2016, 41(3): 281-283. doi: 10.13898/j.cnki.issn.1000-2200.2016.03.001
Abstract:
Objective:To explore the effects of melatonin combined with cisplatin on the proliferation and apoptosis of Eca109 cell and study the molecular mechanism.Methods:Eca109 cells were con-cultured with different concentrations of melatonin with or without cisplatin for 24 h.The inhibitory rate of Eca109 cells were examined using MTT assay, and the cell apoptosis was analyzed using flow cytometry with propidium iodide staining.Western blot for apoptosis-related protein Bcl-2 and Bax were used to analyze possible mechanisms of the synergistic anti-proliferation effect of melatonin in combination with cisplatin. Results:Different concentrations of melatonin combined with cisplatin showed an obviously enhanced inhibitory effect on colony formation of Eca109 cells. When melatonin combined with cisplatin, the cell apoptosis rate increased to 29%, significantly higher than that in cells with cisplatin treatment alone(9.7%)(P<0.01).The expression of Bcl-2 was high but Bax was low in Eca109 cells, after stimulating with melatonin and cisplatin, the expression of Bcl-2 decreased but expression of Bax increased. Conclusions:Melatonin can enhance the anti-proliferative effect of cisplatin on Eca109 cell and can enhance cisplatin-induced apoptosis. The possible mechanism may related to the downregulation of Bcl-2 and upregulation of Bax protein expression.

2016, 41(3): 284-287. doi: 10.13898/j.cnki.issn.1000-2200.2016.03.002
Abstract:
Objective:To explore the effects of tetramethylpyrazine(TMP) on the levels of RhoA, Rho associated coiled coil forming protein kinase 2(ROCK2), myosin light chain(MLC) phosphorylation and ezrin-radxin-moesin(ERM) phosphorylation in the injury of human umbilical vein endothelial cells(HUVECs) induced by lipopolysaccharide(LPS).Methods:The HUVECs were cultured in vitro, and divided into the control group, LPS group and TMP groups(0.5, 1.0, 1.5 mg/mL).The mRNA levels of RhoA, ROCK2 and p-EZR in endothelial cells were detected by real-time fluorescence quantitative PCR, and the protein levels of RhoA, ROCK2, p-MLC and p-ERM were examined using western blotting. Results:Compared with the control group, the protein levels of RhoA, ROCK2, p-MLC and p-ERM, and the mRNA levels of RhoA, ROCK2 and p-Ezr in LPS group significantly increased(P<0.01).The differences of the protein and mRNA levels of RhoA between LPS group and 3 TMP groups were not statistically significant(P>0.05).Compared with the LPS group, the protein levels of ROCK2, p-MLC and p-ERM, and the mRNA levels of ROCK2 and p-Ezr decreased significantly in 3 TMP groups(P<0.01).The differences of the expression of ROCK2, p-MLC and p-ERM within 3 TMP groups were statistically significant(P<0.05 to P<0.01).Conclusions:TMP may protect the HUVECs against LPS-induced injury by inhibiting the expressions of ROCK2, p-MLC and p-ERM in Rho/ROCK signaling pathways, and reducing the injury of LPS on endothelial cell skeleton.

2016, 41(3): 357-360. doi: 10.13898/j.cnki.issn.1000-2200.2016.03.026
Abstract:
Objective:To investigate the cognition and application of antibiotics in vocational college students, analyze the risk factors of the abuse of antibiotics, and provide the scientific evidence for developing health education of antibiotics. Methods:The students from two vocational schools in Suzhou were investigated using the cluster sampling method. The investigation content included the profession, grade, cognition of antibiotics, use of antibiotics and purchasing prescription antibiotics. The differences of the cognition and use of antibiotics between two school students were compared by the cognitive scores and percentage of each indicator. Results:Four thousand four hundred and sixty-two effective questionnaires were collected, which included 2543 medical students and 1919 nonmedical students.Among 4462 students, 3621 students(81.2%) prepared for the antibiotic. One thousand and fifteen medical students(40%) and 666 nonmedical students(34.7%) with similar symptom took it by themselves. One thousand one hundred and forty-four medical students(45.0%) and 943 nonmedical students(49.1%) read the instruction carefully before using the antibiotic.Seven hundred and one medical students(27.6%) and 276 nonmedical students(14.4%) knew that the licensed pharmacist must be equipped in community pharmacy according to the state law.One thousand five hundred and sixty-eight medical students(61.7%) and 1063 nonmedical students(55.4%) thought that they were never asked to show the prescription when they bought the antibiotics, the difference between medical student and nonmedical student was statistically significant(P<0.01).Conclusions:The low antibiotic cognition and abuse of antibiotics in vocational students is common. The different cognition of antibiotic between medical student and nonmedical student results from their different knowledge.

Abstract:
Objective:To measure the liver shear wave velocity(SWV) in hepatolenticular degeneration(HLD) patients by virtual touch tissue quantification(VTQ) technology, evaluate the texture of the liver, and explore the correlations of liver SWV with copper biochemical indicators and serum liver fibrosis indexes. Methods:The SWV value in HLD patients was recorded by Siemens ACUSON-S2000 color doppler ultrasonic diagnostic instrument, which was equipped with the acoustic radiation force impulse imaging software and VTQ technology. The copper biochemical indicators including copper ion(Cu2+), ceruloplasmin(CP), and copper oxidase(SCO) and serum liver fibrosis indexes including procollagen type Ⅲ(PC-Ⅲ), collagen type Ⅳ(Ⅳ-C), laminin(LN) and hyaluronic acid(HA) were measured. The correlation of the liver SWV value with Cu2+, CP, SCO, PC-Ⅲ, Ⅳ-C, LN and HA were analyzed. Results:The liver SWV value was not correlation with Cu2+ and CP(P>0.05), negative correlation with SCO value(P<0.05).The SWV value was positive correlation with serum liver fibrosis indexes(PC-Ⅲ, Ⅳ-C, LN and HA)(P<0.05 to P<0.01).Conclusions:The ultrasound VTQ technology can reflect the Cu2+ transport activity and liver fibrosis degree to a certain extent, which has high clinical application value in the diagnosis of HLD.

Abstract:
Objective:To investigate the status of posttraumatic growth (PTG) and analyze its influencing factors among patients with myocardial infarction so as to provide basis for improving the PTG in myocardial infarction patients. Methods:A total of 172 patients with myocardial infarction were surveyed by filling out the social-demographic and disease-related information forms, the Life Events Scale and the Posttraumatic Growth Inventory by convenience sampling. Results:Different levels of PTG occurred in 78.5% of the patients with myocardial infarction, with a total score of 69.09±13.88.The total score of PTG was 69.09±13.88.Each dimension score of PTG from high to low in order was as follows:appreciation of life(21.78±4.34), new possibilities(13.65±3.21), self-transformation(13.52±3.34), relation with others(10.56±2.44) and personal ability(9.78±2.19).Single-factor analysis showed that the career of the patients, duration of the disease, monthly family income, religious faith, knowledge about the disease and whether or not combined with other diseases affected the PTG score significantly(P<0.01).Multiple-factor analysis showed that whether or not combined with other diseases, knowledge about the disease and monthly family income finally entered into the regression equation and accounted for 34.6 percent of the PTG.Conclusions:The essential predictors include whether or not the patients are combined with other diseases, the patients' knowledge about the disease and their monthly family income.
