells. The WASF3-AS may inhibit the proliferation, cloning and migration of NSCLC by regulating the PTEN-PI3K/Akt signaling pathway.
Study on the miR-431-5p inhibiting the proliferation, migration, invasion and promoting the apoptosis of pancreatic cancer cells by regulating AKT1

Abstract:
ObjectiveTo investigate the effects of miR-431-5p on the proliferation, migration, invasion and apoptosis of pancreatic cancer cells, and its underlying mechanism.MethodsThe qRT-PCR was used to detect the expression levels of miR-431-5p in human normal pancreatic epithelial cells hTERT-HPNE and pancreatic cancer cells CFPAC-1 and PANC-1. After the miR-431-5p mimics were transfected into the pancreatic cancer CFPAC-1 cells to overexpress miR-431-5p, the MTT assay was used to detect the cell proliferation activity, the Transwell assay was used to detect the cell migration and invasion, the flow cytometry was used to detect the apoptosis, and the Western blotting was used to detect the expression levels of Cyclin D1, p21, Bax, Bcl-2, E-cadherin, MMP-2 and AKT1 proteins. The regulatory relationship between miR-431-5p and AKT1 was investigated using the dual luciferase reporter gene experiment.ResultsCompared with hTERT-HPNE cells, the expression levels of miR-431-5p in pancreatic cancer cell PANC-1 and CFPAC-1 significantly decreased(P < 0.05). After the miR-431-5p in CFPAC-1 cells was overexpressed, the cells activity and number of migrating and invading cells decreased(P < 0.05), the apoptosis rate increased(P < 0.05), the expression levels of Cyclin D1, Bcl-2 and MMP-2 protein decreased(P < 0.05), and the expression levels of p21, Bax and E-cadherin protein increased(P < 0.05 to P < 0.01). The results of the dual luciferase reporter gene experiment showed that the miR-431-5p negatively regulated the AKT1 expression in CFPAC-1 cells. After the miR-431-5p and AKT1 in CFPAC-1 cells were overexpressed, the CFPAC-1 cell activity, the number of migrating and invading cells increased, the apoptosis rate decreased, the protein expression levels of Cyclin D1, Bcl-2 and MMP-2 increased, while the protein expression levels of p21, Bax and E-cadherin decreased, and the differences of which were statistically significant(P < 0.05).ConclusionsThe miR-431-5p can inhibit the proliferation, migration and invasion of pancreatic cancer CFPAC-1 cells, and promote apoptosis by targeting AKT1. It may be a potential molecular therapeutic target for pancreatic cancer.
Analysis of the related factors of residual lesions after cervical conization for high-grade cervical intraepithelial neoplasia

Abstract:
ObjectiveTo investigate the residual status and related factors of lesions after conization of high-grade cervical intraepithelial neoplasia(CIN), and explore the necessity of supplementary surgery after conization.MethodsThe clinical data of 200 high-grade CIN(CIN Ⅱ-Ⅲ) patients treated with conization and total hysterectomy one month after surgery were retrospectively analyzed. The correlation between the age, pregnancy history, HR-HPV genotype, CIN grade, conic resection method, resection margin status and residual lesions were analyzed using univariate and logistic regression model.ResultsOf the 200 patients, the residual lesions in the hysterectomy specimens in 98 cases(49.0%) were found, and there was not correlation between the age, birth rate, coning method and postoperative residual lesions. The CIN grade, positive preoperative HPV-16 or HPV-18 and margin status were positively correlated with the residual lesions. The results of logistic regression analysis showed that the CIN grade, positive margin and HPV-16 or HPV-18 test results were the high risk factors of residual lesions.ConclusionsThe CIN grade, positive margin and preoperative HPV-16 or HPV-18 genotype are the reliable predictors of residual lesions after conization for high-grade CIN.
Risk factors of cerebral microbleeds in ischemic stroke and the correlation between cerebral microbleeds and cognitive function

Abstract:
ObjectiveTo explore the risk factors of cerebral microbleeds(CMBs) and the correlation between CMBs and cognitive function in patients with ischemic stroke.MethodsThe data of 70 patients with ischemic stroke were collected. According to SWI, they were divided into cerebral microbleeds group(CMBs group, n=29) and non-cerebral microbleeds group(N-CMBs group, n=41). The data of the two groups were collected and the risk factors of CMBs were analyzed. MoCA and MMSE scale were used to evaluate the cognitive function of the two groups, and the correlation between CMBs and cognitive function was analyzed.ResultsAge, hypertension, history of antiplatelet drugs, leukoaraiosis and history of stroke were the risk factors for CMBs(P < 0.05 to P < 0.01). There were significant differences in the scores in MoCA, MMSE and the number of cases of cognitive impairment between CMBs group and N-CMBs group(P < 0.01). Spearman correlation analysis was carried out between cognitive function and the degree of CMBs, the results showed that MoCA score was negtively correlated with CMBs degree(rs=-0.889, P < 0.05). MMSE score was negtively correlated, and with CMBs degree(rs=-0.726, P < 0.05).ConclusionsCMBs can lead to the decline of cognitive function, and more numbers of CMBs can induce the more severe cognitive impairment. Age, hypertension, history of antiplatelet drugs, leukoaraiosis and history of stroke are the risk factors of CMBs, and age is the independent risk factor of CMBs.
Influence of different time of administration of parecoxib in three doses method on postoperative analgesia of gallbladder stones

