Effect of over-expression of Periostin protein on the biological function of endothelial progenitor cells

Abstract:
Objective To construct the over-expression of Periostin protein lentiviral vector, and investigate its effects on the biological function of endothelial progenitor cells(EPCs).Methods The mouse EPCs were isolated from bone marrow, cultured and identified.The LV5/Periostin lentiviral vector was constructed, and transferred into the EPCs.The expression of green fluorescent protein(GFP) in transfected cells was observed under fluorescence microscope.The expression level of Periostin in EPCs was verified by Western blotting, the proliferation and apoptosis of infected cells were detected using the cell counting kit-8(CCK-8) assay and flow cytometry, and the migration ability and invasion ability of cells were detected using the scratch and Transwell assgy.Results The expression of GFP in EPCs was obvious.Compared with the control group, the proliferation activity of cells was lower, and the number of apoptosis cells were more in lentiviral vector group(P < 0.05).Compared with the control group, the migration rate of cells was higher, the number of invasion cells were more in lentiviral vector group(P < 0.05).Conclusions EPCs can be isolated from bone marrow, and cultured and identified.The over-expression lentiviral vector of Periostin protein can reduce the growth and proliferation, promote the apotosis, and enhance the migration and invasion ability of EPCs.
Cloning of human HNA-3a gene and its expression in HEK-293 cells

Abstract:
ObjectiveTo construct an eukaryotic expression vector containing full-length sequences of human HNA-3a gene, explore an experimental method for detecting human anti-HNA-3a antibody, and observe the protein expression of HNA-3a.Methods The primers were designed and synthesized according to human HNA-3a gene sequences in Genebank.The full-length of HNA-3a gene was reversely transcribed using RT-PCR, and cloned into pEGFP-N3 vector.The pEGFP-N3 expression vector containing human HNA-3a gene was transfected into the HEK-293 cells for transient expression using LipofectamineTM 2000 reagent, and the expression of the target gene was identified by Western blotting.Results The eukaryotic expression vector containing the full-length sequences of human HNA-3a gene was successfully constructed, and the target gene was highly expressed in HEK-293 cells.Conclusions The eukaryotic expression vector with high expression of HNA-3a protein is successfully constructed, which lays the foundation for the detection of the anti-HNA-3a antibody in human plasma.
Protective effect of IL-33 preconditioning on hepatic ischemia reperfusion injury in mice

Abstract:
Objective To explore the effects of interleukin(IL)-33 preconditioning on hepatic ischemia reperfusion in mice, and its possible mechanism.MethodsForty-five male C57BL/6 mice were randomly divided into the sham operation group(Sham group), ischemia and reperfusion injury group(PBS+I/R group, control group) and IL-33+ischemia and reperfusion injury group(rIL-33+I/R group)(15 mice each group).The hepatic artery and portal vein and bile duct of left, middle and caudate lobe of liver in control group were separated, and blocked using no injury microvascular clip for 60 min after 1 h of intravenous injection of the same amount of phosphate buffer.The hepatic artery and portal vein and bile duct of left, middle and caudate lobe of liver in experimental group(rIL-33+I/R group) were separated, and blocked using no injury microvascular clip for 60 min after 1 h of intravenous injection of recombinant IL-33 protein.The treatment of the sham operation group(Sham group) was the same as the control group except for no using no injury microvascular clip.Each group was divided into three subgroups(5 mice each subgroup), the serum levels of ALT, AST and IL-17, and expression levels of IL-33 and myeloperoxidase(MPO) in ischemic liver tissue were detected after 6 h, 12 h and 24 h of reperfusion.Results The expression of IL-33 in muse liver significantly increased during liver ischemia and reperfusion.After 12 h of reperfusion, the serum levels of ALT and AST in PBS+I/R group significantly increased compared with the Sham group(P < 0.01), and the serum levels of ALT and AST in rIL-33+I/R group significantly decreased compared with the PBS+I/R group(P < 0.01).The Results of HE staining in rIL-33+I/R group showed that the hepatocyte damaged, the neutrophil infiltration was found, and the MPO content in liver tissue was significantly lower than that in PBS+I/R group(P < 0.01).The Results of ELISA showed that the serum levels of IL-17A in rIL-33+I/R group was significantly lower than that in PBS+I/R group(P < 0.01).Conclusions IL-33 can protect the hepatic ischemia reperfusion injury in mouse, the mechanism of which may be related to the inhibition of IL-17A expression and reduction of neutrophil infiltration.
Effect of anisodamine pretreatment on the lung injury induced by hepatic ischemia-reperfusion in rats

Abstract:
Objective To investigate the effects of anisodamine pretreatment on the lung injury induced by hepatic ischemia-reperfusion in rats.Methods Forty-eight healthy male Sprague-Dawley rats were randomly divided into the sham operation group(group S), hepatic ischemia-reperfusion group(group IR) and anisodamine group(group A) using a random number table(16 rats each group).The hepatic portal in group S was dissected, and the ischemia-reperfusion model of 70% liver in group IR and group A were established.The group A was injected with 2 mg/kg anisodamine before 30 min of blocking liver blood flow and the group S and group IR were injected with the same volume of 0.9% sodium chloride solution.The rats were sacrificed after 3 h of reperfusion, the lung tissue was harvested, stained using HE, and the result of which was observed under light microscope.The ratio of wet to dry(W/D) of lung weight was calculated.The levels of malondialdehyde(MDA), superoxide dismutase(SOD) and myeloperoxidase(MPO) in lung tissue were detected using thiobaituricacid, xanthinoxidase and Hydrogen oxide reduction method, respectively.The expression levels of heme oxygenase-1(HO-1) and inducible nitric oxide synthase(iNOS) in lung tissue were determined by immunohistochemistry.Results The Results of HE staining in lung tissue showed that the lung injury in IR and A groups were more serious than that in group S, and the lung injury in group A was lighter than that in group IR.Compared with the group S, the ratio of W/D, MDA content, MPO activity and HO-1 and iNOS protein expression level significantly increased(P < 0.01), and the SOD activity significantly decreased in IR and A groups(P < 0.01).Compared with the group IR, the ratio of W/D, MDA content, MPO activity and iNOS protein expression level significantly decreased(P < 0.01), and the HO-1 protein level and SOD activity in lung tissue significantly increased in group A(P < 0.01).Conclusions Anisodamine pretreatment can alleviate the lung injury induced by hepatic ischemia-reperfusion in rats.
Study on the external fixator of titanium dioxide-coated screw in preventing pinhole infection

