Effect of quercetin on hydrogen peroxide-induced AKT/mTOR signaling pathway and autophagy in human umbilical vein endothelial cells

Abstract:
ObjectiveTo analyze the effect of quercetin on autophagy and proliferation of human umbilical vein endothelial cells and its relationship with AKT/mTOR signaling pathway.MethodsMTT method was used to detect cell viability.MDC staining was used to detect the level of autophagy vesicles.Western blotting was used to detect the levels of autophagy-related proteins, and 3-MA was used to study the effect of quercetin on autophagy and cell proliferation.ResultsQuercetin increased the viability of human umbilical vein endothelial cells and up-regulated the fluorescent particles labeled by MDC staining in the cells followed with the increase in concentration, which promoted the expressions of autophagy protein LC3 Ⅱ/LC3 Ⅰ in umbilical vein endothelial cells and reduce the levels of p-AKT/AKT and p-mTOR/mTOR(P < 0.01).Compared with the quercetin group, the cell viability and LC3 Ⅱ/LC3 Ⅰ of the quercetin+3-MA group were significantly reduced, and p-mTOR/mTOR was significantly increased(P < 0.05).ConclusionsQuercetin can regulate the proliferation of umbilical vein endothelial cells by inhibiting the AKT/mTOR signaling pathway and inducing autophagy.
Study on the correlation between inflammatory factors and myeloperoxidase in the treatment of ulcerative colitis with mesalasize combined with dandelion polysaccharide

Abstract:
ObjectiveTo observe the correlation between the levels of inflammatory factors interleukin (IL) -6, IL-4, IL-8, IL-10, tumor necrosis factor-α (TNF-α) and myeloperoxidase(MPO) in the ulcerative colitis(UC) model rats treated with dandelion polysaccharide.MethodsThe UC model of rats was established by 2, 4, 6-trinitrobenzenesulfonic acid combined with 50% ethanol solution.The refined dandelion polysaccharide was extracted by water extraction and alcohol precipitation method.Fifty male rats were randomly divided into 5 groups: normal control group, model control group, mesalazine group, dandelion polysaccharide group and mesalazine combined with dandelion polysaccharide treatment group(combined treatment group), with 10 rats in each group.The levels of serum inflammatory factors(IL-6, IL-4, IL-8, IL-10, TNF-α) and the expression of MPO in colon tissue were detected by enzyme-linked immunosorbent assay.ResultsCompared with the normal control group, the serum levels of IL-6, IL-8 and TNF-α and MPO content in colon tissues were significantly increased, and the levels of IL-4 and IL-10 were decreased in the model control group and dandelion polysaccharide group (P < 0.05), meanwhile, the serum levels of IL-6 and TNF-α were increased and the levels of IL-4 and IL-10 were significantly decreased in the mesalazine group (P < 0.05), and there was no significant difference in the levels of each factor in the combined treatment group (P>0.05).Compared with the model control group, the mesalazine group, dandelion polysaccharide group and combination treatment group had significant reductions in the serum levels of IL-6, IL-8, TNF-α and MPO content in colon tissues, and significant increases in the levels of IL-4 and IL-10 (P < 0.05).Compared with the mesalazine group and the dandelion polysaccharide group, the combined treatment group had significant reductions in the serum levels of IL-6, IL-8, TNF-α and the MPO content in colon tissues, and significant increases in the levels of IL-4 and IL-10 (P < 0.05).The expression levels of IL-6, IL-8 and TNF-α were positively correlated with MPO level, and the levels of IL-4 and IL-10 were negatively correlated with the levels of MPO(P < 0.05).ConclusionsThe effect of mesalazine combined with dandelion polysaccharide in the treatment of UC is better than that of mesalazine and dandelion polysaccharide alone.The levels of IL-6, IL-8 and TNF-α are positively correlated with the levels of MPO and the progression of inflammation, while the levels of IL-4 and IL-10 are negatively correlated with the levels of MPO and the progression of inflammation.
Effect of sesamin on high glucose-induced vascular endothelial cell injury through lncRNA WEE2-AS1/miR-515-5p pathway

Abstract:
ObjectiveTo explore the effect of sesamin on human umbilical vein endothelial cells (HUVEC) injury induced by high glucose and its possible mechanism.MethodsHigh glucose stimulated the HUVEC to establish the cell damage model, and the cells were treated with different concentrations of sesamin.qRT-PCR was used to detect the expressions of LncRNA WEE2-AS1 and miR-515-5p.After transfection of sh-NC and sh-WEE2-AS1 into HUVEC, the cells were treated with 30 mmol/L glucose(HG+sh-NC group, HG+sh-WEE2-AS1 group).HUVEC cells stably overexpressing WEE2-AS1 were constructed.HG+WEE2-AS1-LV group was treated with 30 mmol/L glucose.HG+SES+WEE2 -AS1-LV group was treated with 40 μmol/L sesamin and 30 mmol/L glucose.MTT and flow cytometry were used to detect cell proliferation and apoptosis rates.The levels of SOD, LDH, and MDA were tested according to the kit instructions.The dual luciferase reporter gene experiment was used to detect the targeting relationship between WEE2-AS1 and miR-515-5p.Western blotting was used to detect the expression of cleaved-caspase3 protein.ResultsSesamin reduced the expression of WEE2-AS1, the rate of apoptosis, the protein level of cleaved-caspase3, the activity of LDH and the level of MDA in HUVEC after stimulated with high glucose (P < 0.05).It could promote the expression of miR-515-5p and enhance the cell viability and the activity of SOD (P < 0.05) in a dose-dependent manner.Compared with the HG+sh-NC group, the expression of miR-515-5p in the HG+sh-WEE2-AS1 group was increased (P < 0.05), the cell viability and the activity of SOD were increased (P < 0.05), and apoptosis rate and the protein level of cleaved-caspase3 were decreased (P < 0.05), and the activity of LDH and the level of MDA were decreased (P < 0.05).WEE2-AS1 could targetedly regulate miR-515-5p.Compared with the HG+SES group, the expression of miR-515-5p in the HG+SES+WEE2-AS1-LV group was decreased (P < 0.05), the cell viability and the activity of SOD were decreased (P < 0.05), the apoptosis rate and the protein level of cleaved-caspase3 were increased (P < 0.05), the activity of LDH and the level of MDA were increased (P < 0.05).ConclusionsSesamin can promote cell proliferation and inhibit apoptosis and oxidative stress by regulating WEE2-AS1/miR-515-5p, thereby reducing the damage of vascular endothelial induced by high glucose.
Serum levels of adiponectin, resistin and chemerin in patients with nodular goiter and their correlations

