2010, 35(3): 217-220.
Abstract:
Objective: To investigate the influence of arsenic trioxide (As2O3) on migration and adhesion capability in melanoma B16 cells, and evaluate the effect of As2O3 on anti-invasion and anti-metastasis ability. Methods: The invasion and migration capacities of melanoma B16 cells were evaluated using Transwell Permeable Supports. The adhesion capacity of melanoma B16 cells was evaluated by MTT assay. The expression of adhesion molecule CD44 was detected by FCM.Results: As2O3 significantly inhibited the adhesion and migration capacities of melanoma B16 cells (P<0.05 to P<0.01) and down-regulated the expression level of CD44 (P<0.01).Conclusions: As2O3 has the effect of antagonizing invasion and metastasis in melanoma B16 cells, the mechanism maybe related to decrease the expression of adhesion molecule CD44.

2010, 35(3): 220-222.
Abstract:
Objective: To study the occurence of apoptosis and the related mechanism of the human umbilical vein endothelial cells (HUVEC) induced by Ureaplasma urealyticum (UU).Methods: The occurence of apoptosis of HUVEC was determined by flow cytometry at different times (2,4,8 and 12 h) after infected by UU.The productions of TNF-α,caspase-3 and caspase-8 in HUVEC after 8-hour infection of UU were detected by colorimetric assay, and the apoptosis rates were determined by flow cytometry when HUVEC was treated with TNF-α antibody,caspase-3 inhibitor or caspase-8 inhibitor prior to UU.Results: UU could induce the apoptosis occurrence of HUVEC with significant difference compared with control group. The productions of TNF-α,caspase-3 and caspase-8 were increased significantly in HUVEC infected by UU in contrast to control group, and the apoptosis occurence was significantly decreased when HUVEC was treated with TNF-α antibody,caspase-3 inhibitor or caspase-8 inhibitor. Conclusions: The results suggested UU could induce the apoptosis occurrence of HUVEC with time-dependence.The mechanism was likely related to extrinsic death pathway dependent on TNF death receptor which induced caspase-8 and caspase-3 activations.

2010, 35(3): 223-225.
Abstract:
Objective: To evaluate the efficiency of a new paraaortic counterpulsation device (PACD) assist therapy in acute heart failure animal modle. Methods: Eight sheep weighing (44.9±2.5)kg were selected.PACD was made by ourself. The blood chambers were connected to descending aorta through a valveless artifical vessel, and air chambers were connected to the intra-aortic ballon pump (IABP) console. The counterpulsation therapy was fulfilled by filling air during heart systole and deflating air during heart diastole.Acute heart failure was induced in all by snaring the left anterior descending coronary artery and (or) branch of left circumflex artery. The hemodynamic effects of PACD were assessed.Hemodynamic parameters during the PACD-assisted beats were compared with those during the unassisted beats to evulate the role of PACD.Results: PACD activation increased cardiac output and mean arterial pressure by 6.29% and 2.04%,respectively,while systole aortic pressure and diastolic aortic pressure were decreased significantly by 3.67% and 8.39%,respectively.The left carotid artery blood flow was increased by 8.12% (P<0.01),and left main coronary artery blood flow was increased by 13.88% (P<0.01).In assistant beats, the peak-augmented diastolic pressure was (97.25±13.67) mmHg,exceeding of unassisted significantly (P<0.01).Conclusions: In acute heart failure model,PACD is an effective counterpulsator which decreases left ventricular afterload and increases hemoperfusion of heart and brain.

2010, 35(3): 226-229.
Abstract:
Objective: To study the relationships between Helicobacter pylori L-form (Hp-L)infection and the invasion and metastasis of gastric carcinoma BGC-823 cells.Further to explore the probable mechanism between Hp-L infection and the progression of gastric carcinoma. Methods: Hp were incubated and induced into Hp-L.Then gastric cancer BGC-823 cells were co-incubated with them for 24 hours in different ratio and a control group was set.The ability of invasion and metastasis of BGC-823 cells were evaluated by using Transwell chambers. The expression of ezrin in BGC-823 cells was assessed by Western-blotting.Results: With the increasing of the concentrations of Hp-L forms, the cells which permeated the membrane were increased (P<0.05 to P<0.01),and the expression of ezrin in gastric carcinoma BGC-823 cells was risen (P<0.01).Conclusions: The infection of Hp-L form can increase the invasion and metastasis ability of gastric carcinoma cells. The mechanism may have relationship with the over-expression of ezrin.

2010, 35(3): 231-234.
Abstract:
Objective: To investigate the expression of caspase-3 protein and Ki-67 in bladder transitional cell carcinomas (BTCC) and its relationship with the grading and staging of the tumor. Methods: The expression of caspase-3 protein and Ki-67 in 55 cases of bladder transitional cell carcinomas and 10 cases of normal bladder mucosas were determined by immuohistochemisitry (SP method). Results: The positive expression of caspase-3 in BTCC (47.3%) was significantly lower than that in normal bladder mucosas (90.0%).The expression level of caspase-3 was positively correlated with the histological grades of BTCC (P<0.05).No positive expression of Ki-67 was observed in normal bladder mucosas, while it was 52.7% in BTCC (P<0.01).The expression of Ki-67 was related positively with the histological grade and negatively with the tumor stage of BTCC (P<0.05).Conclusions: Caspase-3 is closely correlated with the tumorigenesis of BTCC,and may act as a useful marker of invasion of BTCC.Increased expression of Ki-67 maybe related with the genesis and development of BTCC.

2010, 35(3): 234-235.
Abstract:
Objective: To investigate the association of maternal age and environmental factors with the risk of delivering a child with Down's syndrome (DS).Methods: The peripheral blood lymphocytes of the mother of the children with DS were cultured to prepare human G-banding chromosome examples for karyotype analysis, and the association of the maternal age and environmental factors with the risk of delivering a child with DS was analyzed. Results: One hundred and twenty-six of the 145 mothers of children with DS (86.90%) were below the age of 35.All the mothers had contacted with harmful physical or chemical factors to different degree before pregnancy. Conclusions: The age of the mother delivering a child with DS tends to be younger, which is probably due to the harmful environmental factors.Popularizing prepotency knowledge and enlarging the scope of prenatal screening and prenatal diagnosis may decrease the incidence of DS.