Abstract:
Objective To evaluate the antibacterial activity of titanium dioxide-coated screw, and observe its effects on pinhole infection in external fixator treating fractures.Methods Titanium dioxide-coated screw was prepared, and its antibacterial experiment was performed in vitro.Thirty New Zealand white rabbits were randomly divided into the pinhole group(MO group) and titanium oxide-coated screw group(TG group)(30 rabbits each group).A fracture complicated with pinhole infection model was established.Two groups were treated with taking X-ray, taking photo, harvesting materials for HE staining, culturing bacteria and counting.Results The bacteriostatic radius in TG group[(2.96±2.86)cm] was greater that in MO group[(33.96±6.10)cm] (P < 0.01).The inflammation scores of soft tissue in MO group after 1 d, 1 week and 4 weeks were higher that in TG group(P < 0.05 to P < 0.01).The difference of bacteria count after 1 d of operation between two groups was not statistically significant(P>0.05), which in TG group after 1 week and 4 weeks of operation decreased compared with the MO group(P < 0.01).A large number of inflammatory cells infiltrated, and gradually increased in MO group after 1 d, 1 week and 4 weeks of operation, and a few inflammatory cells infiltrated in TG group.Partial bone tissue with thread destroy absorption in MO group, and pinhole with clear and no damage and absorption in TG group were identified.At the first week, X-ray findings showed that fracture line began to blur in TG group, while fracture line in MO group remained clear.At the fourth week, the fractures in TG group had reached the bony union, and the displacement of the fracture end in MO group was still obvious and did not reach bony union.Conclusions Titanium dioxide-coated screw has a photocatalytic bactericidal effect, which can effectively prevent the pinhole infection in external fixator treating fracture.
Clinical pathological significance of vasculogenic mimicry in rectal adenocarcinoma

Abstract:
Objective To study the presence or absence of vasculogenic mimicry(VM) in rectal adenocarcinoma by some relevant data, and explore its clinical and pathological significance.MethodsThe clinicopathological data of 176 patients with rectal adenocarcinoma were collected.The VM was detected using immunohistochemical EliVisionTM plus method, and the cases were divided into the positive and negative groups.The relationship between VM and clinicopathological parameters was analyzed.Results Among 176 tissue samples, the positive expression of VM in 35 cases was identified, and the positive rate of which was 19.89%.The differences of the formation of VM among different gender, age and length of tumor were not statistically significant(P>0.05).The positive rates of VM in poor differentiation group, vascular and nerve invasion group and lymph node metastasis group significantly increased(P < 0.01).The Results of multivariate regression analysis showed that the survival time of patients became short with the increasing of the positive rate of VM(P < 0.01), the survival time of patients with lymph node metastasis was shorter than that in patients without lymph node metastasis(P < 0.01), and the survival time of patients with vascular and nerve invasion was shorter than that in patients without vascular and nerve invasion(P < 0.05).Conclusions VM is present in rectal adenocarcinoma, and the formation of VM is of great significance to the occurrence, development and prognosis of rectal adenocarcinoma.
Effect of different reconstruction ways of digestive tract on the reflux after laparoscopic gastric cancer surgery

Abstract:
ObjectiveTo analyze the related factors of the gastrointestinal reflux after laparoscopic gastric cancer surgery, and explore its treatment measures.MethodsThe clinical data of 1835 patients treated with laparoscopic gastric cancer surgery were retrospectively analyzed.Three hundred and eighty-six cases were treated with gastroduodenal anastomosis(B-Ⅰ type), 1002 cases were treated with gastric jejunum anastomosis(B-Ⅱ type), 412 cases were treated with stomach empty Roux-en-Y anastomosis, and 35 cases were treated with triangle anastomosis.ResultsPostoperative stageⅠreflux in 13 cases, stage Ⅱ reflux in 12 cases, stage Ⅲ and Ⅳ reflux in 5 cases were identified in the patients treated with B-Ⅰtype anastomosis.Postoperative stageⅠreflux in 39 cases, stage Ⅱ reflux in 67 cases, stage Ⅲ and Ⅳ reflux in 71 cases were identified in the patients treated with B-Ⅱtype anastomosis.Postoperative stageⅠreflux in 9 cases, stage Ⅱ reflux in 7 cases, stage Ⅲ and Ⅳ reflux in 3 cases were identified in the patients treated with stomach empty Roux-en-Y anastomosis.Postoperative stageⅠreflux in 1 case, and no stage Ⅱ, Ⅲ and Ⅳ reflux case were identified in the patients treated with triangle anastomosis.ConclusionsLaparoscopic triangle anastomosis in gastric cancer surgery has more obvious advantages of anti-reflux.
Effect of antepartum hemorrhage on maternal and infant outcomes in late pregnancy