Abstract:
ObjectiveTo explore the effects of different time of administration of parecoxib three doses method on postoperative analgesia of gallbladder stones.MethodsOne hundred and fifty-two patients with gallbladder stones were investigated using a prospective randomized controlled study, and divided into the control group and observation group according to random number table method(76 cases in each group). Two groups were treated with laparoscopic cholecystectomy. The control group was given parecoxib empirically for 40 mg once after surgery, the patients with incision pain were given the medicine one more time, and the total dose was not more than 80 mg a day. The observation group was given parecoxib three doses method after 0 h, 8 h and 16 h of surgery, 20 mg each time for postoperative analgesia. The serum stress-related factors norepinephrine(NE), blood glucose(Glu), cortisol(Cor), pain degree(VAS score), sedation[Ramsay sedation score, agitation(Riker-SAS agitation score), cognitive function(MMSE score)], incidence rates of adverse reactions(including nausea, pruritus, vomiting, flatulence, edema at injection site), postoperative painless time of getting out of bed, morphine dosage, length of hospital stay, total hospitalization cost and patient's satisfaction between two groups before operation(T0), and at postoperative 3 h(T1), postoperative 9 h(T2) and postoperative 18 h(T3) were compared.ResultsAt T1, T2 and T3, the serum levels of NE, Glu and Cor in two groups were higher than those at T0(P < 0.05), but those in observation group were lower than those in control group(P < 0.01). The VAS scores in observation group at T1, T2 and T3 were lower than that in control group(P < 0.01). At T1 and T2, the Ramsay sedation score in observation group was higher than that in control group, and lower than that at T3(P < 0.01). The Riker-SAS agitation score in observation group at T1 and T2 were lower than that in control group, and which in observation group at T3 was higher than that in control group(P < 0.01). The total incidence rate of adverse reaction in observation group was lower than that in control group(P < 0.05). Compared with the control group, the postoperative painless time of getting out of bed and hospitalization time in observation group were shorter(P < 0.01), and the amount of use morphine and total hospitalization cost in observation group were lower than those in control group(P < 0.01). The satisfaction of postoperative analgesia in observation group was higher than that in control group(P < 0.05).ConclusionsThe parecoxib three doses method can significantly reduce the serum levels of stress-related factors, improve the effect of analgesia and sedation, reduce the incidence rate of adverse reactions, promote the postoperative recovery of patients and reduce the amount of morphine and total hospitalization costs, which has high patient's satisfaction.
Infiltration and significance of tumor infiltrating lymphocytes in triple negative breast cancer

Abstract:
ObjectiveTo count the tumor infiltrating lymphocytes(TILs) in the primary tumor nests and stroma in patients with triple negative breast cancer(TNBC), and analyze the infiltration of TILs in TNBC and its relationship with clinical prognosis.MethodsForty-five patients with TNBC were selected from The First Affiliated Hospital of Bengbu Medical College. The expression of TILs in primary tumor nests and stroma were counted by HE staining, and the relationship between TILs and clinical prognosis was analyzed according to the corresponding clinical data and pathological features.ResultsThe stromal TILs(STILs), intracellular TILs(ITILs) and global TILs were the protective factors of overall survival(OS) and disease-free survival(DFS) in TNBC patients. The number of positive lymph nodes and clinical stage were the risk factors of OS and DFS in TNBC patients. The higher the infiltration levels of STILs, ITILs and TILs were, the better the prognosis was.ConclusionsThe levels of STILs, ITILs and TILs are the dependent prognostic factors in patients with TNBC.
Effect of atorvastatin on TLR4 signaling pathway and cognitive function in elderly patients with cerebral infarction

Abstract:
ObjectiveTo investigate the changes of Toll-like receptor 4 (TLR4) signaling pathway in elderly patients with cerebral infarction complicated with cognitive dysfunction, and the role of atorvastatin in regulating this pathway and its influence on cognitive function.MethodsA total of 98 elderly patients with cerebral infarction and cognitive dysfunction were randomly divided into the observation group and control group(49 cases in each group). The control group was treated with aspirin enteric-coated tablets on the basis of routine treatment, while the observation group was treated with aspirin enteric-coated tablets combined with atorvastatin on the basis of routine treatment. The cognitive function, serum oxidized low-density lipoprotein(ox-LDL), transforming growth factor-β1(TGF-β1), high-sensitivity C reactive protein(hs-CRP), total cholesterol (TC), triglyceride(TG), low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C), TLR4 and nuclear transcription factor(NF)-κB, tumor necrosis factor(TNF)-α and interleukin(IL)-6 and the occurrence of adverse reactions in the two groups were compared before and six months after treatment.ResultsAfter treatment, there were statistically significant differences in the levels of TC, TG, LDL-C, HDL-C, ox-LDL, hs-CRP, TGF-β1, TLR4, NF-κB, TNF-α and IL-6 between the two groups compared with before treatment(P < 0.05 to P < 0.01);the levels of TC, TG, LDL-C, ox-LDL, hs-CRP, TLR4, NF-κB, TNF-α and IL-6 in the observation group were lower than those in the control group, and the levels of HDL-C, TGF-β1 were higher than those in the control group(P < 0.01). After treatment, the MMSE and MoCA scores in the two groups were improved compared with those before treatment, and which in the observation group was higher than that in the control group(P < 0.01). There were no significant differences in the incidence of adverse reactions such as abnormal liver and kidney function, dizziness and headache, rash, myalgia and gastrointestinal reactions between two groups(P>0.05).ConclusionsThe TLR4 signaling pathway is involved in the occurrence of cerebral infarction combined with cognitive dysfunction in elderly, and atorvastatin can exhibit the cerebral protective role in regulating TLR4 signaling pathway.
Efficacy of the sufentanil patient-controlled intravenous analgesia combined with anterior quadratus lumborum block in elderly lower limb surgery

Abstract:
ObjectiveTo investigate the value of sufentanil patient-controlled intravenous analgesia combined with anterior quadratus lumborum block in lower limb surgery in the elderly.MethodsEighty-six elderly patients treated with lower limb surgery were divided into the observation group and control group by random number table method(43 cases in each group). After the operation, the affected side of the observation group received anterior quadratus lumborum block under the guidance of ultrasound, and 30 mL of 0.447% ropivacaine mesylate was injected. Two groups received patient-controlled intravenous analgesia of sufentanil. The static and motor visual analog scale(VAS), sufentanil dosage and delirium occurrence in two groups were analyzed after 6 h(T1), 12 h(T2), 24 h(T3) and 48 h(T4) of operation, and the maximum flexion and abduction of hip joint in two groups at T2, T3 and T4 were observed. The levels of serum inflammatory factors[(including tumor necrosis factor α(TNF-α), C reactive protein(CRP) and interleukin-6(IL-6)] were compared between two groups on the next day after surgery, and the incidence rates of adverse reactions within 48 h after surgery in two groups were analyzed.ResultsThe static and motor VAS scores, dosage of sufentanil and incidence rates of delirium in observation group were lower than those in control group at T1, T2, T3 and T4(P < 0.05 to P < 0.01). The maximum flexion and abduction of the hip joint in observation group at T2, T3 and T4 were higher than those in control group(P < 0.01). The levels of TNF-α, CRP and IL-6 in observation group were lower than those in control group on the next day after operation(P < 0.01). The incidence rate of adverse reactions within 48 h after operation in observation group was lower than that in control group(P < 0.05).ConclusionsThe sufentanil patient-controlled intravenous analgesia combined with anterior quadratus lumborum block in elderly lower limb surgery has a precise postoperative analgesia effect, which can reduce the postoperative sufentanil dosage, incidence rate of postoperative delirium, level of early serum inflammatory factors, and has high safety.
Determination of peripheral blood neutrophil extracellular traps and its significance in patients with gout