Abstract:
ObjectiveTo investigate the serum levels of adiponectin, resistin and chemerin in patients with nodular goiter and their correlations.MethodsOne hundred and fifty patients with nodular goiter initially diagnosed in physical examination from 2017 to 2018 and 150 normal people matched with gender and age were selected.Blood samples were collected from subjects in the two groups, general clinical data were collected, and general biochemical indexes and serum levels of adiponectin, resistin and chemerin were recorded and analyzed.Body mass index(BMI) and insulin resistance index(HOMA-IR) were further measured and calculated.Correlation between the serum level of adiponectin, resistin or chemerin and the pathogenesis of nodular goiter was finally analyzed.ResultsCompared to subjects in the control group, patients with nodular goiter showed higher levels of triglyceride(TG), low density lipoprotein(LDL-C), fasting blood glucose(FBG), HOMA-IR, adiponectin and chemerin(P < 0.01), but lower level of resistin(P < 0.01).For patients with nodular goiter, the level of adiponectin was found to be negatively correlated with fasting insulin(FINS) and HOMA-IR(P < 0.01);the level of resistin was found to be negatively correlated with TG, FBG, FINS and HOMA-IR(P < 0.05 to P < 0.01);and the level of chemerin was found to be positively correlated with TG, FBG, FINS and HOMA-IR(P < 0.01).Quartile analysis showed that the proportion of cases in either the observation group or the control group increased with the increase of the levels of adiponectin and chemerin, and the level of resistin was lower in cases of the highest quartile than those of the lowest quartile in each group.In patients with nodular goiter, compared to the normal thyroid function subgroup, hyperthyroidism subgroup exhibited an increase in the level of free triiodothyronine(FT3), free thyroxine(FT4), adiponectin and chemerin, but a decrease in the level of thyroid stimulating hormone(TSH) and resistin(P < 0.01).In the meantime, compared to the normal thyroid function subgroup, hypothyroidism subgroup showed higher levels of TSH and chemerin, but lower levels of FT3, FT4 and resistin(P < 0.01).In hyperthyroidism subgroup, level of adiponectin was negatively correlated with level of TSH(P < 0.01);level of resistin was negatively correlated with levels of TSH, FBG, FINS and HOMA-IR(P < 0.01), and positively correlated with levels of FT3 and FT4(P < 0.01);level of chemerin was negatively correlated with levels of FT3 and FT4(P < 0.01), and positively correlated with levels of TSH, FBG, FINS and HOMA-IR(P < 0.01).In hypothyroidism subgroup, while levels of adiponectin, resistin and chemerin had no significant correlation with level of FT3, FT4, TSH or TG(P>0.05), levels of adiponectin and chemerin was negatively correlated with levels of FBG, FINS and HOMA-IR(P < 0.01), and level of resistin was positively correlated with levels of FBG, FINS and HOMA-IR(P < 0.05).ConclusionsIncrease in the serum levels of adiponectin and chemerin, and decrease in the serum level of resistin, may be related to the pathogenesis of nodular goiter.
Clinical study on balance ability of patients with cerebral apoplexy by core muscle training combined with virtual reality technology

Abstract:
ObjectiveTo observe the effect of core muscle training combined with virtual reality (VR) technology on lower limb balance ability of patients with cerebral apoplexy.MethodsA total of 72 patients with cerebral apoplexy were randomly divided into core muscles training group and VR technology group, 36 cases in each group.The VR group adopted routine rehabilitation training combined with VR training, and the core muscle training group added core muscle training on the basis of the VR group.Berg balance scale (BBS), B-PHY balance function examination and training instrument, and activities of daily living (ADL) were used to assess the two groups before treatment, 4 weeks and 8 weeks after treatment.ResultsAfter 4 weeks of treatment, BBS score, COP elliptical area and ADL score of patients in both groups were improved compared with those before treatment (P < 0.05).After 8 weeks of treatment, BBS scores and ADL scores of patients in both groups were significantly improved compared with those before and after 4 weeks of treatment (P < 0.05 to P < 0.01).Compared with the same group before treatment and after 4 weeks of treatment, the area of COP ellipse measured when the core muscle group opened and closed eyes was significantly improved (P < 0.05).In the same period, the BBS score, COP elliptical area and ADL score of the core muscle group were significantly better than those of the VR technology group (P < 0.05 to P < 0.01).ConclusionsCore muscle training combined with VR technology can significantly improve the balance ability of patients with cerebral apoplexy, improve the quality of life, and is simple and easy to operate.
Analysis of the relationship between N-acetyltransferase 10 gene and prognosis in patients with acute myeloid leukemia

Abstract:
ObjectiveTo explore the relationship between N-acetyltransferase 10(NAT10) and prognosis of patients with acute myeloid leukemia(AML).MethodsA total of 84 patients with newly treated AML(initial treatment group), 20 patients with relapsed AML(relapsed group), 20 patients with complete remission of AML(remission group) and 20 healthy people(control group) from September 2015 to January 2018 were investigated.The expression level of NAT10 mRNA in bone marrow mononuclear cells was detected by RT-PCR.The receiver operator characteristic(ROC) curve was used to analyze the value of NAT10 mRNA in the early diagnosis of AML, and the area under the curve(AUC), sensitivity and specificity were calculated.Eighty-four AML patients were divided into the high expression group and low expression group according to the expression level of NAT10 mRNA, and the survival prognosis between two groups was compared.ResultsThe relative expression levels of NAT10 mRNA in bone marrow mononuclear cells in initial treatment group and recurrence group were higher than that in remission group and control group(P < 0.05 to P < 0.01).The AUC of NAT10 mRNA in the early diagnosis of AML was 0.827(95%CI: 0.723-0.930, P < 0.01), and the sensitivity and specificity were 83.6% and 82.6%, respectively.The results of COX multivariate analysis showed that the FLT3-ITD/TKD mutation and high expression of NAT10 mRNA were the independent influencing factors of the overall survival time and event-free survival time in AML patients(P < 0.05).The median overall survival time in the high NAT10 mRNA expression group was 4 months, which was lower than that of 10 months in low NAT10 mRNA expression group(P < 0.01).The median progression-free survival time in the group with high NAT10 mRNA expression group was 3 months, which was lower than that of 8 months in low NAT10 mRNA expression group(P < 0.01).ConclusionsNAT10 may have some value in the early diagnosis and evaluation of AML.The high expression of NAT10 mRNA is related to the poor survival and prognosis of AML patients.
Evaluation of the efficacy and safety of apatinib mesylate combined with XELOX regimen in the first-line treatment of advanced gastric cancer

Abstract:
ObjectiveTo compare the efficacy and safety of XELOX regimen with or without apatinib mesylate in the first-line treatment of advanced gastric cancer.MethodsThe clinical data of 65 patients with unresectable advanced gastric cancer were retrospectively analyzed.Thirty-five patients who received first-line chemotherapy with apatinib mesylate combined with XELOX regimen were included in the apatinib combined chemotherapy group, and 30 patients who received first-line chemotherapy with XELOX regimen were included in the chemotherapy group.Before the start of treatment and after the end of two cycles of treatment, the curative effect was evaluated, and the survival time and adverse reactions of patients were counted.ResultsThe short-term objective response rate (60.00% vs 23.33%) and disease control rate (88.57% vs 50.00%) of the apatinib combined chemotherapy group were better than those of the chemotherapy group (P < 0.01).By the time of follow-up, the median progression free survival was 8.9 months (95%CI: 0.679-2.382) and the median overall survival was 15 months (95%CI: 0.689-2.699) in the apatinib combined chemotherapy group.The median progression free survival was 7 months (95%CI: 0.420-1.474) and the median overall survival was 11 months (95%CI: 0.371-1.451) in the chemotherapy group.The survival time of apatinib combined with chemotherapy group was better than that of chemotherapy group (P < 0.05).The adverse reactions of the two groups were similar.There was no significant difference in the incidence of any level of adverse reactions between the apatinib combined chemotherapy group and the chemotherapy group(P>0.05).ConclusionsApatinib mesylate combined with XELOX is a safe and effective option for the first-line treatment of advanced gastric cancer.
Value analysis of the serum levels of AT-Ⅲ and G6PD in the early diagnosis and prognosis of neonatal sepsis