Abstract:
ObjectiveTo investigate the influence of antepartum hemorrhage on maternal and infant outcomes in late pregnancy.MethodsOne hundred late pregnant women were divided into the control group(82 cases, no antepartum hemorrhage) and observation group(28 cases, antepartum hemorrhage).The clinical data of two groups were analyzed.The causes of antepartum hemorrhage in late pregnancy were analyzed, and the pregnancy outcomes of pregnant women and neonates were compared.ResultsThe late pregnancy bleeding causes were placental abruption, placenta previa, cervical and others in observation group, and the incidence rates were 50%, 32.14%, 14.29% and 3.57%, respectively.The incidence rate of postpartum hemorrhage between two groups was not statistically significant(P>0.05).The rate of vaginal delivery in observation group was lower than that in control group(P < 0.01).The incidence rates of cesarean section, hysterectomy, and premature birth and neonatal asphyxia in observation group were higher than those in control group, respectively(P < 0.01 and P < 0.05).The differences of the incidence rates of fetal distress and stillbirth between two groups were not statistically significant(P>0.05).ConclusionsThe incidence rates of the poor outcomes in late pregnancy women with antepartum hemorrhage increase significantly.The targeted clinical intervention should be implemented in medical personnel according to the specific situation.
Study on the screening method of macroprolactin in hyperprolactinemia

Abstract:
ObjectiveTo explore the effects of macroprolactin on the detection of prolactin(PRL), and the feasibility of the serum dilution polyethylene glycol(PEG) precipitation method in the screening of hyperprolactinemia(MP).MethodsThe PRL levels in blood samples of 84 patients with serum PRL level >30.0 ng/mL were detected using the 5-fold dilution 25% PEG and PEG precipitation method.The recovery rate of PRL less than 50% was used as the standard for MP screening.The value of 25% PEG and 5-fold dilution 25% PEG in the diagnosis of MP were evaluated using ROC curve.ResultsThe recovery rates of PRL in the eutrophic and hypertrophic groups were 116.0% and 63.0% using 5-fold dilution PEG precipitation method, and which was higher than those in traditional PEG precipitation method(42.0% and 101.0%), respectively(P < 0.05 and P < 0.01).The detection rate of dilution PEG precipitation method in genuine MP was 98.53%, which was higher than that of PEG precipitation method(88.23%)(P < 0.05).The area under the ROC curve of dilution PEG and PEG precipitation methods in diagnosing MP were 0.939(0.851 to 1.0) and 0.932(0.828 to 1.0), respectively.ConclusionsThe 5-fold dilution PEG precipitation method can increase the recovery rate of PRL and detection rate of genuine MP.
Analysis of the efficacy and influencing factors of 131Ⅰ in the treatment of lung metastasis of differentiated thyroid carcinoma

Abstract:
ObjectiveTo analyze the efficacy and influencing factor of 131Ⅰ in the treatment of lung metastasis of differentiated thyroid carcinoma(DTC).MethodsThe clinical data of 124 DTC patients with lung metastasis treated with 131Ⅰ were retrospectively analyzed.The age, gender, pathological type, local lymph node metastasis, distant metastasis of lung, serum Tg level before treatment with clearance of residual thyroid gland, operation method, postoperative treatment time and 131Ⅰ form of lung uptaking in all patients were analyzed, and the therapeutic efficacy and influencing factor of 131Ⅰ were evaluated.ResultsThe age, serum Tg level before treatment with clearance of residual thyroid gland and distant metastasis of lung were related to the curative effects(P < 0.05 to P < 0.01).The Results of multivariate analysis showed that the distant metastasis of lung was the important factor in influencing the therapeutic effect of 131Ⅰ(P < 0.05).ConclusionsThe effect of 131Ⅰ in the treatmet of the lung metastasis of DTC is good, and the distant metastasis of lung is the exact factor influencing the curative effect.
Efficacy analysis of percutaneous biliary stent combined with 125Ⅰ intracavitary implantation in the treatment of malignant obstructive jaundice

Abstract:
ObjectiveTo analyze the clinical efficacy of percutaneous biliary stent combined with 125Ⅰ intracavitary implantation in the treatment of malignant obstructive jaundice.MethodsA total of 137 malignant obstructive jaundice patients with inoperability were treated with percutaneous biliary stent implantation.According to the implantation of 125Ⅰ particle, the patients were divided into observation group(75 cases) and control group(62 cases).The tumor volume and liver function before and after treatment, stent patency time and survival time between two groups were analyzed and compared.ResultsCompared with the control group, the maximum and minimum diameter of tumor significantly decreased, and the liver function indexes were significantly improved in observation group at postoperative 3 months(P < 0.01).The stent patency rates in observation group at postoperative 9, 12 and 15 months were significantly better than those in control group(P < 0.01).The 1-year survival rate in observation group and control group was 72.0% and 33.9%, respectively, and the difference of which was statistically significant(P < 0.01).ConclusionsPercutaneous biliary stent combined with 125Ⅰ intracavitary implantation can inhibit the tumor growth, improve liver function and prolong the stent patency and survival time, and the treatment effect of which is superior to that of simple transcutaneous biliary stent implantation.
Analysis of the clinical characteristics of reactive hemophagocytic syndrome