Abstract:
Objective To compare the expression levels of neutrophil extracellular traps(NETs) between gout patients and healthy controls, and explore the possible role of NETs in gout. Methods The serum levels of free NETs in 40 gout patients and 24 healthy controls were detected by MPO-DNA ELISA, and the association between serum NETs and ESR, CRP, uric acid, length of the course of the disease were analyzed. The difference of the serum level of NETs between the colchicine group(19 cases) and noncolchicine group(21 cases) was compared. Results The serum levels of NETs in gout group[(2 119.62±1 843.36)pg/mL] were significantly higher than that in healthy control group[(1 894.66±113.52)pg/mL](Ρ < 0.01). The serum level of NETs in gout group was positively correlated with ESR and CRP(P < 0.05 and P < 0.01), but not correlated with uric acid and length of the course of the disease(P>0.05). The serum level of NETs in gout group treated without colchicine[(2 138.24±173.60) pg/mL] was high compared with the gout group treated with colchicine[(2 099.05±196.23) pg/mL], but the difference of which was not statistically significant(P>0.05). Conclusions The neutrophils may be involved in the pathogenesis of gout arthritis by forming NETs in a new mode of death. The serum level of NETs may be related to the gout disease activity.
Effect of dexmedetomidine combined with dyclocainemucilage analgesia on the incidence rate of postoperative catheter related bladder discomfort after general anesthesia

Abstract:
Objective To investigate the effects of dexmedetomidine combined with dyclonine mucilage analgesia on the incidence rate of postoperative catheter related bladder discomfort(CRBD) after general anesthesia. Methods A total of 90 male patients with general anesthesia were divided into the dexmedetomidine group(group D), dexmedetomidine combined with dclonine mucilage(group DD) and control group(group C) according to the random number table method(30 cases each group). Before induction of anesthesia, the 1.0 μg/kg dexmedetomidine was intravenously injected into the group D and group DD. After induction, the vulva and urethral orifice in the group DD were routinely disinfected, and the 5 mL dacronine glue was injected into the group DD through the urethra for analgesia, and 2 mL dacronine glue was evenly smeared on the urethra for lubrication. The group C were treated with equal amount of 0.9% sodium chloride solution for routine urethral catheterization. The changes of hemodynamic indexes, agitation score(RS score), Ramsay sedation score, anesthesia recovery time and incidence rate and degree of postoperative CRBD in three groups were observed and compared before anesthesia induction(T0), before extubation(T1), during extubation(T2) and after 15 min of extubation(T3). Results Compared with T0, the systolic blood pressure(SBP), diastolic blood pressure(DBP) and heart rate(HR) in the group C at T1, T2 and T3 significantly increased(P < 0.05), the SBP, DBP and HR in group D at T1 and T2 significantly increased(P < 0.05), while there were not significant changes in the group DD(P>0.05).Compared with the group C, the SBP and HR at T2 and T3, and DBP at T2 in the group D were lower(P < 0.05), and the SBP and HR at T1, T2 and T3, and DBP at T2 in the group DD were lower(P < 0.05).Compared with the group D, the SBP in the group DD at T1 and T2 were lower(P < 0.05), and there was no significant difference in the rest(P>0.05).Compared with the T0, the RS score and Ramsay sedation score in the group D and DD at T1, T2 and T3 were significantly decreased(P < 0.05), while there were not significant changes in group C(P>0.05).Compared with the group C, the RS scores were lower(P < 0.05), and the Ramsay sedation scores were higher in the group D and group DD at each time point(P < 0.05).Compared with the group D, the differences of the RS scores in group DD among all time points were not statistically significant(P>0.05), and the Ramsay sedation scores at all time points except T0 were higher(P < 0.05).The differences of the respiratory recovery time, recovery time and extubation time among three groups were not statistrically significant(P>0.05).Compared with the group C, the incidence rate of postoperative CRBD in group D and group DD were lower(P < 0.01), and the degree of CRBD was less(P < 0.01).Compared with the group D, the incidence rate of postoperative CRBD in group DD was lower(P < 0.01), and the degree of CRBD was milder(P < 0.01). Conclusions The dexmedetomidine combined with dyclonine mucilage analgesia can effectively inhibit the nerve impulses of patients with general anesthesia in the department of urology, reduce the local adverse stimulation, make hemodynamic parameters more stable in the process of anesthesia, relieve the degree of agitation and suffering of patients, effectively prevent the CRBD, reduce the level of the CRBD and increase the comfort and safety of urethral catheterization.
Study on the expression of Piwil2 protein in malignant phyllodes tumor tissue and its clinical prognosis

Abstract:
Objective To investigate the expression levels of Piwil2 protein in breast malignant lobar tumor tissues, and analyze the relationship between Piwil2 protein expression and clinicopathological factors related to breast malignant lobar tumor and its influence on 10-year survival and prognosis of patients. Methods The clinical data of 50 female patients with malignant lobular tumor of breast were collected, and 70 cases of normal breast tissue resected due to mastitis were selected as the control group. The expression levels of Piwil2 protein in malignant lobar tumor and normal breast tissues were detected by EnVisionTM immunohistochemistry. Results The positive expression rate of Piwil2 protein in malignant lobar tumor tissue was 86.00%, which was significantly higher than that in normal breast tissue(0.00%)(χ2=93.82, P < 0.01). The positive expression of Piwil2 protein in malignant lobar tumor tissue was related to the tumor diameter≥10 cm, Ki-67≥25%, oncocyte atypia, CyclinD1 protein positive expression and Karyokinesis≥15/10HPF(P < 0.05 to P < 0.01), but not related to the age and postoperative recurrence(P>0.05). The ten-year survival rate of malignant lobar tumor patients with positive expression of Piwil2 protein(65.12%) was lower than that of patients with negative expression of Piwil2 protein(100.00%), and the difference of which was statistically significant(χ2=3.95, P < 0.05). Conclusions The abnormal expression of Piwil2 protein is involved in the evolution and development of malignant breast lobar tumor, and can also be used as a reference index for predicting the survival and prognosis of patients.