Abstract:
ObjectiveTo detect the serum levels of antithrombin-Ⅲ(AT-Ⅲ) and glucose-6-phosphate dehydrogenase(G6PD) in neonatal sepsis, and analyze its value in the early diagnosis and prognosis of neonatal sepsis.MethodsSeventy-seven children with neonatal sepsis, 50 children with bacterial infection without sepsis and 100 healthy newborns from January 2018 to January 2020 were divided into the sepsis group, infection group and control group, respectively.The serum level of AT-Ⅲ was detected using the developing substrate method, the serum level of G6PD was detected using the simple colorimetric method, the serum procalcitonin(PCT) level and white blood cell count(WBC) were determined suing the automatic biochemical analyzer, and the neonatal critical disease score(NCIS score) was recorded.The differences of the serum levels of AT-Ⅲ and G6PD among the sepsis group, infection group and control group, and between the good prognosis group and poor prognosis group were compared.The diagnostic value of serum levels AT-Ⅲ and G6PD in the neonatal sepsis and death were analyzed, and the risk factors of affecting neonatal sepsis were determined.ResultsCompared with the control group, the serum levels of AT-Ⅲ and G6PD in the infection group and sepsis group significantly decreased, and the serum levels of AT-Ⅲ and G6PD in sepsis group were lower than those in infection group(P < 0.01).The area under the curve(AUC) of serum levels of AT-Ⅲ and G6PD in the early diagnosis of neonatal sepsis were 0.811(95%CI: 0.748-0.874) and 0.847(95%CI: 0.788-0.905), respectively.Compared with the survival group, the serum levels of PCT and WBC significantly increased, the NCIS score and serum levels of AT-Ⅲ and G6PD were significantly decreased, the differences of which were statistically significant(P < 0.05 to P < 0.01).The AUC of serum AT-Ⅲ and G6PD were 0.791(95%CI: 0.671-0.911) and 0.800(95%CI: 0.678-0.922), respectively.The results of logistic regression analysis showed that the levels of AT-Ⅲ and G6PD decreasing were the risk factors for neonatal septicemia death.ConclusionsThe serum levels of AT-Ⅲ and G6PD increasing are closely related to the pathogenesis and poor prognosis of neonatal sepsis, and which have certain value in the early diagnosis and poor prognosis of neonatal sepsis.
Predictive analysis of UACR combined with serum KIM-1 and NGAL on postoperative contrast-induced nephropathy in patients with coronary heart disease

Abstract:
ObjectiveTo investigate the evaluation value of urine micro-albumin/urinary creatinine ratio(UACR) combined with serum kidney injury molecule-1(KIM-1) and neutrophil gelatinase-associated lipocalin(NGAL) on postoperative contrast-induced nephropathy(CIN) in patients with coronary heart disease.MethodsOne hundred and sixty-two patients with coronary heart disease were selected.All patients underwent coronary angiography and percutaneous coronary intervention.According to the occurrence of CIN after operation, they were divided into CIN group(22 cases) and non-CIN group(140 cases).The baseline data, UACR, serum KIM-1 and NGAL levels at 24 h after operation were compared between the two groups.Multivariate logistic regression was used to analyze the risk factors of CIN.Receiver operating characteristic(ROC) curve was used to analyze the diagnostic value of UACR, KIM-1 and NGAL in predicting CIN.ResultsThe levels of NGAL, serum KIM-1, UACR and preoperative UCr of CIN patients 24 h after operation were significantly higher than those in non-CIN group, while preoperative eGFR was significantly lower than that in control group(P < 0.05 to P < 0.01).Multivariate logistic regression analysis showed that the increased levels of UACR, KIM-1, NGAL were the risk factor for CIN(P < 0.01).ROC curve analysis showed that the area under the curve for NGAL, KIM-1, UACR and the combined diagnosis to predict CIN were 0.826, 0.801, 0.790, 0.886, the area under the curve of the combined diagnosis to predict CIN was significantly higher than that in the single diagnosis(P < 0.05).ConclusionsUACR, serum KIM-1 and NGAL are the independent risk factors for CIN in patients with coronary heart disease after surgery.High-risk patients with CIN are all at a high level in UACR, serum KIM-1 and NGAL.The combined prediction of the three indicators can improve the predictive value of CIN.
Expression and clinical significance of serum VEGF in patients with white matter ischemic lesions

Abstract:
ObjectiveTo explore the expression level of serum vascular endothelial growth factor(VEGF) in patients with white matter ischemic lesions(WMIL) and its relationship with WMIL.MethodsPatients with WMIL confirmed by craniocerebral magnetic resonance were selected as the research subjects, and the degree of white matter lesions was evaluated according to the Fazekas rating scale.Those with normal craniocerebral magnetic resonance matched with age and sex during the same period were selected as the control group.Serum VEGF concentrations were detected with enzyme-linked immunosorbent assay.The general clinical data and VEGF protein levels were compared between the two groups.Pearson correlation analysis was used to analyze the relationship between serum VEGF levels and the degree of white matter lesions, and multivariate logistic regression analysis was used to analyze the influencing factors of WMIL.ResultsA total of 115 patients in the WMIL group and 69 in the control group were included in this study.The proportion of hypertension and diabetes mellitus, and the level of VEGF in the WMIL group were significantly higher than those in the control group(P < 0.01).There was a significant positive correlation between serum VEGF level and Fazekas score in the WMIL group(r=0.871, P < 0.01).Multivariate logistic regression analysis showed that hypertension, diabetes mellitus and elevated VEGF levels were independent risk factors for WMIL(OR=5.350, 3.010, 19.382, P < 0.01).ConclusionsThe expression level of serum VEGF in WMIL patients is significantly increased.The increased level of serum VEGF may be an independent risk factor for WMIL and is positively correlated with the severity for WMIL.It may be used as one of the effective biological indicators of chronic white matter damage.
The relationship between the expression levels of Thr38, KCa3.1 and KLF4 in uterine smooth muscle and postpartum hemorrhage based on RhoA/Rho signaling pathway

Abstract:
ObjectiveTo investigate the relationship between the expression of p-cpi-17 (Thr38), KCa3.1 and Kr in uterine smooth muscle tissue of puerpera and postpartum hemorrhage based on RhoA/Rho signaling pathway.MethodsA total of 80 patients of postpartum hemorrhage due to uterine inertia were selected as study group, and 80 patients without postpartum hemorrhage in the same period were selected as control group.The spontaneous contractile activity of isolated uterine smooth muscle and the contractile activity of uterine smooth muscle with Rho kinase inhibitor were measured by isometric tension test.Real-time fluorescence quantitative reverse transcription polymerase chain reaction method was used to detect the expression levels of RhoA, ROCKⅠ, and ROCKⅡmRNA in uterine smooth muscle tissues.Western blotting was used to detect the expression levels of RhoA, ROCKⅠ, ROCKⅡ, Thr38, KCa3.1, and KLF4.Pearson correlation was used to analyze the relationship between RhoA protein and uterine smooth muscle contractility and Thr38, KCa3.1, KLF4.ResultsThe uterine smooth muscle contraction activity, contraction frequency and contraction amplitude of the study group were lower than those of the control group (P < 0.05 to P < 0.01).After Y-27632 was added, the uterine smooth muscle contraction activity was lower than before (P < 0.05).The expression levels of RhoA/Rho signaling pathway related mRNA and protein (RhoA, ROCKⅠ, ROCK Ⅱ) in the study group were lower than those in the control group (P < 0.01).The expression levels of Thr38, KCa3.1, and KLF4 in the study group were lower than those in the control group (P < 0.01).Pearson correlation analysis showed that RhoA/Rho signaling pathway related proteins and contractile activity were positively correlated with Thr38, KCa3.1, and KLF4 expression levels (P < 0.05).ConclusionsThe RhoA/Rho signaling pathway and expression of Thr38, KCa3.1, and KLF4 decrease in uterine smooth muscle tissue of parturients with postpartum hemorrhage due to uterine asthenia, and are positively correlated with the decrease of contractility, and jointly participate in the occurrence of postpartum hemorrhage.
Effect of different delivery modes on pelvic floor muscle potential and muscle strength of primiparas in early postpartum period