Abstract:
ObjectiveTo analyze the clinical presentation, laboratory data, treatment and prognosis of reactive hyemophagocytic syndrome(RHS).MethodsThe clinical presentation, laboratory test, treatment protocols in 22 cases with RHS were retrospectively analyzed.The patients were divided into two groups according to the bone marrow hemocytology, and the difference between two groups was compared.ResultsAmong 22 patients, the incidence rates of hyperpyrexia, lymphadenectasis, blood system involovement rate, anemia and thrombocytopenia were 95.5%, 81.8%, 72.7% and 50.0%, respectively.The levels of WBC, HB, ALB, TG and fibrinogen(Fbg) in RHS patients were(19.5±8.1)×109/L, (95.2±14.4)g/L, (26.1±4.4)g/L, (3.2±2.1)mmol/L and(1.5 ±0.7)g/L, respectively.The bone marrow cytology Results showed the hemophagocytic phenomenon in 11 cases were indentified, and the difference of the positive indicators of examination between two groups were not statistically significant(P>0.05).The 50% patients alleviated after treatment, and discharged.Ten cases(one case lose to follow-up) died, which included adult Still disease in 5 cases, lymphoma in 1 case, dermatomyositis in 1 case, vasculitis related to anti-neutrophil cytoplasmic antibody in 1 case, systemic lupus erythematosus in 1 case and mesenteric lipid membrane inflammation in 1 case, and the respiratory failure was the death cause.ConclusionsThe adult still disease in RHS caused by rheumatic disease is often found, the lymphoma is more, poor prognosis and high death rate, and the main death cause of which is respiratory failure.The related indicator test can help to early diagnose the RHS.Hormones combined with cyclophosphamide can control the progression of disease.
Effect of neoadjuvant chemoradiotherapy on clinical effects and prognosis in postoperative patients with rectal cancer

Abstract:
ObjectiveTo investigate the effects of neoadjuvant chemoradiotherapy on the levels of microvessel density(MVD), proliferating cell nuclear antigen(PCNA) and vascular endothelial growth factor(VEGF) and prognosis in postoperative patients with rectal cancer.MethodsNinety-six rectal cancer patients treated with neoadjuvant chemoradiotherapy combined with resection and 86 rectal cancer patients treated with surgical resection were divided into the observation group and control group, respectively.The observation group was treated with rectal cancer radical surgery after routine segmental radiotherapy using 6 MV X-ray linear accelerator combined with neoadjuvant chemoradiotherapy for 4 to 6 weeks.The levels of MVD, PCNA and VEGF in surgical specimens in observation group before and after neoadjuvant chemoradiotherapy were detected.The expression levels of MVD, PCNA and VEGF in control group before and after operation were detected.The expression levels of MVD, PCNA and VEGF, and prognosis between two groups were compared before and after operation.The recurrence rates in two groups during 3-year following up were recorded.ResultsAfter operation, the expression levels of MVD and PCNA in observation group were significantly lower than those in control group(P < 0.01).There was not statistical significance in VEGF expression between two groups(P>0.05).The recurrence rates in control group and observation group during 3-year following up were 9.3% and 2.1%, respectively, the difference of which was not statistically significant(P>0.05).ConclusionsNeoadjuvant chemoradiotherapy can decrease the expression levels of MVD and PCNA in rectal cancer patients.
The postoperative application value of the vacuum sealing drainage technique in patients with giant malignant thyroid

Abstract:
ObjectiveTo explore the application effects of vacuum sealing drainage (VSD) technique on wound healing of cervical giant thyroid carcinoma after operation.MethodsEighty-nine cervical giant thyroid carcinoma patients treated with radical surgery were randomly divided into the control group(n=44) and observation group(n=45).The control group was treated with radical tissue flap, and the observation group was treated with radical tissue flap combined with VSD covering wound.After operation, the observation group was continously drainaged under the pressure of 6.65 to 7.98 mmHg for 72 h, then intermittently drainaged.After 1 to 2 weeks of operation, the VSD was stopped according to the postoperative wound recovery, and the wound was treated with skin graft according to the growth of granulation tissue.The wound repair between two groups was compared.ResultsThe VSD in 39 cases of observation group was stopped after 2 weeks of operation, the wound granulation tissue survival was good, completely covered the important organization and blood vessels of neck, and no pus secretion was found.The wound was timely grafted with skin, and the skin healing was good.The granulation tissue growth in 6 cases was poor in observation group after 2 weeks of operation, the wound was treated with debridement for 1 for 2 times and VSD, the skin graft surgery was successfully implemented after granulation tissue growth was good.Thirteen cases with wound infection in control group were found, the wound healed after repeated dressing wounds, and the obvious scars were identified.The skin flap necrosis in 7 cases were identified in control group, and the debridement and flap transplantation should be performed again.The difference of the infection rate of wound between two groups was statistically significant(P < 0.01).The average healing time of skin in observation group[(3.57±1.83) weeks] was significantly lower than that in control group[(6.75±2.42) weeks] (t=7.00, P < 0.01).ConclusionsThe application of VSD technique after resection of giant thyroid carcinoma can reduce the probability of infection, promote the wound healing, and improve the tolerance of patients.It is a simple, safe and effective method for repairing postoperative tissue defect of giant thyroid carcinoma.
The value of the retinal blood flow change in evaluating the prognosis of patients with wet age-related macular degeneration

ObjectiveTo investigate the value of the retinal blood flow change in evaluating the prognosis of patients with wet age-related macular degeneration.MethodsSeventy-two patients with wet age-related macular degeneration(72 eyes) and 72 healthy elderly(72 eyes) were divided into the observation group and control group, respectively.The vitreous chamber in observation group was injected with ranibizumab.The retinal blood flow levels in observation group before and after treatment, which in control group were observed using the ultrasound the physical examination.ResultsThe diastolic velocity and peak systolic velocity of ophthalmic artery and central retinal artery in observation group after treatment were significantly higher than before treatment(P < 0.05 to P < 0.01), and the difference of which between two groups was not statistically significant(P > 0.05).The differences between the resistance index in observation group before and after treatment, and resistance index in control group were not statistically significant(P > 0.05).The best corrected visual acuity in observation group after treatment was significantly better than before treatment(P < 0.01).The incidence rate of complications during treatment was 6.9%, and the complication was improved after symptomatic treatment.Linear correlation analysis showed that the diastolic velocity, peak systolic velocity of ophthalmic artery and central retinal artery were negatively correlated with the vision of prognosis(P < 0.05).ConclusionsThe retinal blood flow velocity in patients with wet age-related macular degeneration significantly decreases, the ranibizumab injection can effectively improve the vision and retinal blood flow of patients, and the change of retinal blood flow velocity has an important value on assessing the prognosis of patients in clinic.
Effect of low dose propofol maintenance anesthesia on the hemodynamics and recovery agitation after vocal cord polypectomy