Abstract:
Objective To compare the application effects between the platelet-rich fibrin(PRF) combined with coronally advanced flap(CAF) and coronally advanced flap alone in the treatment of gingival recession(GR). Methods Thirty-six patients with GR were randomly divided into the observation group(18 cases, 25 teeth with GR) and control group(18 cases, 24 teeth with GR). The observation group and control group were treated with PRF combined with CAF and CAF, respectively. The microsurgery in two groups were used.The base line, and periodontal probing depth(PD), vertical gingival regression height(VGR), keratinized gingival width(KTH), gingival thickness(GT), patient comfort(PCS), sensitivity(HS), aesthetic index(PES) and total satisfaction after 3 and 6 months of operation in two groups were recorded and compared. Results The differences of the PD and KTH in two groups between before and after operation were not statistically significant(P>0.05). In terms of root surface coverage, the difference of the VGR in two groups between before and after operation was statistically significant(P < 0.01). The difference of the GT in the observation group between before and after operation was statistically significant(P < 0.01). The differences of the patients' satisfaction(including the PCS, HS and PES) in the observation group were statistically significant between before and after surgery(P < 0.01), especially for HS, the patients were generally satisfied after surgery. Conclusions The PRF membrane combined with CAF in the treatment of GR can achieve the good root surface coverage, and the postoperative GT and patient's satisfaction have more prominent advantages compared with the CAF alone.
Efficacy of oxiracetam combined with Xingnaojing in the treatment of traumatic brain injury and its effect on neuronal injury

Abstract:
Objective To comprehensively analyze the clinical effects of oxiracetam combined with Xingnaojing in the treatment of traumatic brain injury, and its effects on neuronal injury. Methods Seventy-six patients with traumatic brain injury were randomly divided into the observation group and control group(38 cases in each group). On the basis of comprehensive treatment, the control group was treated with oxiracetam, and the observation group was treated with oxiracetam combined with Xingnaojing. The Glasgow coma score(GCS), acute physiological and chronic health status scores(APACHE Ⅱ), degree of neuron injury[serum levels of glial fibrillary acidic protein(GFAP), neuron specific enolase(NSE), calcium binding protein S100B and myelin basic protein(MBP)], nerve injury repair indexes[levels of heat shock protein(HSP)-90, HSP-70, HSP-27 and nerve growth factor(NGF)], inflammatory chemokines[serum monocyte chemokine 1(MCP-1), aspartate cysteine-specific proteinase-3(Caspase-3) and factors regulating the expression and secretion of activated normal T cells(RANTES)] in two groups were compared between before treatment and after 14 and 28 days of treatment. The prognosis and adverse reactions between two groups were compared. Results Compared with before treatment, the GCS scores in two groups increased, the APACHE Ⅱ scores decreased in two groups after 14 and 28 days of treatment, and which in observation group was better than that in control group(P < 0.01). The results of evaluation after 28 days of treatment showed that there was no statistical significance in the rate of good prognosis between two groups(P>0.05). After 14 and 28 days of treatment, the serum levels of GFAP, NSE, S100B, MBP, HSP-90, HSP-70, HSP-27 and NGF in two groups decreased compared with before treatment, and the decrease amplitude in observation groups was higher than that in control group(P < 0.05 to P < 0.01). The serum levels of MCP-1, Caspase-3 and RANTES in two groups after 14 and 28 days of treatment were lower than that before treatment, and which in observation group was lower than that in control group(P < 0.05 to P < 0.01). There was no statistical significance in the incidence rate of adverse reactions between two groups(P>0.05). Conclusions Oxiracetam combined with Xingnaojing has significant clinical effect in the treatment of traumatic brain injury, which can effectively relieve the nerve injury, reduce the serum content of MCP-1, Caspase-3 and RANTES, and alleviate local inflammation. The recovery and prognosis of patients are good.
Effectiveness analysis of EV-A71 vaccination against hand-foot-mouth disease in children

Abstract:
Objective To analyze the effectiveness of EV-A71 vaccination in preventing hand-foot-mouth disease(HFMD) and EV-A71 virus infection in children. Methods Three hundred children with HFMD at 0.5-6.0 years old were divided into the the vaccination group(122 cases) and unvaccination group(178 cases). The nucleic acid of EV-A71, CV-A16 and CV-A6 enterovirus were detected using the throat swabs and/or feces of children. The correlation between EV-A71 vaccine and various types of virus infection of HAND were analyzed. Results The EV-A71 infection rate in vaccination group was significantly lower than that in unvaccination group(P < 0.01). The differences of the CV-A16, CV-A6 and other infection between two groups were not statistically significant(P>0.05). The EV-A71 infection rate in children vaccinated with one dose of EV-A71 vaccine was higher than that in children vaccinated with two doses(P < 0.05). There was no statistical significance in the CV-A16, CV-A6 and other infection rates between children vaccinated with one dose of EV-A71 vaccine and those vaccinated with two doses(P>0.05). There was no statistical significance in the severity of EV-A71 virus infection between patients with vaccination and patients without vaccination(P>0.05). Conclusions Full-dose prophylactic EV-A71 vaccination is a protective factor for HFMD caused by EV-A71 virus infection, but not for HFMD caused by other virus infections.
Clinical effect analysis of double titanium plate with loop Tight Rope fixation under endoscopy in the treatment of acromioclavicular joint dislocation