Abstract:
ObjectiveTo investigate the effects of different delivery modes on pelvic floor muscle potential and muscle strength of primiparas in early postpartum period.MethodsA prospective study has been conducted on primiparas who received prenatal and postnatal pelvic floor function examination in The First Hospital of Hebei Medical University since September 2018.According to the delivery mode, the primiparas were divided into the vaginal delivery group, emergency cesarean section group and elective cesarean section group, and the vaginal delivery group was subdivided into the perineal lateral resection group and non-perineal lateral resection group.The maximum electromyographic value of pelvic floor muscles, strength and fatigue degree of class Ⅰ and Ⅱ muscle fibers were quantitatively measured at 36-38 weeks of gestation and 6-8 weeks of postpartum.ResultsTo August 2019, a total of 221 parturient women were included, 139 cases in the vaginal delivery group(61 cases in the perineal resection group and 78 cases in the non-perineal resection group), 27 cases in the emergency cesarean section group and 55 cases in the elective cesarean section group.At 36-38 weeks of gestation, the differences of the maximum myopotential, muscle fiber strength and fatigue of pelvic floor muscle among three groups were statistically significant(P > 0.05).At postpartum 6-8 weeks, the maximum myopotential, strength and fatigue degree of class Ⅰ and Ⅱ muscle fibers in vaginal delivery group were lower than those in emergency cesarean section group and selective cesarean section group(P < 0.05 to P < 0.01).The differences of the maximum myopotential, strength and fatigue degree of class Ⅰ and Ⅱ muscle fibers between the perineal lateral resection group and non-perineal lateral resection group were not statistically significant(P > 0.05).ConclusionsThe effects of vaginal delivery on the pelvic floor muscle potential and muscle strength in early postpartum period is more obvious than that of elective or emergency cesarean section.Compared with the emergency cesarean section, the elective cesarean section has no benefit on pelvic floor muscle function.Lateral perineal resection does not reduce the effects of vaginal delivery on pelvic floor muscle function.
Correlation analysis of the triglyceride-glucose index and severity of coronary lesions in patients with coronary artery disease

Abstract:
ObjectiveTo investigate the correlation between the triglyceride-glucose(TyG) index and severity of coronary lesions in patients with coronary artery disease.MethodsFrom 2018 to 2019, 1 804 patients with coronary heart disease diagnosed by coronary angiography from the Department of Cardiology of The Affiliated Zhongda Hospital of Southeast University were investigated, and the clinical characteristics, TyG index, other biochemical indexes and echocardiography imaging were recorded.The Gensini score was calculated according to the results of coronary angiography.According to the TyG index, the patients were divided into the group 1(451 cases, TyG index ≤ 8.38), group 2(451 cases, 8.38 < TyG index ≤ 8.71), group 3(451 cases, 8.71 < TyG index ≤ 9.14) and group 4(451 cases, TyG index >9.14).ResultsThere was a positive linear correlation between the TyG index and Gensini score in patients with coronary heart disease(r=0.049, P < 0.05).The higher TyG index was, the more severe the coronary artery lesions were.In addition, the patients with high TyG index(group 4) were young, high incidence rates of hyperlipidemia, hypertension and diabetes mellitus, and high levels of blood glucose, total cholesterol, triglyceride and low density lipoprotein(P < 0.05 to P < 0.01).The area under ROC curve for TyG index to evaluate severe coronary artery disease(Gensini score ≥ 60 points) was 0.754(95%CI: 0.722-0.787, P < 0.05), and the best diagnostic value was TyG index ≥ 9.27.At this time, the sensitivity and specificity of diagnosis were 0.612 and 0.805, respectively.ConclusionsThe TyG index is positively correlated with the severity of coronary artery disease, and closely related to the risk factors of cardiovascular disease.It can be used as an effective tool to identify the severity of coronary artery disease.
Effects of application of labor analgesia at different stages of the first stage of labour on maternal and infant outcomes

Abstract:
ObjectiveTo explore the effects of the use of epidural block delivery analgesia at different stages of the first stage of labour on maternal and infant outcomes.MethodsA total of 144 full-term primipara were selected.The patients who did not perform labor analgesia were selected as the control group, and the patients who performed labor analgesia were selected as the observation group.According to the timing of the implementation of labor analgesia, they were divided into observation group 1 (uterine orifice dilation < 3 cm), observation group 2 (3 cm ≤ uterine orifice dilation ≤ 4 cm) and observation group 3 (uterine orifice dilation >4 cm).The timing of labour, the rate of transit caesarean section, the rate of perineal cleavage, the rate of midwifery delivery, and the Apgar score of newborns were compared among the groups.ResultsThe first stage of labor time in the observation group 1 was significantly longer than that in other groups(P < 0.01), and the difference in the time of second and third stages of labor was not statistically significant in each group (P > 0.05).The natural birth rate in the observation group was higher than that in the control group(P < 0.05).The rate of transit caesarean section in the observation group was lower than that in the control group(P < 0.01).There was no statistically significant difference in the comparison of the rate of lateral resection and the rate of midwifery delivery between the observation groups (P > 0.05).The natural birth rate of the observation groups 3 was significantly higher than that in the observation group 1 (P < 0.01), and the rate of transit caesarean section in the observation group 3 was significantly lower than that in the observation group 1 (P < 0.01).The 1-minute Apgar score and 5-minute Apgar score of neonates in the observation group 1, 2 and 3 and the control group were compared, and the results were not statistically significant (P > 0.05).ConclusionsThe application of labour analgesia can significantly reduce the rate of caesarean section during delivery and reduce the adverse effects of caesarean section on mother and infant.It can be promoted and applied in clinical work.Therefore, labor analgesia can be preferentially selected when the uterine orifice is dilated to 3-4 cm.
Efficacy analysis of TACE combined with MWA in the treatment of early and middle hepatocellular carcinoma