Abstract:
ObjectiveTo investigate and analyze the effects of low dose propofol maintenance anesthesia on hemodynamics and recovery agitation in patients with vocal cord polypectomy.MethodsOne hundred patients scheduled by vocal cord polypectomy were divided into the observation group and control group according to random number table method(50 cases each group).The control group was treated with routine anesthesia, and the observation group was treated with 0.5 mg/kg propofol by intravenous injection.The gender, age, body weight, operation time and intraoperative bleeding in two groups were observed, heart rate(HR), mean arterial pressure(MAP) and oxygen saturation(SpO2) before and after operation, agitation score, recovery during recovery period (including the recovery time of spontaneous breathing, eye opening, extubation and incidence rate of agitation) in two groups were analyzed.ResultsHR and MAP of patients in two groups were significantly lower than those before operation(P < 0.01), HR and MAP were significantly higher at extubation, HR and MAP in observation group were significantly lower than those in control group(P < 0.01), HR and MAP in two groups were significantly lower 30 minutes after extubation(P < 0.01), SpO2 in two groups was significantly lower at extubation(P < 0.01), and recovered 30 minutes after extubation(P < 0.01).At different time after operation, the restlessness score of the observation group was lower than that of control group(P < 0.05 to P < 0.01).There was a significant difference in the restlessness score between the two groups at different time after operation(P < 0.01).The restlessness score at extubation was significantly higher than that at awakening and 30 minutes after extubation(P < 0.01).There was no significant difference in extubation time between the two groups(P > 0.05).The recovery time of spontaneous breathing and the opening time of eyes in the observation group were significantly longer than those in the control group(P < 0.01), and the incidence of restlessness in the observation group was significantly lower than that in the control group(P < 0.01).ConclusionsThe low dose propofol maintenance anesthesia can effectively reduce the fluctuation of hemodynamics and recovery agitation, and improve the breathing recovery time and opening eye time in patients with vocal cord polypectomy.
Influence of minimally invasive percutaneous tracheotomy in the treatment of laryngeal cancer on the clinical effect and stress response of patients

Abstract:
ObjectiveTo explore the influence of minimally invasive percutaneous tracheotomy in the treatment of laryngeal cancer on clinical effect and stress response of patients.MethodsOne hundred and twenty patients with laryngeal cancer were randomly divided into the control group(60 cases) and observation group(60 cases).The control group was treated with conventional tracheotomy, and the observation group was treated minimally invasive percutaneous tracheotomy.The level of blood rheology, operation, stress response index and postoperative complications between two groups were compared.ResultsAt T2, T3, T4 and T5, the SBP, DBP, HR and breathing rate in two groups significantly increased compared with at T1, and the differences of those among different time-points were statistically significant(P < 0.01).The SBP, DBP, HR and breathing rate in observation group at T2, T3, T4 and T5 were significantly lower than those in control group(P < 0.01).The operation time, incision length, intraoperative blood loss and wound healing time in observation group were significantly lower than those in control group(P < 0.01).The levels of blood glucose, cortisol, IL-6 and and CRP significantly increased in two groups at T'2, T'3 and T'4, and the differences of those among different time-point were statistically significant(P < 0.05 to P < 0.01).The levels of blood glucose, cortisol, IL-6 and and CRP in observation group were significantly lower than those in control group(P < 0.05 to P < 0.01).The incidence rate of postoperative complications and skin dent rate in observation group were significantly lower than that in control group(P < 0.01).ConclusionsThe minimally invasive percutaneous tracheotomy in treating laryngeal cancer has good effect and simple operation, which can significantly decrease intraoperative blood loss, operation time, stress response indicators and postoperative complications, and avoid postoperative skin dents.

ObjectiveTo investigate of the hypertension prevalence among adult residents in Tongling county, and its influencing factors.MethodsThe general condition, hypertension and its related factors, blood pressure, height, body weight and waist circumference in 3 221 adult residents from 2 091 households in 13 townships were investigated using a multistage cluster random sampling method.ResultsThe hypertension prevalence of adult residents in Tongling county was 33.9%, and the difference of which between male and female was not statistically significant(P > 0.05).The differences of the hypertension prevalence among adult residents with different occupations and whether participating in exercise were not statistically significant(P > 0.05).The differences of the hypertension prevalence among adult residents with different age, body mass index, education level, waist circumference, smoking, drinking, taste, daily intake of fresh vegetable, daily intake of fresh fruit and bacon frequency were statistically significant(P < 0.05 to P < 0.01).Logistic regression analysis showed that high education level, moderate taste, moderate daily intake of vegetable and occasional consumption of cured bacon were the protective factors of hypertension, and the age, body mass index and stopping smoking were the risk factors of hypertension.ConclusionsThe hypertension prevalence among adult residents is higher in Tongling county.Strengthening the knowledge of prevention and control of hypertension and forming a good behavior and dietary habit can comprehensively prevent and control the hypertension.