Abstract:
Objective To study the effects of double titanium plate with loop(Tight Rope) fixation under endoscopy in the treatment of acromioclavicular joint dislocation. Methods Forty acromioclavicular joint dislocation patients included 27 cases of Rockwood type Ⅲ and 13 cases of type Ⅳ. Twenty cases treated with Tight Rope internal fixation under arthroscopy and 20 cases treated with open reduction and clavicular hook plate internal fixation were divided into the observation group and control group, respectively. Two groups were followed up for more than 24 months. The surgical indicators and complications in two groups were recorded. The comprehensive efficacy in two groups before operation, after 1 and 24 months of operation were evaluated using the shoulder joint system(UCLA), pain visual analogue(VAS) score and Constant-Murley shoulder score. Results The average blood loss and hospital stay in observation group were significantly lower than that in control group(P < 0.01), and the difference of the operation time between two groups was not statistically significant(P>0.05). The pain VAS scores in two groups after 1 and 24 months of surgery were lower than that before surgery, and the UCLA and Constant-Murley scores were higher than those before surgery(P < 0.05 to P < 0.01). The differences of the UCLA and Constant-Murley scores between two groups before operation were not statistically significant(P>0.05). The UCLA score and Constant-Murley score after 1 month of operation, and UCLA score after 24 months of operation in observation group were higher than those in control group(P < 0.01). The differences of the excellent and good rate of operation and in cidence rate of complications between two groups after 24 months of operation were not statistically significant(P>0.05). Conclusions The double titanium plate with loop fixation under endoscopy in the treatment of acromioclavicular joint dislocation can firmly fix the dislocated acromioclavicular joint, reduce pain symptoms and improve joint function, and also has the advantages of small trauma and low complication rate.
Efficacy and safety of gabapentin in the treatment of pruritus in peritoneal dialysis patients

Abstract:
Objective To observe the efficacy and safety of gabapentin in the treatment of pruritus in peritoneal dialysis patients. Methods A randomized controlled trial was performed. Peritoneal dialysis patients with pruritus were selected and randomly divided into experimental group and control group. Twenty-three patients in the control group received loratadine 10 mg orally before sleep every night, while 25 patients in the experimental group received gabapentin 100 mg orally before sleep every day. After 4 weeks, efficacy was observed according to visual analog score(VAS) and 5-D pruritus scale score, and adverse reactions were recorded. Laboratory data such as blood urea nitrogen, blood creatinine, hemoglobin, Kt/V, blood calcium, blood phosphorus and parathyroid hormone of patients before and after treatment were collected. Results After treatment, VAS score and 5-D pruritoneal scale score in the experimental group decreased compared with the control group, and the differences were statistically significant(P < 0.05). Four cases in the control group showed drowsiness and 6 cases in the experimental group showed fatigue and drowsiness, all of which were relieved without any treatment, and none of them developed serious complications that led to discontinuation of treatment. There was no significant difference in the incidence of adverse reactions among patients with 24 h urine volume >400 mL, 100-400 mL and < 100 mL in the observation group(P>0.05). Conclusions Gabapentin is effective in improving pruritus in patients with peritoneal dialysis complicated with pruritus and small-dose daily administration is a safe way for using.

Abstract:
Objective To analyze the correlation between hypertensive disorder complicating pregnancy and different in vitro fertilization techniques, and its pregnancy outcomes. Methods The data of 546 infertility patients with in vitro fertilization were analyzed retrospectively. According to the method of fertilization, the patients were divided into the group A(313 cases with conventional in vitro fertilization-embryo transfer) and group B(233 cases with intracytoplasmic sperm injection). According to the embryo transfer cycle, the patients were divided into the group C(370 cases with embryo transfer in fresh cycle) and group D(176 cases with embryo transfer in frozen cycle). The incidence rates of hypertensive diseases during pregnancy were compared among four groups. Results Among 546 cases, 24 cases with gestational hypertension, 28 cases with preeclampsia and no eclampsia case were identified.The incidence rate of gestational hypertension in group A(2.66%) was lower than that in B group(6.87%)(P < 0.05), and there was no statistical significance in the incidence of preeclampsia between two groups(P>0.05). There was no statistical significance in the incidence rates of gestational hypertension and preeclampsia between group C and group D(P>0.05). The differences of the total incidence rates of adverse pregnancy outcomes between group A and group B, and between group C and group D were not statistically significant(P>0.05). Conclusions In infertile women, the intracytoplasmic sperm injection increases the risk of gestational hypertension, regardless of conventional in vitro fertilization-embryo transfer or freshness of embryo transfer.
Correlation between sodium iodine transporter expression and BRAFV600E mutation in thyroid papillary carcinoma

Abstract:
Objective To explore the relationship between BRAFV600E mutation and sodium iodide transporter(NIS) in common genetic events in thyroid papillary carcinoma(PTC). Methods A total of 104 PTC patients who underwent surgical treatment PTC were selected. Fresh PTC tissue and normal tissue adjacent to carcinoma specimens were collected. The contents of NIS mRNA were detected by fluorescence quantitative PCR. NIS protein expressions in paraffin blocks were detected by immunohistochemistry, at the same time BRAFV600E gene mutation was tested. Results In 104 cases of PTC, there were 70 cases of BRAFV600E gene mutation(67.3%), which was significantly higher than that in the normal tissue adjacent to carcinoma(0 case)(P < 0.01). The expression of NIS mRNA in PTC tissues was significantly lower than that in the normal tissues adjacent to carcimoma(P < 0.01), but the expression of NIS protein was significantly higher than that in the normal tissues adjacent to carcimoma(P < 0.01). The proportion of NIS mRNA expression in the PTC tissues was lower and the increase of NIS protein expression was significantly higher than that in the normal tissues adjacent to carcimoma(P < 0.01). The expression of NIS mRNA in patients with gene mutation of PTC BRAFV600E was significantly lower than that in the wild type(P < 0.01), but there was no significant difference in NIS protein(P>0.05). Conclusions BRAFV600E mutation only exists in the cancer tissue, which may be associated with the low expression of NIS mRNA in cancer tissue and high expression of the NIS protein. NIS protein in PTC may be mainly expressed in the cytoplasm, which affects the normal function of transportation and causes patients to be unable to gather iodine.
Effect of protein-energy wasting on quality of life in maintenance hemodialysis patients with end-stage renal disease

Abstract:
ObjectiveTo study the effect of protein-energy wasting(PEW) on the prognosis of maintenance hemodialysis(MHD) patients.MethodsA total of 150 patients with end-stage renal disease who were subject to MHD, were enrolled in the invstigation and divided into PEW group(n=33) and non-PEW group(n=117) according to the diagnosis critiria of PEW syndorome. The quality of life scores by KDQOLTM-36 scale in the two groups were compared and analyzed.ResultsThirty-three cases met at least three diagnostic indexes of PEW with a percentage of 22.0%. The values of systolic blood pressure, diastolic blood pressure, heart rate, upper arm circumference, triceps skinfold thickness, hip circumference, grip strength, albumin and creatinine in PEW group were significantly lower than those in non-PEW group(P < 0.05);the time of dialysis in the PEW group was longer than that in the non-PEW group, and body mass index was lower than that in the non-PEW group(P < 0.05). There were significant differences in vitality, social function, social emotion, symptom influence, nephropathy influence and total score between PEW group and non PEW group(P < 0.05 to P < 0.01).ConclusionsPEW is a common complication in end-stage renal disease patients receiving MHD and its impact on patients' quality of life should be given more attention.
Application value of high frequency color Doppler ultrasound in carotid artery reconstruction of patients with ocular ischemic syndrome