Abstract:
ObjectiveTo explore the clinical value of the sequential treatment of transcatheter arterial chemoembolization(TACE) combined with percutaneous microwave ablation(MWA) in early and middle hepatocellular carcinoma.MethodsThe clinical data of 108 patients treated with TACE alone or TACE combined with MWA were retrospectively analyzed.The propensity matching was performed at 1:1 in two groups, and 26 patients were included in each group.The short and long-term clinical efficacy in two groups were evaluated, and the subgroup analysis was also conducted based on tumor size and lesions.ResultsThe median survival time in TACE group and TACE-MWA group were 13 and 28 months, respectively.The survival rates of 1, 2 and 3 years in TACE-MWA group and TACE group were(84.6%, 59.0% and 35.2%, respectively) and(57.7%, 30.3% and 19.5%, respectively), and the survival rate in TACE-MWA group was better than that in TACE group(P < 0.01).The short-term efficacy in TACE-MWA group(61.5%) was higher than that in TACE group(30.8%)(P < 0.05).The disease control rate in TACE-MWA and TACE group was 80.8% and 46.2%, respectively, and the difference of which was statistically significant(P < 0.05).When the tumor size was < 5 cm, and the number of lesions was 2-4, the benefit in TACE+MWA group was greater than that in TACE group(P < 0.05).ConclusionsThe TACE with MWA sequential treatment can effectively improve the survival rate, and prolong the survival time in early and middle hepatocellular carcinoma patients.
Multifactorial analysis and treatment of intracranial arteriovenous malformation complicated with proximal aneurysm

Abstract:
ObjectiveTo investigate the risk factors of cerebral arteriovenous malformation(AVM) compolicated with proximal intracranial aneurysm(APIA) and its treatment.MethodsThe clinical data of 352 patients with AVM were retrospectively analyzed, and the patents were divided into the non-APIA group(321 cases) and APIA group(31 cases) according to the presence or absence of APIA.The gender, age, blood supply and size of AVM, drainage vein type, bleeding and bleeding site were compared between two groups, and the multivariate analysis was conducted.The patients with AVM complicated with APIA were divided into the endovascular treatment group, surgery treatment group and surgery combined with endovascular treatment group according to different treatment methods.The blood supplying artery, maximum diameter of aneurysm, maximum dimeter of AVM, post-treatment rebleeding rate and GOS prognosis score were compared among three groups.ResultsThe age of patients in APIA group was higher than that in non-APIA group(P < 0.01), and the rate of blood supply of posterior circulation in APIA group was 51.6%(16/31), which was significantly higher than that in non-APIA group(22.7%, 73/321)(P < 0.05).The results of multivariate analysis showed that the age and AVM blood supply were the independent risk factors of APIA.The differences of the aneurysm supplying artery, maximum diameter of aneurysm, maximum diameter of AVM, postoperative rebleeding rate and GOS prognosis score among endovascular treatment, surgery treatment and surgery combined with endovascular treatment groups were not statistically significant(P > 0.05).ConclusionsThe older the patient is, the more likely APIA will occur, and the AVM supplied by the posterior circulation is more likely to be complicated with APIA.Under the guidance of the principle of priority treatment of aneurysms, the surgery treatment, endovascular treatment and surgery combined with endovascular treatment can achieve a better prognosis.
Rehabilitation treatment effect of Yiqi Tongjing decoction combined with acupuncture on spinal cord injury

Abstract:
ObjectiveTo discuss the rehabilitation treatment effect of Yiqi Tongjing decoction combined with acupuncture on spinal cord injury.MethodsNinety-six patients with spinal cord injury were selected and equally divided into two groups by the digital table method.The control group was given conventional treatment, and the observation group was given Yiqi Tongjing decoction combined with acupuncture on the basis of control group.The American spinal cord injury association (ASIA) spinal function classification, muscle strength classification, and electromyography of the two groups were compared before and after treatment.ResultsAfter treatment, the ASIA classification and muscle strength classification of the two groups were significantly improved (P < 0.05 to P < 0.01), and the efficacy in the observation group was better than that in the control group after treatment (P < 0.05 to P < 0.01).After treatment, the amplitude of motor evoked potential (MEP) and skin sensory evoked potential (SEP) in the EMG test results of the two groups were significantly increased, and which in the observation group was significantly higher that in than the control group (P < 0.01).ConclusionsYiqi Tongjing decoction combined with acupuncture can effectively improve the spinal cord function of patients with spinal cord injury, improve muscle strength and motor and sensory functions, and is an effective method of rehabilitation for spinal cord injury.

Abstract:
ObjectiveTo investigate the effects of continuous stellate ganglion block(SGB) on the prevention of cerebral vasospasm during intracranial aneurysm intervention, and levels of plasma melatonin(MT) and endothelin-1(ET-1).MethodsEighty patients treated with intracranial aneurysm intervention were randomly divided into the observation group and control group according to the random number table method.The control group was treated with general anesthesia, and the observation group was additionally treated with continuous SGB on the basis of the control group.The anterior cerebral artery average blood flow velocity(ACA-Vm), anterior cerebral artery pulse index(ACA-PI), middle cerebral artery average blood velocity(MCA-Vm), middle cerebral artery pulsation index(MCA-PI), MT and ET-1 levels and postoperative 24 h complications were compared between two groups.ResultsThe ACA-Vm, ACA-PI, MCA-Vm, MCA-PI, PCA-Vm and PCA-PI in control group were significantly higher than those in observation group after treatment(P < 0.05 to P < 0.01).After treatment, the levels of MT and ET-1 in control group were significantly lower and higher than those in observation group(P < 0.05).After 24 h of operation, the incidence rate of complication in control group(22.50%) was significantly higher than that in observation group(7.50%)(P < 0.05).ConclusionsThe continuous SGB can effectively stabilize the cerebral blood flow after intracranial aneurysm intervention, promote the secretion and synthesis of MT, and inhibit the increasing of ET-1 level, so as to reduce the incidence rate of complications such as cerebral vasospasm.
Study on the correlation of LncRNA CASC2 and miR-155-5p with chemotherapy sensitivity and prognosis in patients with epithelial ovarian cancer

Abstract:
ObjectiveTo analyze the correlation between long chain non-coding RNA(LncRNA) cancer susceptibility candidate 2(CASC2), miR-155-5p and chemotherapy sensitivity and prognosis of patients with epithelial ovarian cancer.MethodsThe expression levels of LncRNA CASC2 and miR-155-5p in 87 patients with epithelial ovarian cancer(epithelial ovarian cancer group) and 72 patients with benign ovarian cancer(control group) were detected by real-time quantitative PCR(QRT-PCR).The expression levels of LncRNA CASC2 and miR-155-5p between two groups were compared.The correlations between LncRNA CASC2, miR-155-5p expression levels and clinicopathological features were analyzed in the epithelial ovarian cancer group.The epithelial ovarian cancer group was subdivided into the chemotherapy-resistant group(n=32) and chemotherapy-sensitive group(n=55) according to the occurrence of secondary drug resistance after chemotherapy, and the relationship between the expression levels of LncRNA CASC2 and miR-155-5p and chemotherapy sensitivity were analyzed.Meanwhile, the Pearson method was used to analyze the correlation between LncRNA CASC2 and miR-155-5p in epithelial ovarian cancer tissues, and the Kaplan-Meier method was used to analyze the relationship between the LncRNA CASC2, miR-155-5p and 5-year survival of patients with epithelial ovarian cancer.ResultsThe expression level of LncRNA CASC2 in epithelial ovarian cancer group was lower than that in control group, while the expression level of miR-155-5p in epithelial ovarian cancer group was higher than that in control group(P < 0.01).The expression levels of LncRNA CASC2 and miR-155-5p in epithelial ovarian cancer group were correlated with the abdominal metastasis, FIGO stage, differentiation degree and lymph node metastasis(P < 0.05 to P < 0.01), but not correlated with the age, tumor size, ascites and histological type(P>0.05).The expression level of miR-155-5p in chemotherapy-sensitive group was lower than that in chemotherapy-resistant group, while the expression level of LncRNA CASC2 in chemotherapy-sensitive group was higher than that in chemotherapy-resistant group(P < 0.01).The LncRNA CASC2 was negatively correlated with miR-155-5p in epithelial ovarian cancer group(r=-0.637, P < 0.05).The 5-year overall survival rate and median survival time in patients with high expression of miR-155-5p were lower than those in patients with low expression of miR-155-5p(P < 0.05), while the 5-year overall survival rate and median survival time in patients with high expression of LncRNA CASC2 were higher than those in patients with low expression of LncRNA CASC2(P < 0.05).ConclusionsThe expression levels of LncRNA CASC2 and miR-155-5p in the cancer tissues of epithelial ovarian cancer patients are closely correlated with the chemotherapy sensitivity and prognosis, and can be used as the effective predictors.
Effect of standard quadruple combined with Jinghua Weikang capsule on the levels of serum gastrin 17 and PG, and immune function in chronic atrophic gastritis patients with positive Hp