Abstract:
ObjectiveTo investigate the obesity status and its influencing factors in school age children, and provide theoretical basis for the intervention of obesity.MethodsThe basic information, children lifestyle and diet habbit in 1 000 children and their parents from Xianyang city were investigated with the self-made questionnaire on children's dietary behavior.ResultsAmong 1 000 subjects, 82 obesity children were found, and the difference of which between boy and girl was statistically significant(P < 0.05).The differences of the obesity rate in different ages between boy and girl were not statistically significant(P > 0.05).With the increasing of age, the obesity rate of children increased.Family history of obesity, family income, maternal taste preference were positively correlated with childhood obesity(P < 0.05 to P < 0.01), while maternal education and exercise time were negatively correlated with childhood obesity(P < 0.01 and P < 0.05).ConclusionsThe prevalence of obesity in school age children is serious, and the effective intervention should be carried out from dietary behavior and lifestyle.

ObjectiveTo sequencing on the various potential risk to the hospital, identify risk events that the hospital need to focus on, analyze the reason of its formation and impact on the hospital, strengthen risk management, and reduce the hospital disaster vulnerability.MethodsUsing the questionnaire, the grading standard of evaluation factor was established according to the possibility of occurrence and severity(injury-prevention).The disaster vulnerability was investigated using comprehensive index method, and the risk event was qualified according to the results.ResultsThe high risk harm of the hospital was the medical dispute(R=129.28), internet public opinion events(R=101.46), elevator accident(R=90.01), network information system disabled(R=89.47) and hospital infection(R=84.81) in turn.ConclusionsThe medical disputes, internet public opinion events, elevator accident, network information system disabled and hospital infection should be prevented.The hospital can strengthen the construction of mechanism and system, set up early warning system, formulate or improve the corresponding plan, organize training and drills, reserve the corresponding emergency supplies, and reduce the impact of disasters on hospital operation and health care work.

ObjectiveTaking the left-behind experience as an entry point, to explore the status quo of psychological health of normal college students under the perspective of positive psychology in order to promote the harmonious development of college students' psychological health.MethodsThe left-behind experience of college students from some normal colleges in Fuyang city and Anqing city of Anhui province investigated, and the level of students' mental health was comprehensively understood using the SCL-90 symptom self-assessment scale.ResultsStudents' mental health level is quite different according to the following factorswhether their parents are both going out for work, how long their parents going out for work, at what age they became unattended children, and how frequent they reunite with their parents.We have compared two groups of students. Students from group one, whose parents are both woking out for more than one year, all became unattented children before primary school age, and they reunite with their parents more than half a year each time.Students from group two, only one of whose parents are working out for no more than a year, all became unattented children after junior middle school age, and they reunite with their parents less than half a year each time.According to statistics, the mental health level of students from group one is lower than that of group two(P < 0.01).ConclusionsThe left-behind experience has certain influence on the mental health of normal college students.The targeted mental health education should be implemented for students.
Investigation on the somatic symptoms of patients with unipolar and bipolar depressive disorders

Abstract:
ObjectiveTo investigate the status and characteristics of somatic symptoms in patients with unipolar and bipolar depressive disorders.MethodsThe general information, somatic symptoms and depression in 94 patients with depressive disorders were investigated using the symptom questionnaire(SSI), Hamilton Depression Scale(HAMD) and general questionnaire, and all data were analyzed using SPSS20.0 software.ResultsThe average score of SSI of patients was(48.16±15.121), and the somatic symptoms of 26.6% patients were the moderate to severe levels.The somatic symptoms of patients with depressive disorders were significantly correlated with the diet, life events and side effects of medication(P < 0.05 to P < 0.01).The total score of somatic symptoms and non-pain somatic symptom score in patients with good diet were significantly lower than those in patients with normal diet and anorexic(P < 0.05 to P < 0.01).The score of pain symptoms in patients with experiencing more than 2 major life events in one year was significantly higher than that in patients without experiencing or with experiencing one major life event(P < 0.05).The total score of somatization symptoms and non-pain symptoms score in patients with three or more adverse drug reactions were significantly higher than those in patients without or with one adverse drug reaction(P < 0.01).The total score of somatic symptoms, dimension of pain and non-pain were positively correlated with the depression degree of patients(r=0.337, 0.325 and 0.316, P < 0.01).ConclusionsSomatic symptoms is more common in patients with depressive disorder.The diet, management of adverse drug reactions, and emotional management of patients experiencing major life events should be concerned by nurses, so that the comprehensive rehabilitation of patients with depressive disorders is promoted.

Abstract:
ObjectiveTo optimize the extraction technology of bishonokiol A from the Magnolia grandiflora seeds.MethodsThe concentration of bishonokiol A in different extracting solvents was determined using RP-HPLC method.The yield rate of bishonokiol A was set as the indictor, the effects of the ratio of material-to-liquid, soaking duration, and extraction times on the extraction technology were observed using L9(34) orthogonal test.ResultsThe orthogonal test showed that the ratio of material-to-liquid for 130, soaking duration for 48 h, and extraction times for three times were the best technological condition of bishonokiol A extraction.ConclusionsThe optimized extraction process designed by orthogonal test is simple, stable and reliable.