Abstract:
ObjectiveTo study the value of high frequency color Doppler ultrasound in carotid artery reconstruction in patients with ocular ischemic syndrome(OIS).MethodsA total of 50 patients with OIS who underwent carotid artery reconstruction from July 2018 to July 2019 were selected and divided into forward flow group(28 cases) and reverse flow group(22 cases) according to the direction of ophthalmic artery blood flow. The mean carotid stenosis, best corrected visual acuity and hemodynamic changes of ophthalmic artery, central retinal artery and posterior ciliary artery were compared between the two groups before and after operation.ResultsBefore and after treatment, the average degree of carotid stenosis in the reverse flow group was higher than that in the forward flow group(P < 0.01). After treatment, the average degree of carotid stenosis in the two groups was lower than that before treatment(P < 0.01). Before treatment, the PSV of ophthalmic artery in the reverse flow group was higher than that in the forward flow group, and the PSV of posterior ciliary artery was lower than that in the forward flow group(P < 0.05 and P < 0.01). After treatment, the PSV of ophthalmic artery, posterior ciliary artery and central retinal artery in the two groups were higher than that before treatment(P < 0.01), and the PSV of ophthalmic artery and posterior ciliary artery in the forward flow group was higher than that in the reverse flow group(P < 0.05 and P < 0.01). Before and after treatment, the best corrected visual acuity in the forward flow group was higher than that in the reverse flow group(P < 0.01). After treatment, the best corrected visual acuity in the forward flow group was higher than that before treatment(P < 0.01).ConclusionsAfter carotid revascularization, the mean carotid stenosis and hemodynamics of ophthalmic artery, central retinal artery and posterior ciliary artery in OIS patients are significantly improved, and the visual acuity in the forward flow group is significantly improved, but not in the reverse flow group. High frequency color Doppler ultrasound can accurately monitor the changes of hemodynamics of main postbulbar blood vessels before and after operation, which is helpful to grasp the improvement of patients' condition in real time.
Study on the feasibility of quantitative evaluation of osteoporosis by dual-energy CT virtual non-contrast scan

Abstract:
ObjectiveTo investigate the feasibility of dual-energy CT(DECT) virtual non-contrast scan in the quantitative evaluation of bone mineral density(BMD) measured by QCT as the gold standard in the absence of a body mode.MethodsA retrospective study was performed on 100 patients with abdominal true non-contrast scan(TNC) and GSI three-phases enhanced scan, and the patients were randomly divided into the training set and validation set(50 cases in each set). The BMD value of L1 vertebral body in two sets was measured using QCT Pro software. In the training set, the difference of L1 vertebral CT value between conventional non-contrast scan image and GSI enhanced virtual non-contrast scan image in portal vein phase was compared, and the correlation between virtual non-contrast scan vertebral CT value and BMD value was determined. The regression model of virtual non-contrast scan vertebral CT value to predict BMD value was established through multiple regression analysis. The validation set was used to evaluate the accuracy of this equation in the diagnosis of osteoporosis and effectiveness of virtual non-contrast scan vertebral CT value in the diagnosis of osteoporosis.ResultsThe CT value of L1 vertebral body of VNC was lower than that of TNC(P < 0.01). After subtracting the TNC, the total radiation dose of image was reduced by 21.94%, the multiple linear regression model of CT value of VNC to predict BMD of L1 vertebra was as follows: BMD=102.375+1.349×CT value-0.321×age-0.433×height(R2=0.979). The ROC curve analysis showed that the cut-off value of virtual non-contrast scan vertebral body CT value based on portal vein phase in the diagnosis of osteoporosis and osteopenia were 54.35 HU and 64.51 HU, respectively.ConclusionsIn the case of independent body model, the DECT virtual non-contrast scan vertebral CT value can reliably measure the BMD, and the established predictive BMD model can quantitatively diagnose the osteoporosis and osteopenia, which is a beneficial supplement to QCT technology.
Value of three-dimensional speckle tracking imaging in the diagnosis of cardiac involvement in patients with AL amyloidosis

Abstract:
ObjectiveTo analyze the diagnostic value of the three-dimensional speckle tracking imaging in cardiac involvement in patients with systemic immunoglobulin light chain(AL) amyloidosis.MethodsEighty AL amyloidosis patients with normal left ventricular ejection fraction(LVEF) were investigated.According to whether the heart was involved, the patients were divided into the non-heart-affected group(group A, 35 cases) and heart-affected group(group B, 45 cases). The clinical data, serological parameters, conventional echocardiographic parameters and three-dimensional speckle tracking imaging parameters were compared between two groups, and the receiver operating characteristic curve(ROC) was used to analyze the specificity and sensitivity of three-dimensional speckle tracking imaging parameters in the diagnosis of AL amyloidosis.ResultsThe level of amino-terminal B-type natriuretic peptide precursor(NT-proBNP) in group B was significantly higher than that in group A(P < 0.01). There was no statistical significance in the conventional ultrasound parameters between two groups(P>0.05). The overall left ventricular longitudinal strain(LVGLS), overall left ventricular area strain(LVGAS) and overall left ventricular radial strain(LVGRS) in group B were significantly lower than those in group A(P < 0.05 to P < 0.01). The results of ROC curve analysis showed that when the 16.12% of LVGLS was used as the cut-off point for diagnosing AL myocardial amyloidosis, the area under curve, sensitivity, specificity and Yuden index were the highest, which were 0.798, 94.18%, 86.84% and 0.619, respectively. The intra-group correlation coefficients of LVGLS, LVGAS and LVGRS were 0.908, 0.837 and 0.839, respectively.ConclusionsThe ultrasound three-dimensional speckle tracking imaging has certain diagnostic value in cardiac involvement in patients with AL amyloidosis, is simple, non-invasive and worthy of recommending.
Analysis of red blood cell parameters for patients with CKD at stage 3-5