Abstract:
ObjectiveTo analyze the effects of standard quadruple combined with Jinghua Weikang capsule on the levels of serum gastrin 17(G-17) and pepsinogen(PG), and immune function in chronic atrophic gastritis patients with positive Hp.MethodsOne hundred and twenty chronic atrophic gastritis patients with positive Hp were divided into the observation group and control group according to the random number table method(60 cases in each group).The standard quadruple therapy in the control group were given, and the Jinghua Weikang capsule treatment in the observation group were additionally given on the basis of the control group.The clinical efficacy, eradication rate of Hp, serum levels of G-17, PGⅠ, PGⅡ, CD4+, CD8+, CD4+/CD8+ before and after treatment, and incidence rate of adverse reactions were compared between two groups.ResultsThe total clinical effective rate and clearance rate of Hp in the observation group were 93.33% and 96.67%, respectively, which were significantly higher than those in control group(75.00% and 81.67%)(P < 0.01).After treatment, the serum levels of G-17 and PGⅠ significantly increased, while the level of PG Ⅱ significantly decreased in two groups; the serum levels of G-17 and PGⅠ in observation group were significantly higher than those in control group, and the PG Ⅱ level in observation group was significantly lower than that in control group(P < 0.05 to P < 0.01).After treatment, the levels of CD4+ and CD4+/CD8+ significantly increased, and the CD8+ level significantly decreased in two groups; the levels of CD4+, CD8+ and CD4+/CD8+ in observation group were significantly better than those in control group(P < 0.05 to P < 0.01).There was no statistical significance in the incidence rate of adverse reactions between two groups(P>0.05).ConclusionsThe standard quadruple combined with Jinghua Weikang capsule can effectively improve the serum levels of G-17, PG and immune function in chronic atrophic gastritis patients with positive Hp, clearance rate of Hp and clinical total effective rate, which is worthy of clinical application.
Correlation between plasma D-dimer, fibrinogen levels and left atrial dimension in patients with coronary heart disease and atrial fibrillation

Abstract:
ObjectiveTo explore the correlation between plasma D-dimer (D-D), fibrinogen (Fg) levels and left atrial dimension in patients with coronary heart disease and atrial fibrillation.MethodsA total of 332 patients were enrolled and divided into coronary heart disease group(n=119), coronary heart disease atrial fibrillation group(n=108) and healthy group(n=105) according to the diseases suffered.Plasma D-D, Fg levels and left atrial dimension were measured and compared of all patients, Pearson method was used to analyze the correlation between the plasma D-D, Fg levels and left atrial dimension.ResultsThe plasma D-D, Fg levels and left atrial end systolic diameter (LAD) in coronary heart disease group and coronary heart disease atrial fibrillation group were higher than those in healthy group, and those in coronary heart disease atrial fibrillation group were higher than those in coronary heart disease group(P < 0.01).The plasma D-D, Fg levels and LAD in acute myocardial infarction were higher than those in coronary heart disease angina pectoris and ischemic cardiomyopathy, and those of angina pectoris of coronary heart disease were higher than ischemic cardiomyopathy(P < 0.01).The plasma levels of D-D, Fg and LAD in permanent atrial fibrillation were higher than those in paroxysmal and persistent atrial fibrillation, and the plasma levels of D-D, Fg and LAD in persistent atrial fibrillation were higher than those in paroxysmal atrial fibrillation (P < 0.01).The plasma D-D and Fg levels were positively correlated with LAD in patients with coronary heart disease and atrial fibrillation (r=0.603, 0.612, P < 0.05 and P < 0.01).ConclusionsThe plasma D-D and Fg levels in patients with coronary heart disease and atrial fibrillation are increased, which are positively correlated with left atrial dimension.
Analysis of the risk factors for recurrence after coning in patients with high-grade cervical intraepithelial neoplasia

Abstract:
ObjectiveTo investigate the high risk factors for recurrence of high-grade cervical intraepithelial neoplasia(CIN) patients after cervical conization.MethodsThe clinical data of 145 patients with high-grade CIN were retrospectively analyzed.According to the recurrence situation within 2 years after operation, the patients were divided into the recurrence group and non-recurrence group.The age, pregnancy and delivery times, menopause, gland involvement, CIN grading and preoperative and postoperative HPV infection bewteen tewo groups were compared.The univariate analysis and multivariate logistic regression were used to analyze the risk factors of recurrence after conical resection.ResultsThe 145 CIN patients were followed up for two years, 30 cases recurrenced, and 115 cases did not recurrence.The results of univariate analysis showed that the postmenopausal status, glandular involvement, CIN pathological grade and postoperatively high-risk human papillomavirus(HPV) infection were correlated with postoperative recurrence(P < 0.05 to P < 0.01), while the age and pregnancy number were not correlated with postoperative recurrence(P>0.05).The results of multivariate logistic regression analysis showed that the menopause, glandular involvement, CIN pathological grade and postoperative HPV infection were the independent risk factors of postoperative recurrence(P < 0.01).ConclusionsThe postmenopausal status, glandular involvement, pathological grade of CIN, and postoperative HPV infection are the high risk factors for recurrence after conization in high-grade CIN patients.
Correlation analysis between the target time in range of type 2 diabetes mellitus and glycosylated hemoglobin

ObjectiveTo analyze the correlation between the target time in range(TIR) and glycosylated hemoglobin A1c(HbA1c) in patients with type 2 diabetes mellitus(T2DM).MethodsOne hundred and forty-two patients with T2DM were divided into the group A(HbA1c < 7.0%), group B(7% ≤ HbA1c < 9%) and group C(HbA1c ≥ 9%) according to the level of HbA1c.The patients were monitored for 72 hours.The differences of the TIR, mean value of blood glucose monitoring(MBG), estimated glycosylated hemoglobin(eHbA1c), standard deviation(SD), coefficient of variation(CV), mean blood glucose fluctuation(MAGE) and fasting blood glucose(PG) were compared among three groups, and the effects of TIR on HbA1c was investigated.ResultsThe differences of the gender, age, course of disease, BMI, SBP, DBP and CV among group A, group B and group C were not statistically significant(P>0.05), but the differences of the HbA1c, eHbA1c, MBG, SD, MAGE, TIR and PG among three groups were statistically significant(P < 0.01).HbA1c was negatively correlated with the TIR(P < 0.01), and positively correlated with the PG, MBG, CV and eHbA1c(P < 0.01).The results of multiple linear regression analysis showed that the PG and TIR were the influencing factors of HbA1c.ConclusionsTIR is negatively correlated with HbA1c.PG and TIR are the influencing factors of HbA1c.