ObjectiveTo optimize the extraction process of Lonicerae flos and to establish an HPLC method to detect chlorogenic acid and isochlorogenic acid A rapidly.MethodsThe content of chlorogenic acid and isochlorogenic acid A was simultaneously determined by HPLC and used as an indicator to compare the effect of the four factors(alcohol concentration, reflux time, ultrasonic power and ultrasonic time) on the extraction process, optimize the extraction process of Lonicerae flos by ultrasonic-assisted alcohol method.ResultsTaking the comprehensive score of chlorogenic acid and isochlorogenic acid A as an indicator, the best extraction process was using 60% alcohol ultrasounding(power 500W) for 45 min, then reflux extraction for 1 hour.ConclusionsAn accurate and sensitive detection method is established, the optimized extraction process is economical and efficient, which can provide theoretical basis for production.
Comprehensive evaluation of brain and carotid ultrasound in perioperative period of carotid endarterectomy

Abstract:
ObjectiveTo analyze the comprehensive evaluation of brain and carotid ultrasound in perioperative period of carotid endarterectomy(CEA).MethodsThe transcranial Doppler(TCD) and carotid ultrasound in 60 patients treated with CEA were examined during the perioperative period.The results of preoperative, intraoperative and postoperative examination were recorded.Using preoperative digital subtraction angiography(DSA) as the golden standard, the accuracy of carotid artery stenosis detected by carotid ultrasound was analyzed.The detection rate of collateral circulation opening was compared between DSA and TCD, and the perioperative indicators were compared between carotid ultrasound and TCD.ResultsThe sensitivity, specificity, accuracy and Kappa value of carotid ultrasound in the detection of 50% to 69% stenosis were 66.67%(6/9), 100.00%(51/51), 95.00%(57/60) and 0.77, respectively.The detections rates of TCD for anterior communicating artery opening, posterior communicating artery opening and internal-external carotid artery opening were 91.07%, 85.96% and 87.50%, respectively.The preoperative carotid ultrasound showed that the diameter of the stenosis narrowed, and the blood flow velocity was higher than that of normal people.Carotid ultrasound found that the vascular stenosis on the operative side was effectively relieved after the stenosis was removed by operation.The intima of upper and lower margins was well fixed, there was no intimal floating in lumen, and the blood flowing was smooth.The PSV, EDV and PSVICA/PSVCCA at carotid artery opening and after 1 week of operation were significantly lower than those before surgery(P < 0.01), and the internal diameter of stenosis was significantly larger than that before surgery(P < 0.01).TCD examination showed that the PSV, EDV, MV and PI at carotid artery opening and after 1 week of operation were significantly higher than those after 5 to 10 minutes of anesthesia(P < 0.01), and all indictors returned to the normal level.The blood flow velocity of middle cerebral artery in 2 cases were more than 200% above the basal level.To avoid excessive perfusion, the proximal common carotid artery was clamped again, the blood pressure value decreased for 10 to 30 minutes, and the flow rate was restored in the end.ConclusionsTCD combined with carotid artery ultrasound can provide the accurate and effective objective basis for preoperative examination, intraoperative monitoring and postoperative evaluation of CEA, which has great application value in successful operation of patients.
Diagnostic value of the gastric ultrasound contrast in the etiology of gastroesophageal reflux anatomy

Abstract:
ObjectiveTo investigate the abnormal anatomy of gastroesophageal reflux using gastric ultrasound contrast, and provide the imaging evidence for the clinical diagnosis and treatment.MethodsForty-eight patients with gastroesophageal reflux and 51 healthy people were divided into the observation group and control group, respectively.The His angle and length of the esophagus in two groups were detected using contrast-enhanced ultrasonography of gastric filling and gastrointestinal meal barium, respectively, and which was compared between two groups.The relationship between the His angle and esophageal length of abdomen was investigated, and the diagnostic value of which in gastroesophageal reflux was evaluated.ResultsThe differences of the diagnositic results of His angle and abdominal esophageal length between two methods were not statistically significant(P>0.05).The results of ultrasonic examination showed that the esophagus length of abdominal section and His angle in observation group were lower and more than those in control group, respectively(P < 0.01).Taking 59.03° as the cut-off value, the sensitivity and specificity of His angle in the diagnosis of gastroesophageal reflux were 80.00% and 85.71%, respectively, and the area under ROC curve was 0.886.Taking 2.743 cm as the cut-off value, the sensitivity and specificity of abdominal esophageal length in the diagnosis of gastroesophageal reflux were 72.86% and 74.29%, respectively, and the ROC curve was 0.800.The abdominal esophageal length detected by ultrasound was negatively correlated with His angle(r=-0.681, P < 0.01).ConclusionsThe contrast-enhanced ultrasonography of gastric filling can accurately evaluate the His angle of patients with gastroesophageal reflux, and is safe and non-invasive operation, which has a certain diagnostic value for gastroesophageal reflux disease.
The detection rate and genotype analysis of Mycoplasma pneumonie in bronchoalveolar lavage fluid

Abstract:
ObjectiveTo detect the distribution of Mycoplasma pneumonie(MP) in bronchoalveolar lavage fluid of 136 children, and preliminarily understand the genotype of MP in Hefei Anhui province.MethodsThe MP infection cases in 136 bronchoalveolar lavage fluid samples were screened using PCR.The genotype of MP was analyzed using nest multiplex PCR, and the DNA temple of FH-MP standard strain was set as control.The recombinant plasmid was constructed using the amplified DNA fragment cloned by nest multiplex PCR, and sequenced.ResultsFifty-two positive MP samples were screened in 136 cases of bronchoalveolar lavage fluid using PCR.The nest multiplex PCR results showed that the genotype of MP positive samples was type P1-Ⅰ, and the genotype of standard strain FH-MP was type PCP1-Ⅱ.The sequencing results of the recombinant plasmid showed that the cloned fragment was right.ConclusionsThe genotype of epidemic strains of MP is type P1-Ⅰ, and the positive rate of MP infection is 38.2% in Hefei.
The detection of drug resistance genes of carbapenem-resistant Klebsiella pneumonia and its homology analysis