Abstract:
ObjectiveTo investigate the characteristics and clinical significance of red blood cell parameters in patients with chronic kidney disease(CKD) at stages 3-5 with the data from Guangdong Provincial Hospital of Chinese Medicine.MethodsOne hundred and twenty cases with CKD in each stage were collected. The red blood cell parameters including red blood cell count(RBC), hematocrit(HCT), hemoglobin(Hb), mean corpuscular volume(MCV), mean corpsular hemoglobin(MCH), mean corpuscular hemoglobin concentration(MCHC) and red cell distribution width(RDW) were obtained from sysmexXE-5000 automatic hematology analyzer.ResultsThere was no significant difference in the age of patients with different genders or different courses of the disease(P>0.05). Regardless of gender, the age of patients with different course of disease was statistically significant(P < 0.05). The MCHC of male patients in all stages of CKD was higher than that of female patients(P < 0.05 to P < 0.01. The RBC, Hb and HCT of male patients were higher than those of female patients in CKD stage 3 and 4(P < 0.01), but there was no statistically significance in stage 5(P>0.05). RDW had no statistical difference between stage 3 and 4(P>0.05), but it was higher that in female patients than infe male patients in stages 5(P < 0.05). There was no significant gender difference in MCV and MCH in each stage of CKD(P>0.05). Hb and HCT decreased with the disease progression(P < 0.01), while RDW was just the opposite(P < 0.01). MCV and MCH decreased in female patients as clinical stages increased(P < 0.01), but they had no significant difference in male patients(P>0.05). There was no significant difference in the MCHC between male and female patients(P>0.05).ConclusionsThe changes of RBC, Hb and HCT are closely related to the development of CKD. The ranges of red blood cell parameters established based on their characteristics can provide the reference for clinicians to evaluate the condition of the patients with CKD.
Relationship between serum tumor markers and EGFR gene mutation in NSCLC based on logistic regression and ROC curve analysis

ObjectiveTo evaluate the correlation between serum tumor markers and epidermal growth factor receptor(EGFR) gene mutation in non-small cell lung(NSCLC) cancer by logistic regression and receiver operating characteristic(ROC) curve.MethodsA total of 128 hospitalized patients with lung adenocarcinoma were collected and 5 mL of venous blood was collected at the initial diagnosis, 3 mL serum was separated, and serum tumor markers were detected. According to the results of EGFR gene detection, they were divided into EGFR gene mutation group and EGFR gene non-mutation group. The general clinical characteristics of two groups of patients with NSCLC were compared. The ROC curve was drawn based on logistic regression model, and the area under the curve(AUC) was calculated to evaluate the value of each index and the combined diagnosis.ResultsEGFR mutation was associated with gender, smoking history, SCC and CA125(P < 0.05 to P < 0.01). The mutation frequency of EGFR in female patients with NSCLC was higher than that in male(P < 0.01). The mutation frequency of EGFR in patients with smoking history was lower than that in patients without smoking history(P < 0.01). The mutation frequency of EGFR in patients with negative serum SCC was higher than that in patients with positive serum SCC(P < 0.01). The mutation frequency of EGFR in patients with negative serum CA125 was higher than that in patients with positive CA125(P < 0.05). Among the serum tumor markers, gender, CA125, SCC and smoking history had higher diagnostic value with EGFR gene mutation(P < 0.05). At the same time, the combined diagnosis of these indicators, such as gender+SCC, gender+CA125+SCC, gender+SCC+smoking history, CA125+SCC+smoking history, gender+CA125+SCC+smoking history, which all had AUC value more than 0.75(P < 0.05), had high diagnostic value.ConclusionsThere is a good correlation between SCC and CA125 in serum tumor markers and EGFR mutation in NSCLC.The combination of SCC and CA125 with other clinical data(gender, smoking history) can enhance the predictive discrimination of EGFR, which is worthy of further clinical application.
Expression and prognostic value of key genes in bladder cancer based on TCGA database

Abstract:
ObjectiveTo explore the differential expressions and the prognostic role of genes in bladder cancer.MethodsThe gene expression data of bladder cancer were downloaded from TCGA database. The differentially expressed genes between tumor and normal tissues were screened by R software, and their potential biological functions and signal pathways were evaluated by enrichment analysis. A protein-coding interaction network was constructed using the online database STRING11.0 to further screen key genes. The expressions of key genes in bladder cancer was verified by GEPIA and UALCAN database. Then the online tool Kaplan Meier Plotter was used to analyze the prognosis.ResultsA total of 1 538 genes were differentially expressed, including 1 088 up-regulated and 450 down-regulated. Enrichment analysis showed that the differential genes were mainly related to signal transduction, substance metabolism, immune responses and other signal pathways. Among the 10 key genes screened, 2 genes have the prognostic value.ConclusionsThere are many differentially expressed genes in bladder cancer which have an impact on the progression of the disease. It is of great significance to find and use them in the individualized treatment of bladder cancer.
Construction and analysis of the prediction model of psychological disorder in colorectal cancer patients with enterostomy

Abstract:
ObjectiveTo explore the construction of a prediction model of psychological disorders in colorectal cancer patients with enterostomy.MethodsA total of 290 colorectal cancer patients with enterostomy were investigated, and randomly set the training set(203 cases) and test set(87 cases). According to the Minnesota multiphasic personality inventory(MMPI), the patients were divided into the psychological disorder group(MMPI≥60 points) and non-psychological disorder group(MMPI < 60 points). The general data in the training set were compared between two groups, the related factors affecting the psychological disorders of colorectal cancer patients with enterostomy were analyzed using the logistic regression model. The R software was used to establish a nomogram prediction model based on the training set, and the effectiveness of model in the test set was evaluated.ResultsTwo invalid questionnaires(one in the training set and one in the test set) were eliminated, and 288 valid questionnaires were obtained with effective recovery rate of 99.31%. Two hundred and two cases were included in the training set, and 67 cases(33.17%) in the psychological disorder group and 135 cases(66.83%) in the non-psychological disorder group were identified. The differences of the gender, enterostomy time, self-management ability score, social support rating scale(SSRS) score and enterostomy complications between two groups were statistically significant(P < 0.05 to P < 0.01). The results of multivariate logistic analysis showed that female, long time of enterostomy, high score of self-management ability and high score of SSRS were the protective factors of psychological disorders, and the enterostomy complication was the risk factor of psychological disorders in colorectal cancer patients with enterostomy(P < 0.05 to P < 0.01). The consistency index of the above factors in predicting psychological disorders in colorectal cancer patients with enterostomy was 0.804(95%CI: 0.743-0.857). The results of further verification by ROC curve showed that the specificity was 88.50%, and the sensitivity was 81.60%. The results of external validation showed that the sensitivity and specificity of this model were 85.71% and 87.93%, respectively.ConclusionsThe psychological disorder of colorectal cancer patients with enterostomy is more severe. The gender, self-management ability, social support, enterostomy complications and time are the influencing factors of psychological disorder. The prediction model has high application value.
Study on the self-perceived burden of patients after enterostomy and its influencing factors