ObjectiveTo explore the application of spot tracking technique in the diagnosis of fetal aortic coarctation.MethodsNinety-six fetuses diagnosed as suspected aortic coarctation by the last echocardiography before delivery were selected.According to the fetus needed aortic constriction or not in the first week after delivery, they were divided into aortic constriction group and non-aortic constriction group(false positive).At the same time, 200 healthy fetuses were selected as the control group.After the conventional echocardiographic pa rameters were measured, the two-dimensional image was used for spot tracking technique analysis, and the relevant parameters were measured and calculated.The mean and standard deviation of the above-mentioned ultrasound parameters of healthy fetuses in the control group were used as the reference, the Z score of 96 fetuses with suspected aortic coarctation was calculated according to the formula.Finally, logistic regression analysis was used to identify the variables that separate the aortic coarctation group from the false positive group.Results46.88%(45/96) of 96 fetuses with suspected coarctation of the aorta were diagnosed as the postpartum aortic coarctation.Another 53.13%(51/96) did not have aortic coarctation after delivery and 1-2-year follow-up.The proportion of fetuses diagnosed with other cardiovascular abnormalities in the aortic coarctation group was significantly higher than that in the false positive group(P < 0.05).The logistic regression analysis identified 28 variable sets, R2 was 0.76.The model predicted aortic coarctation with the sensitivity of 95.56%, the specificity of 96.08%, and the area under the receiver operating characteristic curve of 0.982(95%CI=0.832-0.998).ConclusionsSpot tracking technique analysis of multiple echocardiographic measurements can help further diagnose the fetuses with suspected aortic coarctation in the prenatal examinations.
Development and influencing factors analysis of the genetic counseling cognition-behavior questionnaire for infertility patients

Abstract:
ObjectiveTo compile a genetic counseling cognition-behavior questionnaire for infertility patients, evaluate its reliability and validity, investigate the genetic counseling cognition-behavior level of infertility patients using the questionnaire, and analyze its influencing factors at the same time.MethodsThe study was conducted in two stages, the first stage was to determinate and pre-survey the scale content, and test the reliability and validity of the questionnaire; the second stage was to use the questionnaire to evaluate the genetic counseling cognition-behavior level of infertility patients, and analyze its related influencing factors.ResultsAfter expert revision and preliminary investigation, a final questionnaire with 20 items of genetic counseling cognition-behavior level was developed for infertility patients.The reliability test results of the questionnaire were as follows: the internal consistency of the Chronbach's α coefficient of the total scale was 0.898, and that of each factor was 0.889-0.900, the I-CVI ranged 0.879-1.000, the S-CVI/UA was 0.883, and the S-CVI/Ave was 0.972.The KMO value was 0.886, and the Bartlett sphericity test value was 1 124.960(df=190, P < 0.01).Five common factors with characteristic roots greater than 1 were extracted by orthogonal rotating principal component analysis, and the cumulative contribution rate was 62.128%, the load of 20 items were all ≥ 0.4, and the load range was 0.487-0.796.The subjects were divided into two groups according to gender.The difference of the score between two groups was statistically significant by comparing the general data of two groups(t=4.01, P < 0.01).The results of ordered multiple logistic regression analysis showed that the score of cognitive behavior level of genetic counseling in elderly group(35 years and above) was higher than that in younger group(below 35 years old)[OR=0.566, 95%CI=(0.421, 0.760), P < 0.01].Scores of male were higher than that of female[OR=1.735, 95%CI=(1.338, 2.250), P < 0.01].Compared with unemployed and others, the scores in cadre[OR=7.149, 95%CI=(4.158, 12.293), P < 0.01], full-time professional and technical personnel[OR=6.514, 95%CI=(4.411, 9.621), P < 0.01], business service personnel[OR=2.737, 95%CI=(1.931, 3.885), P < 0.01)] were higher, and the score of patients without adverse pregnancy history was lower[OR=0.336, 95%CI=(0.256, 0.440), P < 0.01].ConclusionsThe questionnaire has good reliability and validity, and can be used as a measurement tool to evaluate the genetic counseling cognition-behavior level and related factors analysis of infertility patients.The cognition-behavior level of genetic counseling in infertility patients is related to the age, sex, occupation, and adverse pregnancy history.
Effect of clozapine on glucose and lipid metabolism, cognitive function and related factors in the treatment of schizophrenic patients

Abstract:
ObjectiveTo investigate the effects of clozapine on glucose and lipid metabolism, cognitive function and related factors in the treatment of patients with schizophrenia.MethodsA total of 80 schizophrenic patients were randomly divided into the observation group and control group(40 cases in each group), and the control group and observation group were treated with risperidone tablets and clozapine tablets, respectively.The levels of estradiol(E2), prolactin(PRL), high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C), total cholesterol(TC), triglyceride(TG) and fasting blood glucose(FPG), and cognitive function scores were compared between two groups.ResultsAfter intervention, the general psychopathological symptoms, negative symptoms and positive symptoms in two groups were significantly lower than those before intervention(P < 0.01).After intervention, the scores of different dimensions of MCCB scale in two groups increased compared with those before intervention(P < 0.05 to P < 0.01), and the scores of working memory, speech learning and problem reasoning dimensions in observation group were significantly higher than those in control group(P < 0.05 to P < 0.01).Except for HDL-C in the control group, the differences of the levels of blood glucose and blood lipid in two groups between before and after treatment were statistically significant(P < 0.05 to P < 0.01).After intervention, the levels of FPG, TC, TG and LDL-C in observation group were higher than those in control group, while the HDL-C level in observation group was lower than that in control group(P < 0.01).After treatment, the E2 level decreased, and the PRL level increased in the control group(P < 0.01), while there was no statistical significance in the levels of E2 and PRL in the observation group between before and after treatment(P>0.05).After treatment, the PRL level in observation group was lower than that in control group, and the E2 level in observation group was higher than that in control group(P < 0.01).ConclusionsIn the treatment of schizophrenia, the application of clozapine intervention can improve the patients' cognition, and has little effect on PRL, but it will have a certain impact on glucose and lipid metabolism.