Abstract:
ObjectiveTo explore the drug resistance genes of carbapenem-resistant Klebsiella pneumonia(CRKP), and its homology analysis.MethodsForty strains of Klebsiella pneumonia were isolated from clinical samples from April to June 2018, and the drug sensitivity of which was identified using VITEK 2 COMPACT automatic bacterial sensitivity instrument.The carbapenem phenotypes of the 40 strains of Klebsiella pneumonia were screened using the modified hodge test(MHT) and modified carbapenem inactivation test(mCIM).PCR method was used to detect the main carbapenem resistance genes(including KPC, OXA-48 and NDM-1 genes).Matrix assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF MS) was used to analyze the homology of genes.ResultsThe 40 strains of CRKP were isolated from ICU ward(55%), mainly from respiratory tract samples(80%), and the resistance rates of those to cephalosporins, aminoglycosides and macrolides were above 80%.Thirty-three strains were positive in MHT test and mCIM test, 2 strains were negative in MHT test and mCIM test, and the remaining 5 strains were positive in MHT test and negative in mCIM test.The KPC gene was detected in 32 strains, and the OXA-48 and NDM-1 genes were not detected.The results of homology analysis showed that 40 strains of CRKP were divided into 7 types, which included type A in 34 strains(85%), and type B, C, D, E, F and G in 1 strain, respectively(each accounting for 2.5%).ConclusionsThe KPC-2 gene is the main genotype in CRKP, which can be divided into 7 kinds of types(type A as the main type).The study can provide the basis in effective treating and preventing the CRKP infection.

ObjectiveTo explore the effects of self-made pen-type vein puncture needle pressing tool in pressing and stopping blood in cancer patients after removing the intravenous infusion needle.MethodsNinety-five tumor patients with intravenous infusion were randomly divided into group A(34 cases), group B(31 cases) and group C(30 cases).Group A was treated using conventional compression method(stopping bleeding by pressing the needle eye with a cotton swab after pulling out the needle), group B was treated with infusion paste above the retaining point(stopping bleeding using self-made pen-type vein puncture needle and pressing belt according to device pressure after pulling out the needle), and group C was treated with hand pressing puncture needle site to stop bleeding after pulling out the needle.ResultsThere was statistical significance in the pain degree among the three groups(P < 0.01), and the differences of the pain degree between group A and B, and between group C and B were statistically significant(P < 0.05).There was statistical significance in pressing time among the three groups(P < 0.01), and the difference of the pressing time between group C and B was statistically significant(P < 0.05).The differences of the incidence rates of ecchymosis and subcutaneous hematoma among the three groups were statistically significant(P < 0.05), which in group B was significantly higher than that in group A(P < 0.05).ConclusionsThe self-made pen-type vein puncture needle pressing tool in the application of removing the intravenous infusion needle in cancer patients is comfortable, and has little pain, good stopping bleeding effect and short time.

ObjectiveTo investigate the effects of the risk assessment and precise nursing in patients with venous thromboembolism during anticoagulant therapy.MethodsOne hundred and eighty patients were randomly divided into the control group and observation group(90 cases each group).The control group were treated with routine care, and the observation group were treated with anticoagulant risk assessment combined with precise nursing.The score of anticoagulation knowledge, therapy compliance, nursing satisfaction, recent quality of life and incidence of hemorrhage between two groups were compared.ResultsThe score of anticoagulation knowledge, therapy compliance, recent quality of life and nursing satisfaction in observation group were significantly higher than those in control group(P < 0.05 to P < 0.01), and the difference of the incidence rate of hemorrhage between two groups was not statistically significant(P>0.05).ConclusionsDuring the anticoagulant therapy in patients with venous thromboembolism, the risk assessment and precise nursing can improve the satisfaction degree of nursing, medical compliance, recent quality of life and knowledge of anticoagulant therapy, and protect the safety of nursing of patients.

Abstract:
ObjectiveTo discuss the application effects of the lean management concept and method in the management of the patients with severe trauma in orthopedics.MethodsOne hundred and twenty patients with severe trauma in orthopedics were randomly divided into the lean group and routine group(60 cases each group).The routine group was nursed with the routine nursing management, and the lean group was nursed with the lean management concept and method based on the rountine group.The knowledge of disease, incidence of adverse nursing event, care satisfaction and anxiety between two groups were compared.ResultsThe disease knowledge and care satisfaction in lean group were significantly higher than those in routine group(P < 0.05).The nursing satisfaction rate, incidence rate of adverse nursing event and SAS scores in lean group after 7 and 14 d of intervention were significantly lower than those in routine group(P < 0.01 and P < 0.05).ConclusionsThe nursing based on the lean management concept and method can effectively improve the anxiety, disease knowledge and care satisfaction, and reduce the incidence rate of nursing events in orthopedic severe trauma patients.It is worthy of further clinical promotion.

Abstract:
ObjectiveTo investigate the status of knowledge, attitude and practice of breast cancer in female college students in Bengbu city, enhance the awareness of prevention of breast cancer, and reduce the threat of breast cancer to female students.MethodsSeven hundred and twenty-one female students from 4 colleges and universities in Bengbu city were randomly selected, and the knowledge, attitude and practice status of breast cancer in female college students were investigated using the self-made questionnaire.ResultsAmong the 22 problems about the knowledge of breast cancer, the awareness rate of 11 problems was less than 60%, and the knowledge of breast cancer prevention and breast cancer risk factors were also poor, the lowest ratio was 25.7%.In the attitude of prevention and treatment, 96.3% students thought that the early discovery was very important to save the life of breast cancer patients.In term of behavior, the self-examination behavior of breast was 15.8%.In term of access to breast cancer prevention resource, only 25.8% of the respondents received breast cancer knowledge health education.ConclusionsThe knowledge of breast cancer in female college students is not enough.Although the attitude towards prevention and treatment is generally positive, the level of behavior execution is relatively low, and the knowledge, attitude and practice is at low level.