Abstract:
ObjectiveTo investigate the status quo of self-perceived burden and self-management behavior among enterostomy patients, and explore its relevant influencing factors.MethodsA total of 170 patients with enterostomy from stoma outpatient department of three tertiary grade A general hospitals in Shanghai were investigated using the general data questionnaire, self-perceived burden scale and self-management behavior questionnaire, and the results of survey were analyzed.ResultsThe total score of self-perceived burden(27.29±0.66) points was at a mild level. Among the three dimensions, the highest score was the physical burden(2.88±0.06) points, followed by economic burden(2.12±0.10) points, and the lowest score was emotional burden(1.92±0.06) points. The total score of self-management behavior(113.97±13.90) points was at a high level.The scores of the psychology, symptoms, information, ostomy nursing and daily life of five dimensions were(4.27±0.04), (4.11±0.05), (3.85±0.05), (3.72±0.06) and(3.27±0.07) points, respectively. The total score of self-perceived burden was significantly negatively correlated with the total scores of self-management behavior, symptom management and psychological management(P < 0.01). The self-management behavior, marital status and caregiver health status were the main influencing factors of self-perceived burden among enterostomy patients. The level of self-management behavior had no mediating effects on the impact of marital status and caregiver health status on self-perceived burden.ConclusionsMedical staff should pay attention to maintaining the mental health of patients with enterostomy, and improve the quality of life of patients by improving self-management behavior and reducing the self-perceived burden.
Analysis of the influencing factors of stigma in colorectal cancer patients after enterostomy

Abstract:
ObjectiveTo investigate the status of stigma in colorectal cancer patients treated with enterostomy, and analyze its main influencing factors.MethodsA total of 290 patients with colorectal cancer enterostomy from the outpatient department of stomostomy and gastrointestinal surgery department of The First Affiliated Hospital of Bengbu Medical College from October 2020 to March 2021 were selected by convenience sampling method. The baseline data(including the general demographic data, anxiety anddepression scale, social impact scale and perceived social support scale) were collected using the questionnaires. According to the score of each scale, the level of patients' stigma was evaluated, and the main influencing factors were analyzed.ResultsThe total score of stigma in colorectal cancer patients with enterostomy was(68.16±13.51) points, which was at a moderate level, and 43.1% patients had severe stigma. The results of multiple linear regression analysis showed that the degree of self-care, stoma acceptance, presence of stoma complications, depression and anxiety, and social support were the factors of influencing the sense of stigma(P < 0.05 to P < 0.01). The social support was negatively correlated with the sense of stigma, while the level of anxiety and depression was positively correlated with the sense of stigma(P < 0.01).ConclusionsThe main factors influencing the sense of stigma in colorectal cancer enterostomy patients include the degree of stoma self-care, acceptance, presence of complications, levels of social support, and anxiety and depression.
Model analysis of the degree of association between post-traumatic growth, locus of control and psychological acceptance of enterostomy patients

Abstract:
ObjectiveTo analyze the relationship between post-traumatic growth, locus of control and psychological acceptance of patients with enterostomy.MethodsEighty-eight patients with rectal cancer enterostomy were selected.The ostomy adjustment inventory-20(OAI-20) was used to evaluate the psychological acceptance, the posttraumatic growth inventory was applied to assess the post-traumatic growth, and the multidimentional health locus of control scale was employed to assess the locus of control. A structural equation model established by AMOS24.0 software was used to verify the mediating effect of locus of control between psychological acceptance and post-traumatic growth.ResultsThe OAI-20 score of 88 patients was(43.66±6.26) points, which was at a medium-low adaptation level; the average score of each dimension item was as follows: positive emotion dimension(2.28±0.38) points, negative emotion dimension(2.42±0.52) points, and social life dimension(2.21±0.46) points. The total score of post-traumatic growth was(68.33±10.46) points, and 54 patients(61.36%) had post-traumatic growth.The score of health internal control was(21.33±4.37) points, the score of health authority control was(24.26±4.68) points, and the score of health opportunity control was(20.35±4.26) points. The locus of control had a mediating effect between psychological acceptance and post-traumatic growth, and the mediating effect accounted for 61.60% of the total effect.ConclusionsThe influence of post-traumatic growth on psychological acceptance of rectal cancer patients should be paid attention to, and effective measures should be taken to improve the post-traumatic growth and locus of control of patients, so as to improve the psychological acceptance of stoma.
Design and application of an adjustable plastic tray cutter for ostomy bag

Abstract:
ObjectiveTo design an adjustable plastic tray cutter for ostomy bag and discuss its application effect.MethodsA total of 84 patients with ostomy surgery from March 2020 to March 2021 were selected and divided into observation group and control group, 42 cases in each group. The control group used special scissors for stoma to cut the plastic disc of the ostomy bag, the observation group used an adjustable ostomy plastic tray cutter to cut the ostomy plastic tray. The time of patients independently completing the tailored ostomy plastic disc, difficulty score of tailoring plastic tray for ostomy bag, number of cases of proper, inappropriate and scrapped plastic tray for ostomy bag, and leakage of the plastic disc in the ostomy bag on the eighth day after surgery were compared.ResultsThe time of patients independently completing the cutting of pocket plastic disc in the observation group was less than that in the control group(P < 0.01). The cutting of colostomy plastic disc in the observation group was better than that in the control group(P < 0.05), the difficulty of plastic disc cutting was lower than that of the control group(P < 0.05), and the leakage rate of plastic disc was lower than that of the control group on the eighth day after operation(P < 0.05).ConclusionsThe adjustable plastic tray cutter for ostomy bag can realize rapid and accurate cutting of pocket plastic disc according to different stoma shapes, with low scrap rate and convenience for patients.