Abstract:
ObjectiveTo analyze the serum interleukin level in patients with adolescent depression disorders, and analyze its correlation with somatic symptoms.MethodsA total of 93 patients with adolescent depression disorders treated in the Hefei Fourth People's Hospital from December 2020 to December 2021 were selected and divided into functional somatic symptoms (FSS) group (n=40) and non-FSS group (n=53) according to whether they were accompanied by FSS.At the same time, thirty-two volunteers were selected as the control group.The Hamilton depression rating scale (HAMD) and childhood somatization inventory (CSI) were used to evaluate the severity of disease.The enzyme-linked immunosorbent assay was applied to detect the levels of serum IL-13 and serum IL-12/23p40.ResultsThe level of IL-13 in FSS group was significantly higher than that of the non-FSS group and control group (P < 0.05), and there was no statistically significant difference between the non-FSS group and control group (P>0.05).The level of IL-12/23p40 in the FSS group significantly higher than that in the non-FSS group (P < 0.05), while both had no significant difference with control group (P>0.05).The level of IL-13 and IL-12/23p40 were positively correlated with CSI total score and its sub-factor scores (IL-13:r=0.360, 0.286, 0.366, 0.310, 0.336, P < 0.01;IL-12/23p40:r=0.333, 0.360, 0.275, 0.280, 0.313, P < 0.01).ConclusionsThe level of serum IL-13 in adolescent depression patients with somatic symptoms is abnormal, and it has a positive correlation patients with somatic symptoms.
Analysis of HCV genotype distribution and natural drug resistance mutation in Huaibei city

Abstract:
ObjectiveTo analyze the distribution characteristics of HCV genotypes and natural resistance mutation of type 1b of patients with hepatitis C virus (HCV) infection in Huaibei city, so as to provide evidence for clinical antiviral treatment and infection prevention and control of HCV.MethodsThe HCV-RNA positive serum of patients with HCV infection[without any direct antiviral agent (DAAs) treatment] who were treated in Huaibei Miners General Hospital from 2019 to 2022 were selected.The samples were extracted and reverse transcribed, nested PCR was used to amplify the gene sequences of the NS5B, NS5A and NS3/4A regions.The electrophoresis-positive products were sent to company for sequencing.Sequence alignment analysis was performed using Mega11.0 software.ResultsThe HCV 1b subtype (68.63%) was the main subtype, followed by type 2a (29.41%), and 2 cases of type 3a (1.96%) was detected.The distribution of HCV subtypes was not significantly correlated with gender and age (P>0.05), type 1b was more common in the age group ≥ 55 years old.The main types of RASs were: C316N (100.00%) in NS5B region; Q54H (40.47%) and Y93H (19.05%) in NS5A region; Q86P (30.88%), I71V+Q86P (19.12%), V170I+Q86P (14.71%) and T72I/A+Q86P (13.24%) in NS3/4A region.The RASs with higher mutation frequency were: C316N (100.00%), Q54H (60.00%), Q86P (53.78%), Y93H (33.33%), I71V/S and (15.97%).ConclusionsHCV type 1b is the main epidemic genotype in Huaibei city, followed by 2a.Type 3a is detected for the first time.NS3/4A region is the main type of natural drug resistance mutation of type 1b, followed by NS5A, and NS5B was single.It is suggested that genotypes and RASs tests should be performed in HCV infected patients before DAAs.

Abstract:
ObjectiveTo explore the relationship between professional identity, knowledge sharing and research ability of nursing undergraduate.MethodsA questionnaire survey of 1 042 nursing undergraduates from a medical college were conducted with professional identity scale, knowledge sharing assessment scale and college students' research ability scale by convenient sampling method.ResultsThe total average scores of professional identity, knowledge sharing and research ability of nursing undergraduates were all above 3 points, and the overall level was above average.There was a significant positive correlation between the professional identity, knowledge sharing and research ability of nursing undergraduates (P < 0.01).Knowledge sharing can not only directly affect research ability, but also indirectly affect research ability through professional identity.ConclusionsProfessional identity plays a partial mediating role between knowledge sharing and research ability.Colleges should provide a good knowledge sharing environment for students, pay attention to students' sense of identity and belonging to their majors, which will help students to actively transform their knowledge and skills and improve their research ability.
Investigation of the nursing needs of maintenance hemodialysis patients with different demographic characteristics

Abstract:
ObjectiveTo explore the needs of maintenance hemodialysis patients based on Maslow's hierarchy of needs, and analyze its correlation with demographic characteristics.MethodsA total of 162 patients with maintenance hemodialysis were investigated by questionnaire, which included Maslow's needs at five levels.The needs of maintenance hemodialysis patients with different demographic characteristics were analyzed using multiple linear step-by-step regression.ResultsThe scores of physical needs, psychological needs, spiritual needs, self-esteem needs and care needs for medical personnel in patients with maintenance hemodialysis were(3.70±0.62) points, (3.21±0.52) points, (3.59±0.61) points, (3.85±0.56) points and(4.08±0.45) points, respectively.The higher the level of education was, the higher the score of patients' physical demand[B=0.096, 95%CI: 0.006-0.186)], demand for medical personnel[B=0.082, 95%CI(0.017-0.146)] and total demand[B=0.075, 95%CI(0.014-0.136)] were.The psychological needs score of patients with financial support from themselves was higher[B=-0.249, 95%CI(-0.407--0.092)], and the self-esteem needs score of patients with younger age was higher[B=-0.128, 95%CI(-0.242--0.015)], the differences of which were statistically significant(P < 0.05 to P < 0.01).ConclusionsThe need of patients for medical staff is highest, and the attention should be paid to the psychological needs of patients with different financial support, different age patients' self-esteem needs and effects of education on the body and needs of medical staff.The demographic analysis of patient needs will help patients and health care providers make joint decisions about the best personalized care.
Influencing factors analysis of rt-PA thrombolytic therapy and investigation of NLR predictive effect in ischemic stroke

Abstract:
ObjectiveTo study the prognostic factors of patients with ischemic stroke after emergency rt-PA thrombolytic therapy and explore the predictive value of neutrophil to lymphocyte ratio(NLR).MethodsA total of 187 patients with ischemic stroke who received emergency rt-PA thrombolytic therapy were selected, the influencing factors that might be related to the prognosis of ischemic stroke were collected.NIHSS score and MRS score at different time points were followed up and collected.The NIHSS score before thrombolysis was subtracted from the NIHSS score 2 weeks after thrombolysis.ΔNIHSS < 2 was considered as poor prognosis group (65 patients), and Δ NIHSS ≥ 2 was considered as good prognosis group (122 patients).The MRS score before thrombolysis was subtracted from the MRS score 3 months after thrombolysis.ΔMRS < 3 was considered as poor prognosis group (68 patients), and ΔMRS ≥ 3 was considered as good prognosis group (119 patients).Independent sample T test, Spearman correlation analysis, linear regression analysis and ROC curve analysis were used, respectively.The factors influencing the prognostic effect of thrombolytic therapy and the possible predictive indicators were analyzed.ResultsThe proportions of patients with hypertension and atrial fibrillation in the poor ΔNIHSS score group were higher than that in the better ΔNIHSS score group (P < 0.01 and P < 0.05).The levels of leukocyte, neutrophils and NLR in the poor ΔNIHSS score group were higher than those in the better ΔNIHSS score group (P < 0.01).The proportion of patients with atrial fibrillation in the poor ΔMRS score group was higher than that in the better ΔMRS score group (P < 0.05).The levels of leukocyte, neutrophils and NLR in the poor ΔMRS score group were higher than those in the better ΔMRS score group (P < 0.01).Further correlation analysis and linear regression analysis showed that NLR was significantly associated with ΔNIHSS score (b=-0.173, t=2.46, P < 0.05) and ΔMRS score (b=0.072, t=2.01, P < 0.05) after thrombolysis.ROC curve analysis showed that NLR could be used as the predictor of ΔNIHSS (AUC=0.595, P < 0.05, the optimal cutoff value was 5.169) and ΔMRS (AUC=0.618, P < 0.05, the optimal cutoff value was 6.228).ConclusionsNLR level before thrombolytic therapy is an independent factor which influencing the prognosis of patients with ischemic stroke after rt-PA thrombolytic therapy, and it can be used as a potential reference index to predict the efficacy of thrombolytic therapy.
